Abstract: SYNOPSIS: In this paper, we investigate the association between the audit committee’s oversight of the internal audit function (IAF) and the nature of IAF activities. The importance of internal controls and of the roles of both the audit committee and the internal audit function in monitoring control activities have grown in recent years. Despite the importance of these topics, relatively little regulatory or best practices guidance addresses the distribution of IAF activities and amount of audit committee involvement with the IAF. We hypothesize that when the balance of oversight over the IAF lies with the audit committee (vis-a`-vis management), the committee will steer the IAF toward a more internal-controls-oriented focus. Our hypothesis is based on the existing practice guidance in this area and the relative incentives of management and the audit committee. Abstract: This essay examines multitudinous definitions of ‘communication’ in the light of the meaning of ‘concept’ as reflected in the literature of the philosophy of science. The examination produced 15 main themes from the definitions. Among the 15 conceptual components there are three upon which the definitions rather critically divide. These three points of conceptual split are examined for their impact on theory construction in communication. Some suggestions are made for conceptual clarification.

Abstract: SARS-CoV-2 infection is controlled by the opening of the spike protein receptor binding domain (RBD), which transitions from a glycan-shielded 'down' to an exposed 'up' state to bind the human angiotensin-converting enzyme 2 receptor and infect cells. While snapshots of the 'up' and 'down' states have been obtained by cryo-electron microscopy and cryo-electron tomagraphy, details of the RBD-opening transition evade experimental characterization. Here over 130 µs of weighted ensemble simulations of the fully glycosylated spike ectodomain allow us to characterize more than 300 continuous, kinetically unbiased RBD-opening pathways. Together with ManifoldEM analysis of cryo-electron microscopy data and biolayer interferometry experiments, we reveal a gating role for the N-glycan at position N343, which facilitates RBD opening. Residues D405, R408 and D427 also participate. The atomic-level characterization of the glycosylated spike activation mechanism provided herein represents a landmark study for ensemble pathway simulations and offers a foundation for understanding the fundamental mechanisms of SARS-CoV-2 viral entry and infection.
