Abstract: Contents In mares, mating-induced persistent endometritis contributes to low fertility. The condition is in part related to delayed clearance of mucus accumulated within the uterine lumen. The objective of this study was to investigate the endometrial response of healthy mares to intrauterine (i.u.) treatment with N-acetylcysteine (NAC). Oestrous mares (n = 12) were randomly assigned to a treatment (TM) or control (C) group and received an i.u. infusion of 5% NAC and saline (total volume 140 ml), respectively. Endometrial biopsies were collected in five of the mares 24 h after treatment, in the remaining seven mares 72 h after treatment. Endometrial biopsies were evaluated for integrity of the luminal epithelium, number of polymorphonuclear neutrophils (PMN), staining for cyclooxygenase 2 (COX2), staining with Kiel 67 antigen (Ki-67), lectins and periodic acid-Schiff (PAS). The integrity of endometrial epithelial cells was not affected by treatment (no statistical differences between groups or times). At 24 h after treatment, the mean number of PMN in endometrial biopsies from NAC- and C-mares did not differ, but at 72 h after treatment, number of PMN was significantly higher (p < 0.05) in C (3.9 ± 0.6 PMN/field) compared with NAC-treated mares (2.3 ± 0.2 PMN/field). At 72 h after treatment, the intensity of staining for COX2 was significantly higher after saline than after NAC treatment (p < 0.05). In the epithelium, no differences in staining for the proliferation marker Ki-67 were seen with respect to time and treatment. Score for the lectin wheat germ agglutinin (WGA) was slightly higher in NAC-treated mares than in C-mares 72 h after treatment (p < 0.05). Score for PAS staining of mucus in deep uterine glands differed significantly between groups at 24 h after treatment (p < 0.05). The present study demonstrates that NAC does not adversely affect the endometrial function. Moreover, an anti-inflammatory effect on the equine endometrium was observed.

Abstract: Summary A disease associated with central nervous system signs was observed in two duck flocks, each consisting of 6000 three-week-old ducklings, a week after their arrival at the receiving farms. The clinical signs included leg weakness, dyspnoea, diarrhoea, and in many cases disturbance of equilibrium, retrograde movement, torticollis and twisting of the neck. Pathological and histopathological examinations revealed lympho-histiocytic meningitis and cerebroventriculitis, airsacculitis with moderate amounts of fibrinous exudate, bronchitis associated with formation of lymphoid follicles, focal or interstitial pneumonia, in some cases with fibrinous serositis, interstitial catarrh and splenomegaly. Large numbers of mycoplasmas were seen by electron microscopy and were isolated from the affected meninges and pericardium. Other diseases were excluded by laboratory examinations. The isolates were identified as Mycoplasma anatis. The pathogenicity and immunogenicity of the strain were investigated in an infection experiment in ducks. Although during this experiment no disease associated with clinical signs or deaths occurred, birds infected into the airsacs or intracranially exhibited pathological lesions similar to, but milder than, those seen in the field outbreak. No immune response was detectable by the serological and cellular tests applied. The pathological significance of M. anatis infection in ducks merits further study. Zusammenfassung Ausbruch einer Mycoplasma anatis-Infektion in Verbindung mit nervosen Symptomen bei Entenbestanden in Massenhaltung In zwei Bestanden mit je 6000 drei Wochen alten Entenkuken wurde eine Erkrankung verbunden mit zentralnervosen Symptomen beobachtet. Zum Symptomenbild gehorten Beinschwache, Atemnot, Durchfall, in vielen Fallen Gleichgewichtsstorung, retrogarde Bewegung, Torticollis und Verdrehung des Halses. Bei der pathologischen und histopathologischen Untersuchung wurden eine lymphohistiozytare Meningitis und Cerebroventriculitis, eine Entzundung der Luftsacke mit geringen Mengen fibrinosen Exsudates, eine fokale oder interstitielle Pneumonie (teilweise mit fibrinoser Serositis), ein interstitieller Katarrh und eine Splenomegalie diagnostiziert. Eine grose Anzahl von Mycoplasmen wurde elektronenmikroskopisch nachgewiesen und aus den affektierten Meningen und dem Perikardium isoliert. Die Isolate wurden als Mycoplasma anatis identifiziert und andere Krankheitsursachen durch Laboruntersuchungen ausgeschlossen. Die Pathogenitat und die immunogenen Eigenschaften des isolierten Stammes wurden in einem Infektionsversuch an gesunden Tieren gepruft. Wahrend des Versuchszeitraumes konnten weder Erkrankungen mit sichtbaren klinischen Anzeichen noch Todesfalle beobachtet werden. Allerdings wurden bei den uber die Luftsacke oder intrakranial infizierten Tieren pathologische Veranderungen festgestellt, die den Befunden der Feldausbruche ahnelten, jedoch milder in der Auspragung waren. Sowohl mit den serologischen als auch den zellularen Testmethoden konnte eine Immunantwort nicht nachgewiesen werden.

Abstract: Trends of certain mastitis markers were studied in udder quarters (n = 201) showing clinical symptoms of acute mastitis. Besides the clinical examination, before the first treatment (baseline sample), and about 3 weeks later, 17 to 24 days following the last treatment (control sample) milk samples were collected for bacteriological identification of the mastitis pathogens and for the determination of certain inflammatory markers: somatic cell count (SCC), N-acetyl-beta-D-glucosaminidase (NAGase) and alpha 1-antitrypsin (ATR) activities, as well as bovine serum albumin (BSA) and chloride (Cl-) concentrations. Based upon the clinical and bacteriological status as well as the SCC recorded at the control investigations, 6 groups were established (recovered, latently infected, subclinical mastitis: bacteriologically positive and negative, as well as clinical mastitis: bacteriologically positive and negative). As compared to the baseline samples, all parameters decreased in the case of recovered udder quarters, as well as in those with abated latent infection or subclinical mastitis at the time of control examination. Comparing the control samples of the different categories, characteristic differences were found in NAGase activity, indicating the grade of cytodamaging effect of mastitis. Of the other markers, ATR and Cl- proved to be more adequate for the differentiation than BSA. It can be concluded that, in addition to SCC, first of all NAGase can be recommended for use as an inflammatory parameter in pharmacodynamic studies. Besides these two parameters, ATR and Cl- can also be chosen as a possible third marker.
