In this paper we try to unify these different examples, which are discussed in detail below. First we give a discription of the results proved in this paper.

Abstract: Cytokeratin polypeptides of human epidermis, of epithelia microdissected from various zones of the pilosebaceous tract (outer root-sheath of hair follicle, sebaceous gland), and of eccrine sweat-glands have been separated by one- and two-dimensional gel electrophoresis and characterized by binding of cytokeratin antibodies and by peptide mapping. The epithelium of the pilosebaceous tract has three major keratin polypeptides in common with interfollicular epidermis (two basic components of mol wts 58,000 and 56,000 and one acidic polypeptide of mol wt 50,000); however, it lacks basic keratin polypeptides in the mol wt range of 64,000-68,000 and two acidic keratin-polypeptides of mol wts 56,000 and 56,500 and contains an additional characteristic acidic cytokeratin of mol wt 46,000. Another cytokeratin polypeptide of mol wt 48,000 that is prominent in hair-follicle epithelium is also found in nonfollicular epidermis of foot sole. Both epidermis and pilosebaceous tract are different from eccrine sweat-gland epithelium, which also contains two major cytokeratins of mol wts 52,500 and 54,000 (isoelectric at pH 5.8-6.1) and a more acidic cytokeratin of mol wt 40,000. A striking similarity between the cytokeratins of human basal-cell epitheliomas and those of the pilosebaceous tract has been found: all three major cytokeratins (mol wts 58,000; 50,000; 46,000) of the tumor cells are also expressed in hair-follicle epithelium. The cytokeratin of mol wt 46,000, which is the most prominent acidic cytokeratin in this tumor, is related, by immunological and peptide map criteria, to the acidic keratin-polypeptides of mol wts 48,000 and 50,000, but represents a distinct keratin that is also found in other human tumor cells such as in solid adamantinomas and in cultured HeLa cells. The results show that the various epithelia present in skin, albeit in physical and ontogenic continuity, can be distinguished by their specific cytokeratin-polypeptide patterns and that the cytoskeleton of basal-cell epitheliomas is related to that of cells of the pilosebaceous tract.

Abstract: Two molecular dynamics simulations of 216 molecules interacting by the ST2 water potential were carried out. Apart from technical improvements, the first simulation was identical to that reported by Stillinger and Rahman [4]. In the second, however, the reaction field method was applied and the influence of long range interactions was carefully studied. 7500 time steps (= 7·5 ps) were generated. The temperature given by the average kinetic energy was 120°C. The atom-atom pair correlation functions and the average dipole energy are rather insensitive to the reaction field. The average cosine between two molecular dipoles, however, is strongly influenced and corresponding curves are of different sign in the second and third coordination shell. For the integral of the average cosine, namely the Kirkwood g-factor g K, the situation is even worse : Large errors occur if the long range interactions are neglected. Abstract: A rapid and highly sensitive procedure for simultaneous determination of serotonin, 5-hydroxyindoleacetic acid, 3,4-dihydroxyphenylacetic acid and homovanillic acid is described. After precipitation of proteins with perchloric acid the samples are applied directly to a high performance liquid chromatograph, with electrochemical detection. As little as 20 pg of serotonin, 5-hydroxyindoleacetic acid, and 3,4-dihydroxyphenylacetic acid and 200 pg of homovanillic acid can be detected. One chromatographic run requires less than 10 min.

Abstract: Satellite DNAs have been isolated from the monocotyledonous plants Scilla siberica, S. amoena, S. ingridae (all are highly GC-rich), and S. mischtschenkoana by using the Ag+ −Cs2SO4 density centrifugation technique. Hybridization in situ has been performed with 3H-cRNA to these satellite DNAs in all four species. In each species, the endogenous satellite DNA is located mainly in intercalary and major heterochromatin bands associated with terminal regions and nucleolar organizer regions (NORs) but not in centromeric regions. Patterns observed after cross-species hybridization show a high degree of satellite DNA homology between S. siberica, S. amoena, and S. ingridae. By contrast, satellite DNA of S. mischtschenkoana consists largely of different, non homologous DNA sequences, with two exceptions: (i) the NORs of all four species contain similar satellite sequences, and (ii) a strong homology exists between the satellite DNA of S. mischtschenkoana and centromeric DNA of S. siberica but not with those of S. amoena and S. ingridae. — Heterochromatin has also been characterized by the AT-specific fluorochromes quinacrine (Q) and DAPI and the GC-specific agent chromomycin A3 (CMA3), in combination with two counterstaining techniques. While CMA3-fluorescence is largely in agreement with data on base composition and location of the specific satellite DNAs, the results with Q and DAPI are conflicting. Prolonged fixation has been found to change the fluorescence character in certain instances, indicating that other factors than the base sequence of the DNA also play a role in fluorochrome staining of chromosomes. The results are discussed in relation to the taxonomy and phylogeny of the four species.

Abstract: A new human plasmacytoma cell line (Karpas 707) has been established from a myeloma patient. The cultured cells are negative for Epstein-Barr viral nuclear antigen and free of mycoplasma. They are similar to plasma cells and secrete only lambda light chains. The cells are hypodiploid and contain the Philadelphia chromosome and other abnormalities. This cell line may be suitable for the production of human monoclonal antibodies.

Abstract: Peripheral M mode contrast echocardiograms were performed at rest and during the Valsalva maneuver in 11 patients with patent foramen ovale and normal right heart pressures documented by catheterization studies. Positive contrast studies (at least five clearly identifiable contrast echoes appearing in the left heart) were obtained in three patients (27 percent at rest) in six (54 percent) during the Valsalva maneuver and in seven patients (64 percent) either at rest or during the Valsalva maneuver. It is concluded that contrast echocardiography offers an opportunity to diagnose a patent foramen ovale noninvasively in patients with suspected paradoxical embolism or with questionable evidence of atrial septal defect. A positive study result does not specifically indicate heart disease since it occurs in healthy persons with patent foramen ovale, normal intracardiac pressures and no other abnormality.

Abstract: A peptide was isolated in pure form from human leukocytes which strongly inhibits the proliferation of immature myeloid cells in vitro (committed stem cells). Structural investigations yielded pGlu-Asp or Glu-Asp or Glu-Cys-Lys-OH as the probable sequence of this peptide. The Glu2, Asp3-analog, prepared synthetically, displayed similar activities and when applied in vivo showed effects on the hemopoietic system ranging from an inhibition of pluripotent and committed stem cells to variations in the bone marrow proliferation and alterations in peripheral blood counts.

Abstract: The red cell-linked antigen-antiglobulin reaction (RCLAAR) with stearoyldextran-coated erythrocytes was used to characterize the immunoglobulin (Ig) classes and IgG subclasses of dextran reactive antibodies (DRA) in 27 dextran reactors (DR) and 96 on reactors (DNR). High titres of dextran reactive IgG were regularly found in sera of patients with severe dextran-induced anaphylactoid/anaphylactic reactions (DIAR) prior to the infusion. In four lethal cases IgG antibodies were found to be in the highest titre range of 16,384 to 32,768. In addition, high IgA and IgM titres were also in severe DIAR. DNR had much lower titres of dextran reactive antibodies of IgG, IgM and IgA classes and IgD antibody was absent in both groups. Dextran reactive IgE antibodies were not demonstrable in DR. Dextran reactive IgG2, IgG3, IgG4 and IgG1 (indirect measurement) were demonstrated in both DR and FNR. Dextran infusion caused variable neutralization in all Ig classes and IgG subclasses, but the contribution of IgG2 was considered most important because of its high titres and most pronounced neutralization in severe DIAR. It is concluded that DRA mainly of the IgG class, play a critical pathogenic role in the induction of severe DIAR, which accordingly is classified as immune complex (Type III) anaphylaxis. The method of RCLAAR allows to delineate a risk group of about 2% of potential reactors.

Abstract: The linear logistic test model (LLTM), a Rasch model with linear constraints on the item parameters, is described. Three methods of parameter estimation are dealt with, giving special consideration to the conditional maximum likelihood approach, which provides a basis for the testing of structural hypotheses regarding item difficulty. Standard areas of application of the LLTM are surveyed, including many references to empirical studies in item analysis, item bias, and test construction; and a novel type of application to response-contingent dynamic processes is presented. Finally, the linear logistic model with relaxed assumptions (LLRA) for measuring change is introduced as a special case of an LLTM; it allows the characterization of individuals in a multidimensional latent space and the testing of hypotheses regarding effects of treatments.
