Abstract: Background Cigarette smoking contributes to lung remodelling in chronic obstructive pulmonary disease (COPD). As part of remodelling, peribronchiolar fibrosis is observed in the small airways of patients with COPD and contributes to airway obstruction. Epithelial to mesenchymal transition (EMT) appears to be involved in the formation of peribronchiolar fibrosis. This study examines the EMT process in human bronchial epithelial cells (HBECs) from non-smokers, smokers and patients with COPD as well as the in vitro effect of cigarette smoke extract (CSE) on EMT. Methods HBECs from non-smokers (n=5), smokers (n=12) and patients with COPD (n=15) were collected to measure the mesenchymal markers α-smooth muscle actin, vimentin and collagen type I and the epithelial markers E-cadherin, ZO-1 and cytokeratin 5 and 18 by real time-PCR and protein array. In vitro differentiated bronchial epithelial cells were stimulated with CSE. Results Mesenchymal markers were upregulated in HBECs of smokers and patients with COPD compared with non-smokers. In contrast, epithelial cell markers were downregulated. In vitro differentiated HBECs underwent EMT after 72 h of CSE exposure through the activation of intracellular reactive oxygen species, the release and autocrine action of transforming growth factor β 1 , the phosphorylation of ERK1/2 and Smad3 and by the downregulation of cyclic monophosphate. Conclusions The EMT process is present in bronchial epithelial cells of the small bronchi of smokers and patients with COPD and is activated by cigarette smoke in vitro.

Abstract: A double logistic function has been used to describe global inventory mapping and monitoring studies (GIMMS) normalized difference vegetation index (NDVI) yearly evolution for the 1981 to 2003 period, in order to estimate land surface phenology parameter. A principal component analysis on the resulting time series indicates that the first components explain 36, 53 and 37% of the variance for the start, end and length of growing season, respectively, and shows generally good spatial homogeneity. Mann-Kendall trend tests have been carried out, and trends were estimated by linear regression. Maps of these trends show a global advance in spring dates of 0.38 days per year, a global delay in autumn dates of 0.45 days per year and a global increase of 0.8 days per year in the growing seasons validated by comparison with previous works. Correlations between retrieved phenological parameters and climate indices generally showed a good spatial coherence.

Abstract: We report on the gamma-ray activity of the blazar Mrk 501 during the first 480 days of Fermi operation. We find that the average LAT gamma-ray spectrum of Mrk 501 can be well described by a single power-law function with a photon index of 1.78 +/- 0.03. While we observe relatively mild flux variations with the Fermi-LAT (within less than a factor of 2), we detect remarkable spectral variability where the hardest observed spectral index within the LAT energy range is 1.52 +/- 0.14, and the softest one is 2.51 +/- 0.20. These unexpected spectral changes do not correlate with the measured flux variations above 0.3GeV. In this paper, we also present the first results from the 4.5-month-long multifrequency campaign (2009 March 15 - August 1) on Mrk 501, which included the VLBA, Swift, RXTE, MAGIC and VERITAS, the F-GAMMA, GASP-WEBT, and other collaborations and instruments which provided excellent temporal and energy coverage of the source throughout the entire campaign. The average spectral energy distribution of Mrk 501 is well described by the standard one-zone synchrotron self-Compton model. In the framework of this model, we find that the dominant emission region is characterized by a size <~ 0.1 pc (comparable within a factor of few to the size of the partially-resolved VLBA core at 15-43 GHz), and that the total jet power (~10^{44} erg s^{-1}) constitutes only a small fraction (~10^{-3}) of the Eddington luminosity. The energy distribution of the freshly-accelerated radiating electrons required to fit the time-averaged data has a broken power-law form in the energy range 0.3GeV-10TeV, with spectral indices 2.2 and 2.7 below and above the break energy of 20GeV. We argue that such a form is consistent with a scenario in which the bulk of the energy dissipation within the dominant emission zone of Mrk 501 is due to relativistic, proton-mediated shocks.

Abstract: This bioessay aims to explain the different effects of maternal ageing and postovulatory oocyte ageing on mammalian oocytes/embryos under the scope of 'the oxygen radical-mitochondrial injury hypothesis of ageing'. This hypothesis assumes a key role in the senescent process of oxygen radical damage to mitochondrial DNA, proteins and lipids. It is proposed that a decrease in intracellular ATP concentrations and glutathione (GSH)/glutathione disulphide (GSSG) ratio together with a concomitant increase in cytosolic Ca2+ are major factors causing the observed detrimental effects of ageing on cytoskeletal fibres, fertilization and embryo development.
