Abstract: miRNAs were recently implicated in the pathogenesis of numerous diseases, including neurological disorders such as Parkinson's disease (PD). miRNAs are abundant in the nervous system, essential for efficient brain function and play important roles in neuronal patterning and cell specification. To further investigate their involvement in the etiology of PD, we conducted miRNA expression profiling in peripheral blood mononuclear cells (PBMCs) of 19 patients and 13 controls using microarrays. We found 18 miRNAs differentially expressed, and pathway analysis of 662 predicted target genes of 11 of these miRNAs revealed an over-representation in pathways previously linked to PD as well as novel pathways. To narrow down the genes for further investigations, we undertook a parallel approach using chromatin immunoprecipitation-sequencing (ChIP-seq) analysis to uncover genome-wide interactions of α-synuclein, a molecule with a central role in both monogenic and idiopathic PD. Convergence of ChIP-seq and miRNomics data highlighted the glycosphingolipid biosynthesis and the ubiquitin proteasome system as key players in PD. We then tested the association of target genes belonging to these pathways with PD risk, and identified nine SNPs in USP37 consistently associated with PD susceptibility in three genome-wide association studies (GWAS) datasets (0.46≤OR≤0.63) and highly significant in the meta-dataset (3.36×10−4

Abstract: This study investigates the multilevel interplay among team-level, job-related, and individual characteristics in stimulating employees' innovative work behavior (IWB) based on the theoretical frameworks of achievement goal theory (AGT) and job characteristics theory (JCT). A multilevel two-source study of 240 employees and their 34 direct supervisors in two medium-sized Slovenian companies revealed significant two- and three-way interactions, where a mastery climate, task interdependence, and decision autonomy moderated the relationship between knowledge hiding and IWB. When employees hide knowledge, a team mastery climate only facilitates high levels of IWB if accompanied by either high task interdependence or high decision autonomy. In the absence of one of these job characteristics, knowledge hiding prevents higher levels of IWB even in the case of strong team mastery climate. The results suggest that multiple job design antecedents are necessary to neutralize the negative influence of knowledge hiding on micro-innovation processes within organizations.

Abstract: The use of probiotics has been suggested in the treatment of acute gastroenteritis (AGE) in addition to early rehydration and avoidance of dietary restrictions. This document provides recommendations for the use of probiotics for the treatment of AGE in previously healthy infants and children based on a systematic review of previously completed systematic reviews and of randomized controlled trials (RCTs) published subsequently to these reviews. The recommendations were formulated only if at least 2 RCTs that used a given probiotic (with strain specification) were available. The GRADE system developed by the Grading of Recommendations, Assessment, Development, and Evaluations Working Group, was used to grade the strength of evidence and grades of recommendations used in these guidelines. It offers 4 categories of the quality of the evidence (high, moderate, low, and very low) and 2 categories of the strength of recommendation (strong or weak). The use of the following probiotics (in alphabetical order) may be considered in the management of children with AGE in addition to rehydration therapy: Lactobacillus rhamnosus GG (low quality of evidence, strong recommendation) and Saccharomyces boulardii (low quality of evidence, strong recommendation). Less compelling evidence is available for Lactobacillus reuteri DSM 17938 (very low quality of evidence, weak recommendation) and heat-inactivated Lactobacillus acidophilus LB (very low quality of evidence, weak recommendation). The latter, although traditionally discussed with other probiotics, does not fit with the definition of probiotics. Other strains or combinations of strains have been tested, but evidence of their efficacy is weak or preliminary.

Abstract: We demonstrate the supramolecular and structural equivalence of two halogen-bond donors (I and Br) and three acceptors (O, NH and S) through the synthesis of seven isostructural halogen-bonded cocrystals, involving six different molecules: 1,4-dibromo- and 1,4-diiodotetrafluorobenzene (donors) and thiomorpholine, thioxane, morpholine, and piperazine (acceptors) The formation of isostructural cocrystals indicates how cocrystallization may be used to overcome shape and functional group dissimilarities that control molecular arrangement in the solid state The differences in composition between the seven isostructural cocrystals directly affect the strength and nature of halogen bonds between their constituents, allowing the systematic variation of cocrystal physical properties, in particular the melting point, without affecting their crystal structure Replacement of each O or S halogen-bond acceptor with an NH group provided an approximate 70 degrees C increase in melting point, whereas the replacement of I with Br as the halogen-bond donor lowered the melting point of the resulting solid by a similar amount

Abstract: Summary The effect of extraction solvents (30, 50 and 70 % aqueous solutions of ethanol and acetone, and 100 % distilled water), extraction temperature (60 and 90 °C) and extraction time (30, 60 and 90 min) on the composition and mass fraction of polyphenolic compounds in Dalmatian wild sage (Salvia officinalis L.) extracts has been investigated. The total polyphenolic content of sage extracts was determined spectrophotometrically using Folin-Ciocalteu method, whereas the individual polyphenols were determined by HPLC UV/PDA method. Results indicated that the main polyphenols in sage extracts were vanillic, caffeic, syringic, salvianolic K and salvianolic I acids, methyl rosmarinate, 6-hydroxyluteolin-7-glucoside, luteolin-7-glucuronide, luteolin-7-glucoside, apigenin-7-glucunoride, apigenin-7-glucoside, with rosmarinic acid and luteolin-3-glucuronide as predominant compounds. The mass fractions of total and individual polyphenols significantly depend on the type of extraction solvent, solvent composition and extraction temperature. The results showed that binary solvent systems are more efficient than mono-solvent systems in the extraction of polyphenolic compounds in regard to their relative polarity. The aqueous solutions of ethanol or acetone (30 %), extraction temperature of 60 °C and extraction time of 30 min were the most efficient for the extraction of polyphenols from dry sage leaves.
