Abstract: Recent studies have suggested that bone marrow cells possess a broad differentiation potential, being able to form new liver cells, cardiomyocytes and neurons1,2. Several groups have attributed this apparent plasticity to ‘transdifferentiation’3,4,5. Others, however, have suggested that cell fusion could explain these results6,7,8,9. Using a simple method based on Cre/lox recombination to detect cell fusion events, we demonstrate that bone-marrow-derived cells (BMDCs) fuse spontaneously with neural progenitors in vitro. Furthermore, bone marrow transplantation demonstrates that BMDCs fuse in vivo with hepatocytes in liver, Purkinje neurons in the brain and cardiac muscle in the heart, resulting in the formation of multinucleated cells. No evidence of transdifferentiation without fusion was observed in these tissues. These observations provide the first in vivo evidence for cell fusion of BMDCs with neurons and cardiomyocytes, raising the possibility that cell fusion may contribute to the development or maintenance of these key cell types.

Abstract: The IBIS telescope is the high angular resolution gamma-ray imager on-board the INTEGRAL Observatory, suc- cessfully launched from Baikonur (Kazakhstan) the 17th of October 2002. This medium size ESA project, planned for a 2 year mission with possible extension to 5, is devoted to the observation of the gamma-ray sky in the energy range from 3 keV to 10 MeV (Winkler 2001). The IBIS imaging system is based on two independent solid state detector arrays optimised for low (15 1000 keV) and high (0:175 10:0 MeV) energies surrounded by an active VETO System. This high eciency shield is essential to minimise the background induced by high energy particles in the highly excentric out of van Allen belt orbit. A Tungsten Coded Aperture Mask, 16 mm thick and1 squared meter in dimension is the imaging device. The IBIS telescope will serve the scientific community at large providing a unique combination of unprecedented high energy wide field imaging capability coupled with broad band spectroscopy and high resolution timing over the energy range from X to gamma rays. To date the IBIS telescope is working nominally in orbit since more than 9 month.

Abstract: [1] Many papers have developed algorithms to retrieve land surface temperature from at-sensor and land surface emissivity data. These algorithms have been specified for different thermal sensors on board satellites, i.e., the algorithm used for one thermal sensor (or a combination of thermal sensors) cannot be used for other thermal sensor. The main goal of this paper is to propose a generalized single-channel algorithm that only uses the total atmospheric water vapour content and the channel effective wavelength (assuming that emissivity is known), and can be applied to thermal sensors characterized with a FWHM (Full-Width Half-Maximum) of around 1 μm actually operative on board satellites. The main advantage of this algorithm compared with the other single-channel methods is that in-situ radiosoundings or effective mean atmospheric temperature values are not needed, whereas the main advantage of this algorithm compared with split-window and dual-angle methods is that it can be applied to different thermal sensors using the same equation and coefficients. The validation for different test sites shows root mean square deviations lower than 2 K for AVHRR channel 4 (λ ≈ 10.8 μm) and ATSR-2 channel 2 (λ ≈ 11 μm), and lower than 1.5 K for Landsat Thematic Mapper (TM) band 6 (λ ≈ 11.5 μm).

Abstract: Chronic restraint stress has been shown to induce structural remodelling throughout the interconnected dentate gyrus-CA3 fields. To find out how this stressor affects the rate of adult hippocampal neurogenesis, we subjected rats to acute or chronic restraint stress and assessed the proliferation, survival and differentiation of newly born cells in the dentate gyrus. We also examined polysialylated neural cell adhesion molecule expression, a molecule normally expressed in immature neurons and important for morphological plasticity. The results show that acute restraint stress did not change either the proliferation of dentate gyrus precursor cells or the expression of polysialylated neural cell adhesion molecule, whereas 3 weeks of chronic restraint stress suppressed proliferation by 24% and increased polysialylated neural cell adhesion molecule expression by 40%. The study was extended for an additional 3 weeks to trace the survival and development of the cells born after the initial 3 weeks of restraint. Rats subjected to 6 weeks of daily restraint stress exhibited suppressed cell proliferation and attenuated survival of the recently born cells after the extended time course, resulting in a 47% reduction of granule cell neurogenesis. Furthermore, 6 weeks of chronic stress significantly reduced the total number of granule cells by 13% and the granule cell layer volume by 5%. Expression of polysialylated neural cell adhesion molecule followed a biphasic time course, displaying a significant up-regulation after 3 weeks of daily restraint stress that was lost after 6 weeks of stress. These studies may help us understand the basis for hippocampal shrinkage and raise questions about the ultimate reversibility of the effects of chronic stress.

Abstract: It is widely assumed that the ability of Candida albicans to switch between different morphologies is required for pathogenesis. However, most virulence studies have used mutants that are permanently locked into either the yeast or filamentous forms which are avirulent but unsuitable for discerning the role of morphogenetic conversions at the various stages of the infectious process. We have constructed a strain in which this developmental transition can be externally modulated both in vitro and in vivo. This was achieved by placing one copy of the NRG1 gene (a negative regulator of filamentation) under the control of a tetracycline-regulatable promoter. This modified strain was then tested in an animal model of hematogenously disseminated candidiasis. Mice injected with this strain under conditions permitting hyphal development succumbed to the infection, whereas all of the animals injected under conditions that inhibited this transition survived. Importantly, fungal burdens were almost identical in both sets of animals, indicating that, whereas filament formation appears to be required for the mortality resulting from a deep-seated infection, yeast cells play an important role early in the infectious process by extravasating and disseminating to the target organs. Moreover, these infecting Candida yeast cells still retained their pathogenic potential, as demonstrated by allowing this developmental transition to occur at various time points postinfection. We demonstrate here the importance of morphogenetic conversions in C. albicans pathogenesis. This engineered strain should provide a useful tool in unraveling the individual contributions of the yeast and filamentous forms at various stages of the infectious process.

Abstract: SPI is a high spectral resolution gamma-ray telescope on board the ESA mission INTEGRAL (International Gamma Ray Astrophysics Laboratory). It consists of an array of 19 closely packed germanium detectors surrounded by an active anticoincidence shield of BGO. The imaging capabilities of the instrument are obtained with a tungsten coded aperture mask located 1.7 m from the Ge array. The fully coded field-of-view is 16degrees, the partially coded field of view amounts to 31degrees, and the angular resolution is 2.5degrees. The energy range extends from 20 keV to 8 MeV with a typical energy resolution of 2.5 keV at 1.3 MeV. Here we present the general concept of the instrument followed by a brief description of each of the main subsystems. INTEGRAL was successfully launched in October 2002 and SPI is functioning extremely well.

Abstract: The JEM-X monitor provides X-ray spectra and imaging with arcminute angular resolution in the 3 to 35 keV band. The good angular resolution and the low energy response of JEM-X plays an important role in the identification of gamma ray sources and in the analysis and scientific interpretation of the combined X-ray and gamma ray data. JEM-X is a coded aperture instrument consisting of two identical, coaligned telescopes. Each of the detectors has a sensitive area of 500 cm 2 , and views the sky through its own coded aperture mask. The two coded masks are inverted with respect to each other and provides an angular resolution of 3 0 across an eective field of view of about 10 diameter.

Abstract: We have observed a narrow state near 2.32 GeV/c(2) in the inclusive D(+)(s)pi(0) invariant mass distribution from e(+)e(-) annihilation data at energies near 10.6 GeV. The observed width is consistent with the experimental resolution. The small intrinsic width and the quantum numbers of the final state indicate that the decay violates isospin conservation. The state has natural spin-parity and the low mass suggests a J(P)=0(+) assignment. The data sample corresponds to an integrated luminosity of 91 fb(-1) recorded by the BABAR detector at the SLAC PEP-II asymmetric-energy e(+)e(-) storage ring.

It also offers an in‐depth study of resistance to change. Through empirical research, the paper analyzes the importance of the sources of resistance to change defined theoretically, and considers both types of changes. The article shows which sources of resistance differ most, according to the scope of change, offering hints about where organizations should pay special attention when initiating a change process.

Abstract: OBJECTIVE —To identify a reliable yet simple indirect method for detection of insulin resistance (IR). RESEARCH DESIGN AND METHODS —A total of 65 subjects (44 men and 21 women aged 30–60 years) were selected by a simple random sampling method. Inclusion criteria were voluntary participation from staff and hospital personnel, absence of abnormal glucose tolerance, and normal results of lipid profile and basic blood chemistry. A blood sample was taken after a 12-h overnight fast to determine plasma lipid, glucose, and insulin levels. An intravenous glucose tolerance test with administration of insulin after 20 min and extraction of multiple blood samples for glucose and insulin measurements and calculation of the minimal model approximation of the metabolism of glucose (MMAMG) S i value were performed. Three indirect indexes used to predict insulin sensitivity or IR were calculated, and metabolic syndrome was diagnosed using the Adult Treatment Panel III (ATP III) criteria. All results were correlated with those of the MMAMG. RESULTS —The 75th percentile value as the cutoff point to define IR corresponded with a fasting plasma glucose level of 12 mU/l, a homeostasis model assessment of 2.6, a 25th percentile for S i value of 21, and QUICKI (quantitative insulin sensitivity check index) and McAuley indexes of 0.33 and 5.8, respectively. The S i index correlated ( P CONCLUSIONS —When compared with the S i index, the most sensitive and specific indirect method was the score proposed by McAuley et al. (specificity 0.91, sensitivity 0.75, 9.2 probability ratio of a positive test), followed by the existence of metabolic syndrome (specificity 0.91, sensitivity 0.66, 7.8 probability ratio of a positive test).

Abstract: Bacterial symbioses are widespread among insects, probably being one of the key factors of their evolutionary success. We present the complete genome sequence of Blochmannia floridanus, the primary endosymbiont of carpenter ants. Although these ants feed on a complex diet, this symbiosis very likely has a nutritional basis: Blochmannia is able to supply nitrogen and sulfur compounds to the host while it takes advantage of the host metabolic machinery. Remarkably, these bacteria lack all known genes involved in replication initiation (dnaA, priA, and recA). The phylogenetic analysis of a set of conserved protein-coding genes shows that Bl. floridanus is phylogenetically related to Buchnera aphidicola and Wigglesworthia glossinidia, the other endosymbiotic bacteria whose complete genomes have been sequenced so far. Comparative analysis of the five known genomes from insect endosymbiotic bacteria reveals they share only 313 genes, a number that may be close to the minimum gene set necessary to sustain endosymbiotic life.

Abstract: The evolutionary approach called scatter search originated from strategies for creating composite decision rules and surrogate constraints. Recent studies demonstrate the practical advantages of this approach for solving a diverse array of optimisation problems from both classical and real--world settings. Scatter search contrasts with other evolutionary procedures, such as genetic algorithms, by providing unifying principles for joining solutions based on generalised path constructions in Euclidean space and by utilising strategic designs where other approaches resort to randomisation. Additional advantages are provided by intensification and diversification mechanisms that exploit adaptive memory, drawing on foundations that link scatter search to tabu search. The main goal of this chapter is to demonstrate the development of a scatter search procedure by demonstrating how it may be applied to a class of non-linear optimisation problems on bounded variables. We conclude the chapter by highlighting key ideas and research issues that offer the promise of yielding future advances.

Abstract: Spontaneous neural activity is a basic property of the developing brain, which regulates key developmental processes, including migration, neural differentiation and formation and refinement of connections The mechanisms regulating spontaneous activity are not known By using transgenic embryos that overexpress BDNF under the control of the nestin promoter, we show here that BDNF controls the emergence and robustness of spontaneous activity in embryonic hippocampal slices Further, BDNF dramatically increases spontaneous co-active network activity, which is believed to synchronize gene expression and synaptogenesis in vast numbers of neurons In fact, BDNF raises the spontaneous activity of E18 hippocampal neurons to levels that are typical of postnatal slices We also show that BDNF overexpression increases the number of synapses at much earlier stages (E18) than those reported previously Most of these synapses were GABAergic, and GABAergic interneurons showed hypertrophy and a 3-fold increase in GAD expression Interestingly, whereas BDNF does not alter the expression of GABA and glutamate ionotropic receptors, it does raise the expression of the recently cloned K(+)/Cl(-) KCC2 co-transporter, which is responsible for the conversion of GABA responses from depolarizing to inhibitory, through the control of the Cl(-) potential Together, results indicate that both the presynaptic and postsynaptic machineries of GABAergic circuits may be essential targets of BDNF actions to control spontaneous activity The data indicate that BDNF is a potent regulator of spontaneous activity and co-active networks, which is a new level of regulation of neurotrophins Given that BDNF itself is regulated by neuronal activity, we suggest that BDNF acts as a homeostatic factor controlling the emergence, complexity and networking properties of spontaneous networks

Abstract: To elucidate the co-evolutionary relationships between phloem-feeding insects and their secondary, or facultative, bacterial symbionts, we explore the distributions of three such microbes--provisionally named the R-type (or PASS, or S-sym), T-type (or PABS), and U-type--across a number of aphid and psyllid hosts through the use of diagnostic molecular screening techniques and DNA sequencing. Although typically maternally transmitted, phylogenetic and pairwise divergence analyses reveal that these bacteria have been independently acquired by a variety of unrelated insect hosts, indicating that horizontal transfer has helped to shape their distributions. Based on the high genetic similarity between symbionts in different hosts, we argue that transfer events have occurred recently on an evolutionary timescale. In several instances, however, closely related symbionts associate with related hosts, suggesting that horizontal transfer between distant relatives may be rarer than transmission between close relatives. Our findings on the prevalence of these symbionts within many aphid taxa, along with published observations concerning their effects on host fitness, imply a significant role of facultative symbiosis in aphid ecology and evolution.

Abstract: Despite copper ions being crucial in proteins participating in plant processes such as electron transport, free-radical elimination and hormone perception and signaling, very little is known about copper inward transport across plant membranes. In this work, a five-member family (COPT1–5) of putative Arabidopsis copper transporters is described. We ascertain the ability of these proteins to functionally complement and transport copper in the corresponding Saccharomyces cerevisiae high-affinity copper transport mutant. The specific expression pattern of the Arabidopsis COPT1–5 mRNA in different tissues was analyzed by RT-PCR. Although all members are ubiquitously expressed, differences in their relative abundance in roots, leaves, stem and flowers have been observed. Moreover, steady-state COPT1 and COPT2 mRNA levels, the members that are most efficacious in complementing the S. cerevisiae high-affinity copper transport mutant, are down-regulated under copper excess, consistent with a role for these proteins in copper transport in Arabidopsis cells.
