Objectives: Human papillomavirus (HPV) in oral carcinoma (OSCC) and potentially malignant disorders (OPMD) is controversial. The primary aim was to calculate pooled risk estimates for the association of HPV with OSCC and OPMD when compared with healthy oral mucosa as controls. We also examined the effects of sampling techniques on HPV detection rates.
Methods: Systematic review was performed using PubMed (January 1966–September 2010) and EMBASE (January 1990–September 2010). Eligible studies included randomized controlled, cohort and cross-sectional studies. Pooled data were analysed by calculating odds ratios, using a random effects model. Risk of bias was based on characteristics of study group, appropriateness of the control group and prospective design.
Results: Of the 1121 publications identified, 39 cross-sectional studies met the inclusion criteria. Collectively, 1885 cases and 2248 controls of OSCC and 956 cases and 675 controls of OPMD were available for analysis. Significant association was found between pooled HPV-DNA detection and OSCC (OR = 3.98; 95% CI: 2.62–6.02) and even for HPV16 only (OR = 3.86; 95% CI: 2.16–6.86). HPV was also associated with OPMD (OR = 3.87; 95% CI: 2.87–5.21). In a subgroup analysis of OPMD, HPV was also associated with oral leukoplakia (OR = 4.03; 95% CI: 2.34–6.92), oral lichen planus (OR = 5.12; 95% CI: 2.40–10.93), and epithelial dysplasia (OR = 5.10; 95% CI: 2.03–12.80).
Conclusions: The results suggest a potentially important causal association between HPV and OSCC and OPMD.

Abstract: This review deals with grapevine responses to water stress by examining perturbations to physiological and molecular processes at the root, shoot, leaf and berry levels. Long-distance signalling among organs is also considered. Isohydric or anisohydric Vitis genotypes are described in relation to their response to drought, which is linked to stomatal behaviour. Stomatal regulation of grapevine under abscisic acid and hydraulic control (the latter being linked to embolism formation and recovery in water pathways upstream the stomata) is reviewed and linked to impairments of photosynthetic assimilation. We define three stages of photosynthesis regulation in grapevines that are subjected to progressive water stress on the basis of the main causes of assimilation decline. Early and late contributions of aquaporins, which play a fundamental role in water stress control, are discussed. Metabolic mechanisms of dehydration tolerance are rewieved, and variation linked to differences in transcript abundance of genes involved in osmoregulation, photosynthesis, photorespiration, detoxification of free radicals and coping with photoinhibition. Results of these defence strategies accumulated in berries are reviewed, together with perturbations of their molecular pathways. Features observed in different organs show that grapevine fits well as a complex model plant for molecular and physiological studies on plant drought avoidance/tolerance.

Abstract: How genomic heterogeneity associated with acquired resistance to targeted agents affects response to subsequent therapy is unknown. We studied EGFR blockade in colorectal cancer to assess whether tissue and liquid biopsies can be integrated with radiological imaging to monitor the impact of individual oncogenic alterations on lesion-specific responses. Biopsy of a patient's progressing liver metastasis following prolonged response to cetuximab revealed a K57T MEK1 mutation as a novel mechanism of acquired resistance. This lesion regressed upon treatment with panitumumab and the MEK inhibitor trametinib. In ctDNA, mutant MEK1 levels declined with treatment, but a previously unrecognized KRAS Q61H mutation was also identified that increased despite therapy. This same KRAS mutation was later found in a separate non-responding metastasis. In summary, parallel analyses of tumor biopsies and serial ctDNA monitoring show that lesion-specific radiographic responses to subsequent targeted therapies can be driven by distinct resistance mechanisms arising within separate tumor lesions in the same patient.

Abstract: In this paper we study the use of Sequence Diagrams and Statecharts of the Unified Modeling Language (UML) for the validation and the performance evaluation of systems. We assume that the system is specified as a set of Statecharts and that Sequence Diagrams are used to represent "executions of interest". UML lacks a formal semantics and hence it is not possible to apply, directly, mathematical techniques on UML models for system validation. To reach this goal we propose an automatic translation of Statecharts and Sequence Diagrams into Generalized Stochastic Petri Nets, and a composition of the resulting net models suitable for reaching a given analysis goal. The translation into Petri nets is based on the abstract syntax of the UML Collaborations and of the State Machines packages (from which Sequence Diagrams and Statecharts are derived) and the construction of the complete model heavily uses compositionality.
