Abstract: Adiponectin is an anti-diabetic adipokine. Its receptors possess a seven-transmembrane topology with the amino terminus located intracellularly, which is the opposite of G-protein-coupled receptors. Here we provide evidence that adiponectin induces extracellular Ca(2+) influx by adiponectin receptor 1 (AdipoR1), which was necessary for subsequent activation of Ca(2+)/calmodulin-dependent protein kinase kinase beta (CaMKKbeta), AMPK and SIRT1, increased expression and decreased acetylation of peroxisome proliferator-activated receptor gamma coactivator-1alpha (PGC-1alpha), and increased mitochondria in myocytes. Moreover, muscle-specific disruption of AdipoR1 suppressed the adiponectin-mediated increase in intracellular Ca(2+) concentration, and decreased the activation of CaMKK, AMPK and SIRT1 by adiponectin. Suppression of AdipoR1 also resulted in decreased PGC-1alpha expression and deacetylation, decreased mitochondrial content and enzymes, decreased oxidative type I myofibres, and decreased oxidative stress-detoxifying enzymes in skeletal muscle, which were associated with insulin resistance and decreased exercise endurance. Decreased levels of adiponectin and AdipoR1 in obesity may have causal roles in mitochondrial dysfunction and insulin resistance seen in diabetes.

Abstract: Long non-coding RNAs (lncRNAs) are largely heterogeneous and functionally uncharacterized. Here, using FANTOM5 cap analysis of gene expression (CAGE) data, we integrate multiple transcript collections to generate a comprehensive atlas of 27,919 human lncRNA genes with high-confidence 5' ends and expression profiles across 1,829 samples from the major human primary cell types and tissues. Genomic and epigenomic classification of these lncRNAs reveals that most intergenic lncRNAs originate from enhancers rather than from promoters. Incorporating genetic and expression data, we show that lncRNAs overlapping trait-associated single nucleotide polymorphisms are specifically expressed in cell types relevant to the traits, implicating these lncRNAs in multiple diseases. We further demonstrate that lncRNAs overlapping expression quantitative trait loci (eQTL)-associated single nucleotide polymorphisms of messenger RNAs are co-expressed with the corresponding messenger RNAs, suggesting their potential roles in transcriptional regulation. Combining these findings with conservation data, we identify 19,175 potentially functional lncRNAs in the human genome.

Abstract: The Fermi surface—the set of points in momentum space describing gapless electronic excitations—is a central concept in the theory of metals. In this context, the normal ‘metallic’ state of the optimally doped high-temperature superconductors is not very unusual: above the superconducting transition temperature, Tc, there is evidence for a large Fermi surface1,2,3, despite the absence of well-defined elementary excitations. In contrast, the normal state of underdoped high-temperature superconductors differs in that there is evidence for a ‘pseudogap’ above Tc (4–7). Here we examine, using angle-resolved photoemission spectroscopy, the temperature dependence of the Fermi surface in underdoped Bi2Sr2CaCu2O8+δ. We find that, on cooling the sample, the pseudogap opens up at different temperatures for different points in momentum space. This leads to an initial breakup of the Fermi surface, at a temperature T*, into disconnected arcs, which then shrink with decreasing temperature before collapsing to the point nodes of the superconducting ground state below Tc. This unusual behaviour, where the Fermi surface does not form a continuous contour in momentum space as in conventional metals, is unprecedented in that it occurs in the absence of long-range order. Moreover, although the superconducting gap below Tc evolves smoothly into the pseudogap above Tc, the pseudogap differs in its unusual temperature-dependent anisotropy, implying an intimate but non-trivial relationship between the pseudogap and the superconducting gap.
