Abstract: Three HRGC systems (1: 30m DB-XLB capillary with MS-SIM detection; 2: 60m DB-XLB capillary with full-scan, ion-trap MS detection; and 3: Parallel dual-column DB-17 and series-coupled HP5/HT5 with ECD detection) were used to completely characterize multiple lots of 8 different-numbered Aroclor mixtures by quantitative calibration against 9 solutions containing primary standards of all 209 PCB congeners. Despite lower absolute sensitivity and more Aroclor congener coelutions than the dual-column ECD system, the MS systems enabled measurement of more congeners per Aroclor since their greater linear response range did not require dilution of samples and standards. Pairs of different lots of Aroclors 1248 and 1254 displayed markedly different proportions of congeners, and the 1254 pair displayed strong differences in the extent of ortho-chlorine substitution. The tables of congener weight percent distributions among Aroclors are more comprehensive and quantitatively precise than those of prior publications. However, the limitations of single-level calibration precluded measurement of all congeners to the ±10% accuracy desirable for establishing these Aroclors as secondary standards for comprehensive, quantitative congener-specific PCB analysis.

Abstract: Multidimensional separation methodology provides a mechanism for a very substantial increase in the resolving power of chromatographic and related systems. After defining the conditions required for multidimensional separation and reviewing the origin of its high resolving power, a comparison is drawn between the two principal kinds of multidimensional systems, represented by continuous two-dimensional operation and coupled column assemblies. It is shown that their roles are complementary. The two-dimensional system gives a broad powerful overview of a sample while the coupled column system can be used flexibly to magnify and resolve selected regions of the sample space.

Abstract: A rapid, reliable and precise capillary gas chromatographic method for routine quantification of short- and long-chain free fatty acids (FFA) in milk and cheese is described. Procedures of (1) lipid extraction, (2) isolation of the FFA from milk and cheese extracts, and (3) capillary gas chromatographic analysis were developed and optimized. FFA can be extracted from cheese for 95–100% with ether-heptane after grinding with sodium sulfate and addition of 2.5 M sulfuric acid. From milk, 95–100 % of the FFA (≤ C8:0) are also extracted with ether-heptane after addition of ethanol and 2.5 M sulfuric acid. Internal standards are used to compensate for the losses of lower FFA (C2:0–C8:0) in the aqueous phase. In view of the excellent recovery (98–100 %) and a considerable saving of time, the use of an aminopropyl column is preferred for the isolation of the FFA from lipid-extracts. The underivatized FFA are separated directly by capillary gas chromatography making use of columns which enable accurate and rapid (≤ 40 min) determination of FFA C2:0–C20:0. With the method described, all major FFA (C2:0–C18:3) in milk and cheese can be quantified with good repeatability (rsd less then 2 %). The method is also applicable to the analysis of short-chain fatty acids in other products.

Abstract: Summary The application of a new silica-based, monolithic-type HPLC-column for fast separations is presented. The column is prepared according to a new sol-gel process, which is based on the hydrolysis and polycondensation of alkoxysilanes in the presence of water soluble polymers. The method leads to “rods” made of a single piece of porous silica with a defined pore structure, i. e. macro- and mesopores. The main feature of silica rod columns is a higher total porosity, about 15% higher than of conventional particulate HPLC columns. The resulting column pressure drop is therefore much lower, allowing operation at higher flow rates including flow gradients. Consequently, HPLC analysis can be performed much faster, as it is demonstrated by various applications.

Abstract: Two types of monolithic silica columns derivatized to form an ODS phase, one prepared in a fused silica capillary (SR-FS) and the other prepared in a mold and clad with an engineering plastic (poly-ether-ether-ketone) (SR-PEEK), were evaluated. The column efficiency and pressure drop were compared with those of a column packed with 5-μm ODS-silica particles and of an ODS-silica monolith prepared in a mold and wrapped with PTFE tubing (SR-PTFE). SR-FS gave a lower pressure drop than a column packed with 5-μm particles by a factor of 20, and a plate height of 20 μm at a linear velocity below 1 mm/s. SR-PEEK showed higher flow-resistance than the other monolithic silica columns, but they still showed a minimum plate height of 8-10 μm and a lower pressure drop than popular commercial columns packed with 5-μm particles. The evaluation of SR-FS columns in a CEC mode showed much higher efficiency than in a pressure-driven mode.
