Abstract: Objective. To analyze the clinical and immunologic manifestations of antiphospholipid syndrome (APS) in a large cohort of patients and to define patterns of disease expression. Methods. The clinical and serologic features of APS (Sapporo preliminary criteria) in 1,000 patients from 13 European countries were analyzed using a computerized database. Results. The cohort consisted of 820 female patients (82.0%) and 180 male patients (18.0%) with a mean +/- SD age of 42 +/- 14 years at study entry. "Primary" APS was present in 53.1% of the patients; APS was associated with systemic lupus erythematosus (SLE) in 36.2%, with lupus-like syndrome in 5.0%, and with other diseases in 5.9%. A variety of thrombotic manifestations affecting the majority of organs were recorded. A catastrophic APS occurred in 0.8% of the patients. Patients with APS associated with SLE had more episodes of arthritis and livedo reticularis, and more frequently exhibited thrombocytopenia and leukopenia. Female patients had a higher frequency of arthritis, livedo reticularis, and migraine. Male patients had a higher frequency of myocardial infarction, epilepsy, and arterial thrombosis in the lower legs and feet. In 28 patients (2.8%), disease onset occurred before age 15; these patients had more episodes of chorea and jugular vein thrombosis than the remaining patients. In 127 patients (12.7%), disease onset occurred after age 50; most of these patients were men. These patients had a higher frequency of stroke and angina pectoris, but a lower frequency of livedo reticularis, than the remaining patients. Conclusion. APS may affect any organ of the body and display a broad spectrum of manifestations. An association with SLE, the patient's sex, and the patient's age at disease onset can modify the disease expression and define specific subsets of APS.

Abstract: This paper discusses an important issue related to the implementation and interpretation of the analysis scheme in the ensemble Kalman filter. It is shown that the observations must be treated as random variables at the analysis steps. That is, one should add random perturbations with the correct statistics to the observations and generate an ensemble of observations that then is used in updating the ensemble of model states. Traditionally, this has not been done in previous applications of the ensemble Kalman filter and, as will be shown, this has resulted in an updated ensemble with a variance that is too low. This simple modification of the analysis scheme results in a completely consistent approach if the covariance of the ensemble of model states is interpreted as the prediction error covariance, and there are no further requirements on the ensemble Kalman filter method, except for the use of an ensemble of sufficient size. Thus, there is a unique correspondence between the error statistics from the ensemble Kalman filter and the standard Kalman filter approach.

Abstract: We develop a systematic method for renormalizing the AdS/CFT prescription for computing correlation functions. This involves regularizing the bulk on-shell supergravity action in a covariant way, computing all divergences, adding counterterms to cancel them and then removing the regulator. We explicitly work out the case of pure gravity up to six dimensions and of gravity coupled to scalars. The method can also be viewed as providing a holographic reconstruction of the bulk spacetime metric and of bulk fields on this spacetime, out of conformal field theory data. Knowing which sources are turned on is sufficient in order to obtain an asymptotic expansion of the bulk metric and of bulk fields near the boundary to high enough order so that all infrared divergences of the on-shell action are obtained. To continue the holographic reconstruction of the bulk fields one needs new CFT data: the expectation value of the dual operator. In particular, in order to obtain the bulk metric one needs to know the expectation value of stress-energy tensor of the boundary theory. We provide completely explicit formulae for the holographic stress-energy tensors up to six dimensions. We show that both the gravitational and matter conformal anomalies of the boundary theory are correctly reproduced. We also obtain the conformal transformation properties of the boundary stress-energy tensors.

Abstract: Local accumulation of the plant growth regulator auxin mediates pattern formation in Arabidopsis roots and influences outgrowth and development of lateral root- and shoot-derived primordia. However, it has remained unclear how auxin can simultaneously regulate patterning and organ outgrowth and how its distribution is stabilized in a primordium-specific manner. Here we show that five PIN genes collectively control auxin distribution to regulate cell division and cell expansion in the primary root. Furthermore, the joint action of these genes has an important role in pattern formation by focusing the auxin maximum and restricting the expression domain of PLETHORA (PLT) genes, major determinants for root stem cell specification. In turn, PLT genes are required for PIN gene transcription to stabilize the auxin maximum at the distal root tip. Our data reveal an interaction network of auxin transport facilitators and root fate determinants that control patterning and growth of the root primordium.
