Abstract: Good computer and video games like System Shock 2, Deus Ex, Pikmin, Rise of Nations, Neverwinter Nights, and Xenosaga: Episode 1 are learning machines. They get themselves learned and learned well, so that they get played long and hard by a great many people. This is how they and their designers survive and perpetuate themselves. If a game cannot be learned and even mastered at a certain level, it won't get played by enough people, and the company that makes it will go broke. Good learning in games is a capitalist-driven Darwinian process of selection of the fittest. Of course, game designers could have solved their learning problems by making games shorter and easier, by dumbing them down, so to speak. But most gamers don't want short and easy games. Thus, designers face and largely solve an intriguing educational dilemma, one also faced by schools and workplaces: how to get people, often young people, to learn and master something that is long and challenging--and enjoy it, to boot.

Abstract: In a prospective-longitudinal study of a representative birth cohort, we tested why stressful experiences lead to depression in some people but not in others. A functional polymorphism in the promoter region of the serotonin transporter (5-HT T) gene was found to moderate the influence of stressful life events on depression. Individuals with one or two copies of the short allele of the 5-HT T promoter polymorphism exhibited more depressive symptoms, diagnosable depression, and suicidality in relation to stressful life events than individuals homozygous for the long allele. This epidemiological study thus provides evidence of a gene-by-environment interaction, in which an individual's response to environmental insults is moderated by his or her genetic makeup.

Abstract: The goal of the International HapMap Project is to determine the common patterns of DNA sequence variation in the human genome and to make this information freely available in the public domain. An international consortium is developing a map of these patterns across the genome by determining the genotypes of one million or more sequence variants, their frequencies and the degree of association between them, in DNA samples from populations with ancestry from parts of Africa, Asia and Europe. The HapMap will allow the discovery of sequence variants that affect common disease, will facilitate development of diagnostic tools, and will enhance our ability to choose targets for therapeutic intervention.

Abstract: The primary rationale for the use of phylogenetically based statistical methods is that phylogenetic signal, the tendency for related species to resemble each other, is ubiquitous. Whether this assertion is true for a given trait in a given lineage is an empirical question, but general tools for detecting and quantifying phylogenetic signal are inadequately developed. We present new methods for continuous-valued characters that can be implemented with either phylogenetically independent contrasts or generalized least-squares models. First, a simple randomization procedure allows one to test the null hypothesis of no pattern of similarity among relatives. The test demonstrates correct Type I error rate at a nominal α = 0.05 and good power (0.8) for simulated datasets with 20 or more species. Abstract: Occasionally, surprisingly large shifts occur in ecosystems. Theory suggests that such shifts can be attributed to alternative stable states. Verifying this diagnosis is important because it implies a radically different view on management options, and on the potential effects of global change on such ecosystems. For instance, it implies that gradual changes in temperature or other factors might have little effect until a threshold is reached at which a large shift occurs that might be difficult to reverse. Strategies to assess whether alternative stable states are present are now converging in fields as disparate as desertification, limnology, oceanography and climatology. Here, we review emerging ways to link theory to observation, and conclude that although, field observations can provide hints of alternative stable states, experiments and models are essential for a good diagnosis.

Abstract: ContextMore persons in the United States die from non–small cell lung cancer (NSCLC) than from breast, colorectal, and prostate cancer combined. In preclinical testing, oral gefitinib inhibited the growth of NSCLC tumors that express the epidermal growth factor receptor (EGFR), a mediator of cell signaling, and phase 1 trials have demonstrated that a fraction of patients with NSCLC progressing after chemotherapy experience both a decrease in lung cancer symptoms and radiographic tumor shrinkages with gefitinib.ObjectiveTo assess differences in symptomatic and radiographic response among patients with NSCLC receiving 250-mg and 500-mg daily doses of gefitinib.Design, Setting, and PatientsDouble-blind, randomized phase 2 trial conducted from November 2000 to April 2001 in 30 US academic and community oncology centers. Patients (N = 221) had either stage IIIB or IV NSCLC for which they had received at least 2 chemotherapy regimens.InterventionDaily oral gefitinib, either 500 mg (administered as two 250-mg gefitinib tablets) or 250 mg (administered as one 250-mg gefitinib tablet and 1 matching placebo).Main Outcome MeasuresImprovement of NSCLC symptoms (2-point or greater increase in score on the summed lung cancer subscale of the Functional Assessment of Cancer Therapy-Lung [FACT-L] instrument) and tumor regression (>50% decrease in lesion size on imaging studies).ResultsOf 221 patients enrolled, 216 received gefitinib as randomized. Symptoms of NSCLC improved in 43% (95% confidence interval [CI], 33%-53%) of patients receiving 250 mg of gefitinib and in 35% (95% CI, 26%-45%) of patients receiving 500 mg. These benefits were observed within 3 weeks in 75% of patients. Partial radiographic responses occurred in 12% (95% CI, 6%-20%) of individuals receiving 250 mg of gefitinib and in 9% (95% CI, 4%-16%) of those receiving 500 mg. Symptoms improved in 96% of patients with partial radiographic responses. The overall survival at 1 year was 25%. There were no significant differences between the 250-mg and 500-mg doses in rates of symptom improvement (P = .26), radiographic tumor regression (P = .51), and projected 1-year survival (P = .54). The 500-mg dose was associated more frequently with transient acne-like rash (P = .04) and diarrhea (P = .006).ConclusionsGefitinib, a well-tolerated oral EGFR-tyrosine kinase inhibitor, improved disease-related symptoms and induced radiographic tumor regressions in patients with NSCLC persisting after chemotherapy.

Abstract: Clarify is a program that uses Monte Carlo simulation to convert the raw output of statistical procedures into results that are of direct interest to researchers, without changing statistical assumptions or requiring new statistical models. The program, designed for use with the Stata statistics package, offers a convenient way to implement the techniques described in: Gary King, Michael Tomz, and Jason Wittenberg (2000). "Making the Most of Statistical Analyses: Improving Interpretation and Presentation." American Journal of Political Science 44, no. 2 (April 2000): 347-61. We recommend that you read this article before using the software. Clarify simulates quantities of interest for the most commonly used statistical models, including linear regression, binary logit, binary probit, ordered logit, ordered probit, multinomial logit, Poisson regression, negative binomial regression, weibull regression, seemingly unrelated regression equations, and the additive logistic normal model for compositional data. Abstract: Rates and reactions of biogeochemical processes vary in space and time to produce both hot spots and hot moments of elemental cycling. We define biogeochemical hot spots as patches that show disproportionately high reaction rates relative to the surrounding matrix, whereas hot moments are defined as short periods of time that exhibit disproportionately high reaction rates relative to longer intervening time periods. As has been appreciated by ecologists for decades, hot spot and hot moment activity is often enhanced at terrestrial-aquatic interfaces. Using examples from the carbon (C) and nitrogen (N) cycles, we show that hot spots occur where hydrological flowpaths converge with substrates or other flowpaths containing complementary or missing reactants. Hot moments occur when episodic hydrological flowpaths reactivate and/or mobilize accumulated reactants. By focusing on the delivery of specific missing reactants via hydrologic flowpaths, we can forge a better mechanistic understanding of the factors that create hot spots and hot moments. Such a mechanistic understanding is necessary so that biogeochemical hot spots can be identified at broader spatiotemporal scales and factored into quantitative models. We specifically recommend that resource managers incorporate both natural and artificially created biogeochemical hot spots into their plans for water quality management. Finally, we emphasize the needs for further research to assess the potential importance of hot spot and hot moment phenomena in the cycling of different bioactive elements, improve our ability to predict their occurrence, assess their importance in landscape biogeochemistry, and evaluate their utility as tools for resource management.

Abstract: Although it has long been recognized that dynamics in supercooled liquids might be spatially heterogeneous, only in the past few years has clear evidence emerged to support this view. As a liquid is cooled far below its melting point, dynamics in some regions of the sample can be orders of magnitude faster than dynamics in other regions only a few nanometers away. In this review, the experimental work that characterizes this heterogeneity is described. In particular, the following questions are addressed: How large are the heterogeneities? How long do they last? How much do dynamics vary between the fastest and slowest regions? Why do these heterogeneities arise? The answers to these questions influence practical applications of glass-forming materials, including polymers, metallic glasses, and pharmaceuticals.

Abstract: Parallel processes for patterning densely packed nanometre-scale structures are critical for many diverse areas of nanotechnology. Thin films of diblock copolymers can self-assemble into ordered periodic structures at the molecular scale (approximately 5 to 50 nm), and have been used as templates to fabricate quantum dots, nanowires, magnetic storage media, nanopores and silicon capacitors. Unfortunately, perfect periodic domain ordering can only be achieved over micrometre-scale areas at best and defects exist at the edges of grain boundaries. These limitations preclude the use of block-copolymer lithography for many advanced applications. Graphoepitaxy, in-plane electric fields, temperature gradients, and directional solidification have also been demonstrated to induce orientation or long-range order with varying degrees of success. Here we demonstrate the integration of thin films of block copolymer with advanced lithographic techniques to induce epitaxial self-assembly of domains. The resulting patterns are defect-free, are oriented and registered with the underlying substrate and can be created over arbitrarily large areas. These structures are determined by the size and quality of the lithographically defined surface pattern rather than by the inherent limitations of the self-assembly process. Our results illustrate how hybrid strategies to nanofabrication allow for molecular level control in existing manufacturing processes.

Abstract: The Moderate Resolution Imaging Spectroradiometer (MODIS) is one of five instruments aboard the Terra Earth Observing System (EOS) platform launched in December 1999. After achieving final orbit, MODIS began Earth observations in late February 2000 and has been acquiring data since that time. The instrument is also being flown on the Aqua spacecraft, launched in May 2002. A comprehensive set of remote sensing algorithms for cloud detection and the retrieval of cloud physical and optical properties have been developed by members of the MODIS atmosphere science team. The archived products from these algorithms have applications in climate change studies, climate modeling, numerical weather prediction, as well as fundamental atmospheric research. In addition to an extensive cloud mask, products include cloud-top properties (temperature, pressure, effective emissivity), cloud thermodynamic phase, cloud optical and microphysical parameters (optical thickness, effective particle radius, water path), as well as derived statistics. We will describe the various algorithms being used for the remote sensing of cloud properties from MODIS data with an emphasis on the pixel-level retrievals (referred to as Level-2 products), with 1-km or 5-km spatial resolution at nadir. An example of each Level-2 cloud product from a common data granule (5 min of data) off the coast of South America will be discussed. Future efforts will also be mentioned. Relevant points related to the global gridded statistics products (Level-3) are highlighted though additional details are given in an accompanying paper in this issue.

Abstract: One of the most pervasive challenges in molecular phylogenetics is the incongruence between phylogenies obtained using different data sets, such as individual genes. To systematically investigate the degree of incongruence, and potential methods for resolving it, we screened the genome sequences of eight yeast species and selected 106 widely distributed orthologous genes for phylogenetic analyses, singly and by concatenation. Our results suggest that data sets consisting of single or a small number of concatenated genes have a significant probability of supporting conflicting topologies. By contrast, analyses of the entire data set of concatenated genes yielded a single, fully resolved species tree with maximum support. Comparable results were obtained with a concatenation of a minimum of 20 genes; substantially more genes than commonly used but a small fraction of any genome. These results have important implications for resolving branches of the tree of life.

Abstract: The Atmospheric Infrared Sounder (AIRS), the Advanced Microwave Sounding Unit (AMSU), and the Humidity Sounder for Brazil (HSB) form an integrated cross-track scanning temperature and humidity sounding system on the Aqua satellite of the Earth Observing System (EOS). AIRS is an infrared spectrometer/radiometer that covers the 3.7-15.4-/spl mu/m spectral range with 2378 spectral channels. AMSU is a 15-channel microwave radiometer operating between 23 and 89 GHz. HSB is a four-channel microwave radiometer that makes measurements between 150 and 190 GHz. In addition to supporting the National Aeronautics and Space Administration's interest in process study and climate research, AIRS is the first hyperspectral infrared radiometer designed to support the operational requirements for medium-range weather forecasting of the National Ocean and Atmospheric Administration's National Centers for Environmental Prediction (NCEP) and other numerical weather forecasting centers. AIRS, together with the AMSU and HSB microwave radiometers, will achieve global retrieval accuracy of better than 1 K in the lower troposphere under clear and partly cloudy conditions. This paper presents an overview of the science objectives, AIRS/AMSU/HSB data products, retrieval algorithms, and the ground-data processing concepts. The EOS Aqua was launched on May 4, 2002 from Vandenberg AFB, CA, into a 705-km-high, sun-synchronous orbit. Based on the excellent radiometric and spectral performance demonstrated by AIRS during prelaunch testing, which has by now been verified during on-orbit testing, we expect the assimilation of AIRS data into the numerical weather forecast to result in significant forecast range and reliability improvements.

Abstract: Three studies address unresolved issues in value-behavior relations. Does the full range of different values relate to common, recurrent behaviors? Which values relate more strongly to behavior than others? Do relations among different values and behaviors exhibit a meaningful overall structure? If so, how to explain this? We find that stimulation and tradition values relate strongly to the behaviors that express them; hedonism, power, universalism, and self-direction values relate moderately; and security, conformity, achievement, and benevolence values relate only marginally. Additional findings suggest that these differences in value-behavior relations may stem from normative pressures to perform certain behaviors. Such findings imply that values motivate behavior, but the relation between values and behaviors is partly obscured by norms. Abstract: Glial cell line-derived neurotrophic factor (GDNF) is a potent neurotrophic factor with restorative effects in a wide variety of rodent and primate models of Parkinson disease, but penetration into brain tissue from either the blood or the cerebro-spinal fluid is limited. Here we delivered GDNF directly into the putamen of five Parkinson patients in a phase 1 safety trial. One catheter needed to be repositioned and there were changes in the magnetic resonance images that disappeared after lowering the concentration of GDNF. After one year, there were no serious clinical side effects, a 39% improvement in the off-medication motor sub-score of the Unified Parkinson's Disease Rating Scale (UPDRS) and a 61% improvement in the activities of daily living sub-score. Medication-induced dyskinesias were reduced by 64% and were not observed off medication during chronic GDNF delivery. Positron emission tomography (PET) scans of [(18)F]dopamine uptake showed a significant 28% increase in putamen dopamine storage after 18 months, suggesting a direct effect of GDNF on dopamine function. This study warrants careful examination of GDNF as a treatment for Parkinson disease.

Abstract: This paper challenges the findings of Frankel, Johnson and Nelson (FJN) (2002). The results of our discretionary accruals tests differ from FJN's when we adjust discretionary current accruals for firm performance. In our earnings benchmark tests, in contrast to FJN we find no statistically significant association between firms meeting analyst forecasts and auditor fees. Our market reaction tests also provide different results than those reported by FJN. Overall, our study indicates that FJN's results are sensitive to research design choices, and we find no systematic evidence supporting their claim that auditors violate their independence as a result of clients purchasing relatively more nonaudit services.

Abstract: Conservation decisions about how, when, and where to act are typically based on our expectations for the future. When the world is highly unpredictable and we are working from a limited range of expecta- tions, however, our expectations will frequently be proved wrong. Scenario planning offers a framework for developing more resilient conservation policies when faced with uncontrollable, irreducible uncertainty. A scenario in this context is an account of a plausible future. Scenario planning consists of using a few con- trasting scenarios to explore the uncertainty surrounding the future consequences of a decision. Ideally, sce- narios should be constructed by a diverse group of people for a single, stated purpose. Scenario planning can incorporate a variety of quantitative and qualitative information in the decision-making process. Often, con- sideration of this diverse information in a systemic way leads to better decisions. Furthermore, the participa- tion of a diverse group of people in a systemic process of collecting, discussing, and analyzing scenarios builds shared understanding. The robustness provided by the consideration of multiple possible futures has served several groups well; we present examples from business, government, and conservation planning that illustrate the value of scenario planning. For conservation, major benefits of using scenario planning are (1) increased understanding of key uncertainties, (2) incorporation of alternative perspectives into conservation planning, and (3) greater resilience of decisions to surprise.

It provides a theoretical approach to dynamical systems and chaos written for a diverse student population among the fields of mathematics, science, and engineering. Prominent experts provide everything students need to know about dynamical systems as students seek to develop sufficient mathematical skills to analyze the types of differential equations that arise in their area of study. The authors provide rigorous exercises and examples clearly and easily by slowly introducing linear systems of differential equations. Calculus is required as specialized advanced topics not usually found in elementary differential equations courses are included, such as exploring the world of discrete dynamical systems and describing chaotic systems. This is a classic text by three of the world's most prominent mathematicians. It continues the tradition of expository excellence. It contains updated material and expanded applications for use in applied studies.

Abstract: Purpose: The authors investigate the impact of hearing loss on quality of life in a large population of older adults. Design and Methods: Data are from the 5-year follow-up Epidemiology of Hearing Loss Study, a population-based longitudinal study of agerelated hearing impairment conducted in Beaver Dam, WI. Participants (N ¼ 2,688) were 53–97 years old (mean ¼ 69 years) and 42% were male. Difficulties with communication were assessed by using the Hearing Handicap for the Elderly—Screening version (HHIE-S), with additional questions regarding communication difficulties in specific situations. Health-related quality of life was assessed by using measures of activities of daily living (ADLs), instrumental ADLs (IADLs) and the Short Form 36 Health Survey (SF-36). Hearing loss measured by audiometry was categorized on the basis of the puretone average of hearing thresholds at 0.5, 1, 2, and 4 kHz. Results: Of participants, 28% had a mild hearing loss and 24% had a moderate to severe hearing loss. Severity of hearing loss was significantly associated with having a hearing handicap and with self-reported communication difficulties. Individuals with moderate to severe hearing loss were more likely than individuals without hearing loss to have impaired ADLs and IADLs. Severity of hearing loss was significantly associated with decreased function in both the Mental Component Summary score and the Physical Component Summary score of the SF-36 as well as with six of the eight individual domain scores. Implications: Severity of hearing loss is associated with reduced quality of life in older adults.

Abstract: The state explosion problem remains a major hurdle in applying symbolic model checking to large hardware designs. State space abstraction, having been essential for verifying designs of industrial complexity, is typically a manual process, requiring considerable creativity and insight.In this article, we present an automatic iterative abstraction-refinement methodology that extends symbolic model checking. In our method, the initial abstract model is generated by an automatic analysis of the control structures in the program to be verified. Abstract models may admit erroneous (or "spurious") counterexamples. We devise new symbolic techniques that analyze such counterexamples and refine the abstract model correspondingly. We describe aSMV, a prototype implementation of our methodology in NuSMV. Practical experiments including a large Fujitsu IP core design with about 500 latches and 10000 lines of SMV code confirm the effectiveness of our approach.
