Abstract: [Objective] To investigate the protective effect of ginsenoside on acute renal failure(ARF) rats and the correlation between the effect and the changes of nNOS-immunoreactivity in proximal convoluted tubules(PCT).[Methods]Fifty-two male SD rats were randomly divided into four groups: ARF+NS group,ARF+GS group,NS+NS group and NS+GS group.Glycerol-induced acute renal failure in rats was employed.Malondialdehyde(MDA) and nitric oxide(NO) in renal cortex homogenate were measured by commercial regents by experiment in vivo.Meanwhile,the changes of nNOS-IR in the PCT were observed by immunohistochemistry.[Results] In ARF rats treated with physiological saline(ARF+NS group),the level of MDA in renal cortex homogenate significantly increased(P0.05),but NO markedly decreased(P0.05).However,in ARF rats treated with ginsenoside(ARF+GS group) the level of MDA in renal cortex homogenate significantly decreased,but NO markedly increased(P0.05).Immunohistochemistry showed an obvious increase of nNOS-IR in the PCT in ARF+NS group(P0.05),compared with that in NS+NS group;but nNOS-IR was further enhanced in ARF+GS group(P0.05),compared with that in ARF+NS group.[Conclusion] Our results indicated that ginsenoside administrated orally in glycerol-induced ARF rats significantly decreased MDA,increased NO,having a strong antioxidative effect.Our results also indicated that ginsenoside administrated orally could further increase the expression of nNOS-IR in the PCT in ARF rats.Consequently,we provided a new evidence that renal nitronergic neuron in the PCT contributed to the renal protective effect of ginsenoside against acute renal failure.

Abstract: [Objective] This study was to investigate the postoperative complications of Dynamic compressive hip(DHS),which were applied to treatment of the intertrochanteric fracture,and to find how to prevent the complications.[Methods] After reviewing and summarizing 204 patients,to whom the technique of DHS was applied from January 1994 to March 2008,the postoperative complications occurring in 34 cases of the 204 patients were analyzed.[Results] Among the 34 patients,eleven of them had their hip joints ental or lateral extended hip varus or valgus,six patients had lower limbs rotatively deformed,one had lower limb angled deformed,two had the distance between femoral trochanter shorten and screws unscrewed,three with minor trochanter fracture failed to be reduced,one had fracture internal fixed unsuccessfully,three patients had pain in hip joints,two had knee joints stiffened,two had pain in knee joints and three patients had fracture healing delayed.[Conclusion] There are many factors resulting in the complications.The main points are summarized in below.The operations became more difficult if they were delayed.The types of fracture couldn't be analyzed and known correctly and completely.The operations weren't performed according to the technical standard.The characteristics of the instruments were unfamiliar,so they couldn't be performed skillfully.After operation,instructing rehabilitation was neglected or functional exercise wasn't done properly or lifting weight was too early after surgery.

Abstract: [Objective] To study if PGC-1α Gly482Ser polymorphism is related to type 2 diabetes mellitus in Chinese.[Methods] One hundred and forty type 2 diabetes(DM) and 98 non-diabetic controls(NC) were selected.Genotyping of PGC-1α Gly482Ser was determined with PCR amplification,followed by MspI digestion.[Results] PGC-1α Ser/Ser genotype frequency in DM was significantly higher than that in NC(29% vs 12.5%,χ2=10.969,P=0.004).Upon stratification with sex Ser/Ser genotype was significantly higher in DM women than NC women(χ2=11.02,P=0.004),while it was not so in men.Both in NC and DM,fasting C-peptide and 2 hour postprandial C-peptide levels were lower in subjects with Ser/Ser genotype than in subjects with Gly/Gly genotype(P0.05).There was no significance for insulin resistance among different genotypes both in DM and NC groups(P0.05).[Conclusion] The PGC-1α Gly482Ser polymorphism might be related to the development of type 2 diabetes in Chinese women.Ser/Ser seems to be associated with lower endogenous insulin secretion.

Abstract: [Objective] To study the effect of Mosla chinensis Maxim on Trichomonas vaginalis in vitro.[Methods] After treated with Mosla chinensis Maxim water extract(MCMWE),Trichomonas vaginalis was observed under light microscope,for the purpose of morphological changes and mortality rate of Trichomonas vaginalis at different time after agent interactions.At the same time,in comparison with the killing effect on Trichomonas vaginalis conducted by Zanthoxylum bungeanum maxim and Dichroa febrifurga Lour.[Result] The killing effect by Mosla chinensis Maxim was positively correlated both with the concentration of agent and the acting time.1∶4(62.5 mg/mL),1∶4 and 1∶1(250 mg/mL) were the minimum effective concentration for MCMWE,Zanthoxylum bungeanum maxim and Dichroa febrifurga Lour to kill Trichomonas respectively.Among the three Chinese herbs,Mosla chinensis Maxim group obtained the best killing effect,Zanthoxylum bungeanum maxim group the secondary and Dichroa febrifurga Lour group the lowest,which was of statistical significance as compared with one another(P0.01).After treated with MCMWE,the cytoplasm of Trichomonas vaginalis was filled with granules and vacuoles,and some of those Trichomonas vaginalis broke and their cellular content overflew.[Conclusion] Mosla chinensis Maxim exhibits a relatively potent effect on Trichomonas vaginalis.

Abstract: [Objective] To observe the effects of paeoniflorin(PA) on contractile activity of isolated rabbit sphincter of Oddi(SO) muscle rings.[Methods] The organ-bath was used to observe the contractile activity of isolated smooth muscle from rabbit sphincter of Oddi muscle rings after cumulatively administrated different concentration of PA,the effect of accumulated PA on SO muscle rings in pre-constricted with acetylcholine(Ach) was observed.The SO muscle rings were incubated with Ca2+-free medium alone,or Ca2+-free medium plus heparin before pre-contraction with Ach.[Results] When the concentration of PA was cumulated among 12.5,25,50 and 100 μmol/L,it caused concentration-dependent relaxation while SO muscle rings were pre-constricted with Ach(10-6mol/L).This effect was not inhibited by Ca2+-free medium.However,the effect was attenuated by the co-incubation with heparin and Ca2+-free medium [without heparin:(74.56±1.47)% vs(92.31±0.84)%,with heparin:(95.98±0.52)% vs(95.03±0.43)%].[Conclusion] The results indicate that PA can relax the rabbit SO muscle rings in vitro.The mechanism may include the inhibition of intracellular calcium ions release by the 1,4,5-triposphate inositol-receptor-dependent pathway in SO muscle cells.

Abstract: [Objective] To study the mutation of ciprofloxacin selection pressure on the gyrA gene with qnrA gene positive strains and the MPC.[Methods] Transconjugants were selected by plasmid conjugation tests,which with qnrA genes and gyrA genes had no mutation.The mutation of gyrA genes and MPC under ciprofloxacin concentration selection pressure was investigated.[Results] The mutation selection exponent(MPC/MIC) of transconjugants(strains 4E and 5E) were 24.0 and 35.0,and the average mutation rate were 0.6852 and 0.5500.The MPC/MIC of negative control Escherichia coli ATCC 25922 was 6.25,and the average mutation rate was 0.2078.The negative control,compared with the transconjugants had an increased significant deviation(P0.05).[Conclusion] The gyrA genes with qnrA genes positive strains are more apt to produce mutation under ciprofloxacin concentration selection pressure,and they play an important role in the high-level quinolone resistance and high MPC.

Abstract: [Objective] To compare the value between MELD score and Child-Pugh classification in determining the short-term prognosis of patients with hepatic cirrhosis.[Methods] Retrospective data from 188 patients with liver cirrhosis were analyzed.Patients were divided into the survival group and the death group.MELD and Child-Pugh score were calculated.Prediction of mortality for three months were analyzed using the area under the receiver's operating characteristics(ROC) curve(AUC).Statistical method: t-test and Che-square test analysis.[Results] ① The mean of MELD scores was 25.57±8.55 in the death group,and was 19.09±6.33 in survival group.There was significant difference between the two groups(t=-2.190,P=0.001).② The mean score of Child-Pugh was 12.07±1.81 in the death group,and was 8.90±1.92 in the survival group.The difference between the two groups was significant(t= 9.701,P=0.000).③ AUC of MELD and Child-Pugh was 0.804 and 0.818 respectively,the difference was not significant(P 0.05).④The incidence of complications between the death group and the survival group was significantly different(P= 0.000~0.045).[Conclusion] Both MELD score and Child-Pugh classification can predict the short term prognosis of patients with decompensated liver cirrhosis.Complications were important indicators for prognosis,and should be reflected in the evaluation system.

Abstract: [Objective] This work is to evaluate the efficacy,survival and toxicity of combination of chemotherapy consisting of Rituximab and CHOP in patients with aggressive B-cell non-Hodgkin's lymphomas.[Methods] Forty patients with histopathologically proven CD20-positive aggressive B-cell non-Hodgkin's lymphomas were assigned into R-CHOP group(19 patients) and CHOP group(21 patients).CHOP regimen consisted of Cyclophosphamide,Vincristine,Epirubicin and Prednisone.The cycle was repeated every 21 days.R-CHOP regimen was Rituximab 375mg/m2 that was infused on the first day,and CHOP that followed on the second day.All of the patients received 6 chemotherapy cycles.The patients were followed up and the therapeutic effect was assessed.[Results] There were 10 CR and 7 PR in CHOP group and the overall response rate was 89.5%(17/19).There were 9 CR and 6 PR in R-CHOP group and the overall response rate was 71.4%(15/21).The response rate was significantly higher in the R-CHOP group than in the CHOP group(P0.05).The one-year survival rates were 89.5% in R-CHOP group and 71.4% in CHOP group respectively.The one-year PFS rates were 78.9% and 52.4% respectively in the two groups,with a significant difference between them(P0.05).The major side effects in the two groups were mild to moderate myelosuppression and gastrointestinal toxicity,without a significant difference between them(P0.05).[Conclusions] Combined chemotherapy including Rituximab and CHOP(R-CHOP) is recommended as the preferred treatment for aggressive B-cell non-Hodgkin's lymphoma because it is effect and well tolerated by the patients.

Abstract: [Objective] This study is to evaluate the systemic stress responses after laparoscopic operation for colorectal cancer patients.[Methods] Patients with colorectal cancer were allocated to curative laparoscopic(n=25) or conventional(n=25) operation.C-reaction protein,interleukin-6 and cortisol were examined in pre and post-operation stages to find the differences between the groups.[Results] Serum C-reaction protein,interleukin-6 and cortisol of post-operation stage in both groups were higher than those of pre-operation,but their post-operation levels in laparoscopic group were significantly less than those in conventional group(P0.05).[Conclusion] Laparoscopic resection for colorectal cancer patients brings less systemic stress responses compared with conventional group.

Abstract: [Objective] To investigate the effects of ischemic postconditioning on the ischemia-reperfusion injury in rat kidney.[Methods]Eighteen male SD rats were randomly divided into three groups as follows:(1)IR group(n=6).(2)Ischemic postconditioning group(n=6)(3)Sham group(n=6): Rats underwent the same surgical procedure without obstruction for 1 h.At the end of reperfusion for 6 h,the blood urea nitrogen(Bun),blood creatinine concentrations(Cr)were measured,the ischemic left renal tissue was collected for the determination of the superoxide dismutase(SOD) activity,malondialdehyde(MDA)content,myeloperoxidase(MPO) activity and the expression of NF-κB;the changes of renal tissue were observed by microscope.[Results] The Bun,Cr,MPO activity were significantly higher in IR and IPO group than those in Sham group(P0.05).SOD activity was lower whereas the MDA content was higher in IR group than those inSham group(P0.05) and the expression of NF-κB in IR group was significantly stronger than that in Sham group.In IPO group the Bun,Cr,MDA and MPO were significantly lower whereas SOD activity was significantly higher(P0.01) than those in IR group(P0.05),the expression of NF-κB was weaker than that in IR group.[Conclusion]Ischemic postconditioning attenuates renal ischemia-reperfusiong injury in rats.The mechanism is related to the enhancement of renal tissue antioxidation,inhibite of the neutrophilinfiltration and of NF-κB expression.

Abstract: [Objective] To study the effect of WT1 antisense oligodeoxynucleotides(ASODN) transfection on the cell proliferation and apoptosis of SKOV3 cells.[Methods] There were 4 groups in our study: normal control group,WT1 ASODN group,WT1 SODN group and Lipofectamine group.The cell apoptosis was observed by flow cytometry.The effect of WT1 ASODN on cell proliferation was assayed by MTT method.The expression level of WT1 mRNA and protein were detected by RT-PCR and Western blot.[Results] The growth of the ovarian cancer cell lines SKOV3 transfected by WT1 ASODN became significantly slow and its activity reduced,with the inhibition rate was 49.48%.WT1 antisense phosphorothioate oligonucleotides not only inhibited the proliferation,arrest cell cycle at G0-G1 checkpoint and induce the apoptosis in SKOV3 ovarian carcinoma cells but also downregulated WT1 mRNA and protein expression that contribute to the apoptosis(P0.05).[Conclusion] WT1 antisense phosphorothioate oligonucleotides could not only inhibite the proliferation but also induce the apoptosis in SKOV3 ovarin carcinoma cell line.Antisense oligonucleotides of WT1 may serve as a potential antitumor strategy in the gene therapy of ovarian carcinoma.

Abstract: VEGF is the most effective mitogen which can promote the growth of vascular endothelial cells.It plays biologic effect by combination with corresponding receptor.VEGF is less expressed in normal skin tissue,but is more expressed in some pathological courses such as skin wound,scar tissue and skin tumor.Nowadays,cellular dermal matrix(ADM) is broadly served as the dermal surrogate,although the mechanism of vascularization is unclear.VEGF plays very important role in the process of the vascularization of ADM.

Abstract: [Objective] To discuss the significance of smears of vaginal secretions in the health examinations as screening standard of gynecological routine item.[Methods] Six hundred and thirty-five female employees of the Dalian Medical University were selected.The gynecologist in the medical examination center obtained the vaginal secretions and smeared slides directly.The dry smears were dyed by professional lab assistant using multifunctional staining solution.Then,the inspections of Trichomonas vaginalis(TV),Candida albicans;vaginal leptothrix,Gardherella vaginalis(GV) and cleaning degree were carried out by light microscope.[Results] Two hundred ninety-eight cases of the Ⅲ~Ⅳ cleaning degree were checked out.Eighteen cases of monilial vaginitis and 77 cases of GV were confirmed.None of TV was detected.Four cases had combined infection.Compared the two groups of Ⅰ~Ⅱ and Ⅲ~Ⅳ cleaning degree,the latter had more monilial vaginitis than the former obviously(P0.005).Moreover,the latter had more clue cells(P0.05) and mixed infection(P0.05) than the former.If two groups of 25~50 year-old and 51~89 year-old were compared,the former was more in Ⅲ~Ⅳ cleaning degree than the latter(P0.005) and more in monilial vaginitis and clue cells than the latter(P0.05).[Conclusion] The smears of vaginal secretions as gynecological routine item of health examination are very important to locate the diseases in time and for early treatment.It is very necessary for adult female to take periodic health examination.
