Abstract: During ischemia, the cell structures are progressively damaged, but restoration of the blood flow, paradoxically, intensifies the lesions caused by the ischemia. The mechanisms of ischemia injury and reperfusion (I/R) have not been completely defined and many studies have been realized in an attempt to find an ideal therapy for mesenteric I/R. The occlusion and reperfusion of the splanchnic arteries provokes local and systemic alterations principally derived from the release of cytotoxic substances and the interaction between neutrophils and endothelial cells. Substances involved in the process are discussed in the present review, like oxygen-derived free radicals, nitric oxide, transcription factors, complement system, serotonin and pancreatic proteases. The mechanisms of apoptosis, alterations in other organs, therapeutic and evaluation methods are also discussed.

Abstract: O presente estudo de revisao visa uniformizar a nomenclatura e os conceitos classificatorios existentes na literatura, relativos aos desenhos ou modelos de estudo cientifico mais comumente utilizados em pesquisas na area clinico-cirurgica geral. Procurou-se facilitar a elaboracao do delineamento de um projeto de pesquisa cientifica, visando sua aplicabilidade. Esta revisao tambem tem o intuito de auxiliar o cirurgiao-pesquisador a identificar qual foi o delineamento proposto em um artigo, assim como desenvolver um senso de critica metodologica mais apurada sobre o mesmo, alem de desmistificar a miscelânea conceitual existente na literatura.

Abstract: OBJETIVO: Delinear os parâmetros que norteiam a escolha de um modelo animal de doenca e verificar na literatura biomedica recente quais as especies animais de uso mais frequente. METODOS: Considerando a revisao da literatura sao discutidos, os conceitos e as caracteristicas de um modelo animal de doenca. Destaca-se o consenso atual sobre quando usar um modelo animal, quais os criterios de sua escolha e os requisitos para um modelo adequado. Descreve quais os tipos de modelos animal de doenca e discute as controversias da similaridade filogenetica e os riscos inerentes a extrapolacao dos modelos para os seres humanos. Baseado em uma pesquisa documental na base de dados da BIREME (Medline, Lilacs, Scielo e Biblioteca Cochrane) investiga quais os animais de experimentacao mais citados nos artigos destas bases de dados. RESULTADOS: Verificou-se que o rato e o camundongo sao os animais mais frequentemente utilizados. O coelho, cao e o suino seguem a lista nas referencias de lingua inglesa. Nas bases de dados da literatura Latino-americana o cao supera o numero de citacoes de coelhos e suinos. Os primatas sao minoria nas citacoes em todas as bases de dados. As revisoes sistematicas tambem tem no rato o maior numero de citacoes, as demais especies sao citadas em igualdade de condicoes. CONCLUSOES: O animal de experimentacao e usado virtualmente em todos os campos da pesquisa biologica nos dias de hoje. A relacao entre os humanos e os animais de outras especies ganhou, atraves dos tempos, contornos mais definidos, a exploracao de outras especies tem regras e uma etica estabelecida, a inducao dos resultados do animal para a especie humana tem criterios claros e objetivos a serem preenchidos. O uso dos modelos experimentais, ou dos modelos animal de doenca, ou dos animais de laboratorio na pesquisa biomedica permite e deverao permitir nos proximos anos discussoes epistemologicas, politicas, sociais, economicas e religiosas.

Abstract: INTRODUCTION: In Brazilian countryside, cataplasm made from Passiflora edulis leaves has been used by the population as a healing agent for infections and skin inflammations in an empiric basis. PURPOSE: the aim of this work was to evaluate the healing process of open wounds in rats, in which Passiflora edulis hydro-alcoholic extract was applied. METHODS: Sixty male, adult Wistar rats were divided into two groups: Passiflora group and Control group. Rats of the first group were treated with Passiflora edulis extract, and those of the second group received distilled water. The daily application of the extract or distilled water was carried out on a 2 cm diameter standardized circular wound on the dorsal region of each animal. Wound assessment was performed macroscopically and microscopically on the 7th, 14th, and 21st postoperative days. Microscopic analysis included hematoxylin-eosine and Masson Trichromium stains, evaluating inflammatory response, fibroplasia and collagen deposition. The wound retraction was evaluated by digital planimetry. RESULTS: No significant difference in the rate of wound healing was detected comparing both groups. However, a significant increase in the number of fibroblastic cells was seen on the 7th PO day, and significantly greater collagen deposition was observed on the 14th day PO day in rats from the Passiflora group (p=0,012). CONCLUSIONS: The application of the Passiflora edulis extract does not accelerate the healing process of open wounds in rats, but is associated with increased number of fibroblastic cells on 7th P.O. day and greater collagen deposition on the 14th PO day.

Abstract: Purpose: To evaluate and characterize the wound healing process profile induced by allantoin incorporated in soft lotion oil/water emulsion using the planimetric and histological methods. Methods: Female Wistar rats (n=60) were randomly assigned to 3 experimental groups: (C) control group-without treatment; (E) group treated with soft lotion O/W emulsion excipients; (EA) group treated with soft lotion O/W emulsion containing allantoin 5%. The emulsions either containing or not allantoin were topically administered for 14 days and the wound area was evaluated by planimetry and by qualitative and quantitative histological analysis of open wound model. Results: The data which were obtained and analyzed innovate by demonstrating, qualitatively and quantitatively, by histological analysis, the profile of healing process induced by allantoin. The results suggest that the wound healing mechanism induced by allantoin occurs via the regulation of inflammatory response and stimulus to fibroblastic proliferation and extracellular matrix synthesis. Conclusion: This work show, for the first time, the histological wound healing profile induced by allantoin in rats and demonstrated that it is able to ameliorate and fasten the reestablishment of the normal skin.
