Abstract: Vibratome sections of perfusion-fixed rat and monkey cerebella were incubated with an antiserum to glial fibrillary acidic protein (GFAP). Immunolabelled elements showed in both species a principally similar distribution but the presence of long, transverse branches of Bergmann fibres was found to be characteristic to the monkey cerebellum. It is concluded that current methods enable fine interspecies differences of GFAP-localization to be demonstrated. This bears a relevance to the use of GFAP-immunostaining as a diagnostic tool in neuropathology.

Abstract: Three strains of the Pasteurella aerogenes complex were isolated as sole pathogens from aborted fetuses of a sow aborted at the 12th week of gestation on a farm of 600 sows. Gross pathology showed no characteristic lesions. The isolates were biochemically identical and resembled P. pneumotropica on the basis of their strong indole and urease positivity but they produced gas, were ornithine decarboxylase negative and fermented mannitol but not trehalose. Only a few differences were apparent in biochemical characteristics between the isolated strains and P. aerogenes. They differed from the type strain of P. aerogenes in ornithine decarboxylase activity, indole production and lactose and mannitol fermentation; however, such strains do occur within this heterogeneous species. At the time of abortion the antibody titre of the aborted sow was 1 in 16 when examined with live bacterial suspension and 1 in 128 if boiled antigen was used. Similar strains could not be isolated from the vaginas of aborted sows or pregnant and newly farrowed sows in the same group. The bacteriological, serological and histological findings support the opinion of other workers on the occasional pathogenic nature of P. aerogenes.

Abstract: Summary Ruminal epithelial cells isolated by fractionated trypsinization were examined for the relative proportions of cell types by phase contrast microscopy and electron microscopy, and were assayed for AP and LDH activities. The fractions consisting for the most part of str. corneum and str. granulosum cells showed AP and LDH activities of 2.19 ± 0.22 and 1.15 ± 0.48 kat/mg protein, respectively, while those consisting mainly of str. spinosum and str. basale cells showed activities of 0.22 ± 0.20 and 40.08 ± 6.35 kat/mg protein, respectively. The opposite tendencies of AP and LDH activity changes reflect the degree of differentiation of ruminal epithelial cells, and can thus be utilized as markers of that property. Zusammenfassung Eine Methode zur Identifizierung des Differenzierungsgrades von isolierten Pansenepithelzellen Bei Pansenepithelzellen (isoliert durch fraktionierte Trypsinisierung) wurden mittels Phasenkontrast- und Elektronenmikroskopie die relativen Anteile der Zelltypen sowie deren AP- und LDH-Aktivitaten bestimmt. Bei Fraktionen, die vorwiegend aus Zellen des Stratum corneum und Stratum granulosum bestanden, betrugen die AP- und LDH-Aktivitaten 2,19 ± 0,22 und 1,15 ± 0,48 kat/mg Protein. Fraktionen, die hauptsachlich Zellen des Stratum spinosum und Stratum basale enthielten, zeigten Aktivitaten von 0,22 ± 0,2 und 40,08 ± 6,35 kat/mg Protein. Die gegensatzlichen Entwicklungen der AP- und LDH-Aktivitaten reflektieren den Differenzierungsgrad der Pansenepithelzellen und konnen daher als Marker fur diese Eigenschaft verwendet werden. Les activites AP et LDH ont ete de 2,19 ± 0,22 et 1,15 ± 0,48 kat/mg proteine chez les fractions avec predominance de cellules du Stratum corneum et du Stratum granulosum. Des activites de 0,22 ± 0,2 et 40,08 ± 6,35 kat/mg proteine ont ete observees avec les fractions contenant principalement des cellules du Stratum spinosum et du Stratum basale. Les developpements opposes des activites AP et LDH ont reflete le degre de differenciation des cellules epitheliales de la panse et peuvent etre ainsi utilises comme marque de ces proprietes. Resumen Un metodo para identificar el grado de diferenciacion de las celulas ruminales epiteliales aisladas Se examinaron en celulas ruminales epiteliales (aisladas mediante tripsinizacion fraccionada) por medio de microscopia de contraste de fases y electronica las proporciones relativas de los tipos celulares asi como sus actividades AP y LDH. En las fracciones que consistian en su mayor parte de celulas del estrato corneo y del estrato granuloso, las actividades AP y LDH eran del orden de 2,19 ± 0,22 y 1,15 ± 0,48 kat./mg. de proteina. Las fracciones que contenian principalmente celulas de los estratos espinoso y basal mostraban actividades de 0,22 ± 0,20 y 40,08 ± 6,35 kat./mg. de proteina. Las tendencias contrarias de las actividades AP y LDH reflejan el grado de diferenciacion de las celulas ruminales epiteliales, pudiendose emplear, por lo tanto, como demarcadores de esta propiedad.
