Ki67 in breast cancer: prognostic and predictive potential

Abstract: The leading parameters that define treatment recommendations in early breast cancer are oestrogen-receptor, progesterone-receptor, and human epidermal growth-factor status. Although some pathologists report Ki67 in addition to other biological markers, the existing guidelines of the American Society of Clinical Oncology do not include Ki67 in the list of required routine biological markers. The advent of new genetic tests has emphasised the role of proliferative genes, including Ki67, as prognostic and predictive markers. Additionally, randomised studies have retrospectively reviewed data and reported on the role of Ki67 in breast cancer. In light of new data, we have re-assessed evidence that could change guidelines to include Ki67 in the standard pathological assessment of early breast cancers. This review provides an update on the current knowledge on Ki67 and of the evidence in the published work about the prognostic and predictive role of this marker, and provides information on the laboratory techniques used to determine Ki67.

The authors' presentation of approximating functions and numerical solution of differential equations are thorough with coverage of splines and boundary value problems. Algorithms are developed in pseudocode (not FORTRAN or Pascal).

SARS-CoV-2 mRNA vaccine design enabled by prototype pathogen preparedness.

Abstract: A vaccine for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is needed to control the coronavirus disease 2019 (COVID-19) global pandemic. Structural studies have led to the development of mutations that stabilize Betacoronavirus spike proteins in the prefusion state, improving their expression and increasing immunogenicity1. This principle has been applied to design mRNA-1273, an mRNA vaccine that encodes a SARS-CoV-2 spike protein that is stabilized in the prefusion conformation. Here we show that mRNA-1273 induces potent neutralizing antibody responses to both wild-type (D614) and D614G mutant2 SARS-CoV-2 as well as CD8+ T cell responses, and protects against SARS-CoV-2 infection in the lungs and noses of mice without evidence of immunopathology. mRNA-1273 is currently in a phase III trial to evaluate its efficacy.

Self-compassion, Achievement Goals, and Coping with Academic Failure.

Abstract: Two studies examined the relationship between self-compassion, academic achievement goals, and coping with perceived academic failure among undergraduates. Selfcompassion entails being kind to oneself in instances of failure, perceiving one’s experiences as part of the larger human experience, and holding painful feelings in mindful awareness. Study 1 (N = 222) found that self-compassion was positively associated with mastery goals and negatively associated with performance goals, a relationship that was mediated by the lesser fear of failure and greater perceived competence of self-compassionate individuals. Study 2 confirmed these findings among students who perceived their recent midterm grade as a failure (N = 110), with results also indicating that self-compassion was positively associated with emotion-focused coping strategies and negatively associated with avoidance-oriented strategies.
