Abstract: Objective To investigate the effects of transient receptor potential M8 (TRPM8) channel on the proliferation and motility of androgen-independent prostate cancer PC-3 cells.Methods After being permanently transfected with an empty vector and cDNA encoding the TRPM8 protein,cells were analysed for cell cycle distribution and motility using flow cytometry and scratch assay.Results Immunocytochemistry and Ca2+ imaging analysis revealed the overexpression of functional TRPM8 channel on both endoplasmic reticulum and plasma membrane of PC-3-TRPM8 cells.Cell cycle distribution and scratch assay analysis revealed that TRPM8 induced cell cycle arrest at the G0/G1 stage ( 71.89% 3.43% vs 54.67%±4.59%、51.48%±3.54%,P0.05 ) and facilitated the cell apoptosis induced by starvation ( 12.56%±1.78% vs 4.67%±1.49%、5.28±1.35%,P0.05 ).Furthermore,TRPM8 inhibited the migration of PC-3-TRPM8 cells (P0.01) through the inactivation of focal-adhesion kinase.Conclusions It appears that TRPM8 was not essential for the survival of PC-3 cells;however,the overexpression of TRPM8 had negative effects on the proliferation and migration of PC-3 cells.Thus,TRPM8 and its agonists may serve as important targets for the treatment of prostate cancer.

Abstract: Objective To evaluate the clinical significance of fluorescence in situ hybridization(FISH)technique for screening bladder transitional cell carcinoma with hematuria,provide a new method for early diagnosis of bladder cancer with cytology of the urine.Methods We selected 20 relatively healthy people as control group.The special urine examination method detects genetic alterations of the urothelial cells found in the urine of the control group,using fluorescent direct labeled DNA probes binding to the peri-centromeric regions of chromosomes 3,7 and 17 as well as on the 9p21 locus,respectively.Thresholds were then established.Morning urine samples collecting from 100 patients with hematuria underwent cytology of the urine and FISH examination,respectively.The sensitivity and specificity of both were analyzed statistically.Results The complete absent,the partial absent and amplification thresholds for p16 gene detected by probes were 3.9%,5.4% and 3.2%,respectively.The absent and amplification thresholds for chromosome 3 were 4.0% and 3.5%;4.0 % and 2.8% for chromosome 7;5.8% and 3.4% for chromosome 17 respectively.The study rated the specificity and sensitivity of the technique 93.6% and 100%,respectively.The study rated the specificity and sensitivity of the urinary exfoliative cytology 46.8% and 100%,respectively.Conclusions This new non-invasive diagnostic application of the FISH technique with high detection rate in the field of urology is prior to urinary cytology.

Abstract: Objective To improve the diagnosis and treatment of von Hippel-Lindau syndrome.Methods Retrograde analyze the diagnosis and treatment of 8 cases of von Hippel-Lindau syndrome,4 received the operations of bilateral focal excision and renal cyst decortications,2 received unilateral focal excision and 2 only received regular follow-up without any operation.Results There were 4 individuals recovery,3 patients survival with tumor and 1 case death.Conclusions Bilateral renal with cystic and solid mass the disease should be considered,focal excision with nephron sparing surgery is the primary therapy and selection pregnancy with gene detection is critical to prevent it occurrence and heredity.

Abstract: Objective To construct the small-molecule double-stranded RNA (dsRNA) expression vector plasmid,and to investigate its activation ability of the expression of cell cycle repressor protein p21WAF1/CIP1(p21) in human prostate cancer cell line PC-3 in vitro.Methods Synthetic the sequence-specific corresponding DNA fragments of the promoter with the p21 gene,as well as the same length of random DNA fragments,by means of the recombinant DNA technology to construct these fragments into the eukaryotic expression plasmids pGenesil-1,constructed with the purpose of the expression of dsRNA fragments plasmid vectors (dsRNAp21-pGenesil-1) and the random fragments of the control dsRNA expression vectors (dsRNACon-pGenesil-1).The dsRNAp21-pGenesil-1 (experimental group),dsRNACon-pGenesil-1 (negative control group) and a blank plasmid pGenesil-1 (control group) were transfected into human prostate cancer cell line PC-3 in vitro respectively with lipofectamine reagent,and transfection efficency was detected by laser scanning confocal microscope.Immunohistochemistry and reverse transcription-PCR were performed to examine the difference expression level of p21 in each group.Results These dsRNAp21-pGenesil-1 and the dsRNACon-pGenesil-1 eukaryotic cell expression vectors were successfully constructed.For each of the groups of plasmids transfected into PC-3 cells,p21 gene expression in the experimental group significantly increased,while there was no significant difference between the negative control group and the blank control group.Conclusions The dsRNAp21-pGenesil-1 express dsRNA molecule successfully,and active the level of expression of p21 mRNA and protein,and provide an effective tool for studying the activation effect of dsRNA.
