Abstract: During fall 2008, an outbreak of hand, foot, and mouth disease (HFMD) with onychomadesis (nail shedding) as a common feature occurred in Finland. We identified an unusual enterovirus type, coxsackievirus A6 (CVA6), as the causative agent. CVA6 infections may be emerging as a new and major cause of epidemic HFMD.

Abstract: More than half of the patients with angiographically confirmed premature coronary heart disease (CHD) have a familial lipoprotein disorder1. Familial combined hyperlipidaemia (FCHL) represents the most common genetic dyslipidemia with a prevalence of 1.0–2.0% (refs 2,3). FCHL is estimated to cause 10–20% of premature CHD (ref. 1) and is characterized by elevated levels of cholesterol, triglycerides, or both3,4. Attempts to characterize genes predisposing to FCHL have been hampered by its equivocal phenotype definition, unknown mode of inheritance and genetic heterogeneity. In order to minimize genetic heterogeneity, we chose 31 extended FCHL families from the isolated Finnish population5 that fulfilled strictly defined criteria for the phenotype status. We performed linkage analyses with markers from ten chromosomal regions that contain lipid-metabolism candidate genes. One marker, D1S104, adjacent to the apolipoprotein A-ll (APOA2) gene on chromosome 1, revealed a lod score of Z=3.50 assuming a dominant mode of inheritance. Multipoint analysis combining information from D1S104 and the neighbouring marker D1S1677 resulted in a lod score of 5.93. Physical positioning of known genes in the area (APOA2 and three selectin genes) outside the linked region suggests a novel locus for FCHL on 1q21-q23. A second paper in this issue (Castellani et al.) reports the identification of a mouse combined hyperlipidaemia locus in the syntenic region of the mouse genome6, thus further implicating a gene in this region in the aetiology of FCHL.

Abstract: Experimental studies on animals have shown that dopamine is a key neurotransmitter in the regulation of working memory (WM) functions in the prefrontal cortex. In humans, blood flow studies show prefrontal involvement in WM functions, but direct evidence for the involvement of the dopaminergic system in WM is lacking. Using positron emission tomography with a recently developed high-affinity dopamine D2 receptor tracer, [11C]FLB 457, we explored frontal, temporal, and parietal D2 receptor availability in 12 healthy volunteers while they were performing verbal WM and sustained attention tasks. During the performance of both tasks, reduced D2 receptor availability was observed in the left ventral anterior cingulate, suggesting an attention or arousal-related increase in dopamine release during these tasks. Compared with the sustained attention task, the verbal WM task reduced D2 receptor availability in the ventrolateral frontal cortex bilaterally and in the left medial temporal structures (amygdala, hippocampus), suggesting that dopamine release in these regions might have a specific role in WM. In addition, correlation analyses indicated that increased dopamine release in the right ventrolateral frontal cortex and the left ventral anterior cingulate during the WM task was associated with faster and more stable WM performance, respectively. Our results indicate that regionally specific components of the frontotemporal dopaminergic network are functionally involved in WM performance in humans.

Abstract: We present new measurements of cosmic infrared background (CIB) anisotropies using Planck. Combining HFI data with IRAS, the angular auto- and cross-frequency power spectrum is measured from 143 to 3000 GHz, and the auto-bispectrum from 217 to 545 GHz. The total areas used to compute the CIB power spectrum and bispectrum are about 2240 and 4400 deg 2 , respectively. After careful removal of the contaminants (cosmic microwave background anisotropies, Galactic dust, and Sunyaev-Zeldovich emission), and a complete study of systematics, the CIB power spectrum is measured with unprecedented signal to noise ratio from angular multipoles � ∼ 150 to 2500. The bispectrum due to the clustering of dusty, star-forming galaxies is measured from � ∼ 130 to 1100, with a total signal to noise ratio of around 6, 19, and 29 at 217, 353, and 545 GHz, respectively. Two approaches are developed for modelling CIB power spectrum anisotropies. The first approach takes advantage of the unique measurements by Planck at large angular scales, and models only the linear part of the power spectrum, with a mean bias of dark matter haloes hosting dusty galaxies at a given redshift weighted by their contribution to the emissivities. The second approach is based on a model that associates star-forming galaxies with dark matter haloes and their subhaloes, using a parametrized relation between the dust-processed infrared luminosity and (sub-)halo mass. The two approaches simultaneously fit all auto- and cross-power spectra very well. We find that the star formation history is well constrained up to redshifts around 2, and agrees with recent estimates of the obscured star-formation density using Spitzer and Herschel. However, at higher redshift, the accuracy of the star formation history measurement is strongly degraded by the uncertainty in the spectral energy distribution of CIB galaxies. We also find that the mean halo mass which is most efficient at hosting star formation is log (Meff/M� ) = 12. 6a nd that CIB galaxies have warmer temperatures as redshift increases. The CIB bispectrum is steeper than that expected from the power spectrum, although well fitted by a power law; this gives some information about the contribution of massive haloes to the CIB bispectrum. Finally, we show that the same halo occupation distribution can fit all power spectra simultaneously. The precise measurements enabled by Planck pose new challenges for the modelling of CIB anisotropies, indicating the power of using CIB anisotropies to understand the process of galaxy formation.

Abstract: Arterial endothelial dysfunction is one of the key early events in atherogenesis, preceding structural atherosclerotic changes. It is also important in the late stages of obstructive atherosclerosis, predisposing to constriction and/or thrombosis. Endothelial function can be measured in coronary arteries and in the periphery by measuring vasomotor function after intra-arterial infusion of pharmacologic substances which enhance the release of endothelial nitric oxide. The disadvantage of these methods is their invasive nature, which generally makes them unsuitable for studies involving asymptomatic subjects. For this reason, noninvasive tests of endothelial function have been developed. In the most widely used of these, an ultrasound-based method, arterial diameter is measured in response to an increase in shear stress, which causes endothelium-dependent dilatation. Endothelial function assessed by this method correlates with invasive testing of coronary endothelial function, as well as with the severity and extent of coronary atherosclerosis. This noninvasive endothelial function testing has provided valuable insights into early atherogenesis, as well as into the potential reversibility of endothelial dysfunction by various strategies, including pharmacological agents (lipid lowering, ACE inhibition), l-arginine, antioxidants and hormones.
