Abstract: It is unclear whether a single clone metastasizes and remains dominant over the course of lethal prostate cancer. We describe the clonal architectural heterogeneity at different stages of disease progression by sequencing serial plasma and tumor samples from 16 ERG-positive patients. By characterizing the clonality of commonly occurring deletions at 21q22, 8p21, and 10q23, we identified multiple independent clones in metastatic disease that are differentially represented in tissue and circulation. To exemplify the clinical utility of our studies, we then showed a temporal association between clinical progression and emergence of androgen receptor (AR) mutations activated by glucocorticoids in about 20% of patients progressing on abiraterone and prednisolone or dexamethasone. Resistant clones showed a complex dynamic with temporal and spatial heterogeneity, suggesting distinct mechanisms of resistance at different sites that emerged and regressed depending on treatment selection pressure. This introduces a management paradigm requiring sequential monitoring of advanced prostate cancer patients with plasma and tumor biopsies to ensure early discontinuation of agents when they become potential disease drivers.

Abstract: A search for the standard model Higgs boson (H) decaying to bb¯ when produced in association with a weak vector boson (V) is reported for the following channels: W(μν)H, W(eν)H, W(τν)H, Z(μμ)H, Z(ee)H, and Z(νν)H. The search is performed in data samples corresponding to integrated luminosities of up to 5.1 inverse femtobarns at s√=7 TeV and up to 18.9 fb−1 at s√=8 TeV, recorded by the CMS experiment at the LHC. An excess of events is observed above the expected background with a local significance of 2.1 standard deviations for a Higgs boson mass of 125 GeV, consistent with the expectation from the production of the standard model Higgs boson. The signal strength corresponding to this excess, relative to that of the standard model Higgs boson, is 1.0±0.5.

Abstract: A detailed description is provided of a new worm algorithm, enabling the accurate computation of thermodynamic properties of quantum many-body systems in continuous space, at finite temperature. The algorithm is formulated within the general path integral Monte Carlo (PIMC) scheme, but also allows one to perform quantum simulations in the grand canonical ensemble, as well as to compute off-diagonal imaginary-time correlation functions, such as the Matsubara Green function, simultaneously with diagonal observables. Another important innovation consists of the expansion of the attractive part of the pairwise potential energy into elementary (diagrammatic) contributions, which are then statistically sampled. This affords a complete microscopic account of the long-range part of the potential energy, while keeping the computational complexity of all updates independent of the size of the simulated system. The computational scheme allows for efficient calculations of the superfluid fraction and off-diagonal correlations in space-time, for system sizes which are orders of magnitude larger than those accessible to conventional PIMC. We present illustrative results for the superfluid transition in bulk liquid $^{4}\mathrm{He}$ in two and three dimensions, as well as the calculation of the chemical potential of hcp $^{4}\mathrm{He}$.
