Abstract: Pelodiscus sinensis Japanese population and Qingxiwubie are the two new varieties of soft-shell turtle.It is very importance to master the genetic relationship in Pelodiscus sinensis for further breeding and cross breeding,the genetic diversity in the new varieties of Pelodiscus sinensis were studied using random amplified polymorphic DNA(RAPD) technique to compare with the usual populations of Taihu and Taiwan.A total of 162 bands,including 110 polymorphic ones were detected from the four populations by using 23 random primers.The results showed that the level of genetic diversity was high in Japanese population,and the average genetic similarity index within each population ranked as Japanese population(0.846 9)Taiwan population(0.842 9)Qingxiwubie population(0.831 7)Taihu population(0.825 4).The maximal genetic distance occurred between Taihu and Qingxiwubie populations,while the minimum occurred between Taiwan and Qingxiwubie populations.The total genetic diversity of Pelodiscus sinensis was 0.236 8 and the intra-population genetic diversity was 0.169 9,which suggest that 71.75% of the genetic variation existed within the populations.The genetic differentiation coefficient was 0.282 7,indicating that there was a certain level of population differentiation.With the methods of UPGMA and NJ on the basis of genetic distances,the results showed that the four populations were clustered into two groups: one with Taihu and Japanese populations,and the other with Qingxiwubie and Taiwan populations.Further appropriate measurements must be taken to protect the germplasm of Pelodiscus sinensis from the reduction of genetic diversity.

Abstract: In order to investigate the effect of some drugs on virulent bacteria isolated from Chinese soft-shelled turtle,the bacteria was isolated from a died Chinese soft-shelled turtle from one Jinxian farm in Jiangxi.The morphological,culture,biochemical characteristics of the isolate were analyzed,and virulent factors detection,virulent test on the mice and challenge test were carried out,too.The effect of 9 drugs on the isolate was analyzed according to MIC of the drugs to the bacteria strain by the method of K-B drug susceptibility and double dilution on the 96 micro-plate.The results showed that the pathogenic Aeromonas hydrophila infection resulted in the Chinese soft-shelled turtle's death and susceptibility test results showed that the strain were highly sensitive to cefixime,cefamandole,aztreonam,kanamycin,netilmicin,Chloramphenicol,levofloxacin;the minimum inhibitory concentration(MIC) of 9 antimicrobial drugs against the bacteria showed that the isolate were highly sensitive to the five cephalosporins,gatifloxacin and oxolinic acid,moderately sensitive to Fosfomycin and flumequine,and cephalosporins was superior to quinolones,ceftizoxime was the better antimicrobial drug than the others.

Abstract: Rex rabbits were selected to analyze the changes of serum malondialdehyde(MDA),total superoxide dismutase(T-SOD) and total antioxidant capacity(T-AOC) by using colorimetric method during heat stress with different intensity(25℃,30 ℃,35 ℃ and 40 ℃)and different time(0.5,1.5,2,2.5,4 and 6 hour).The results showed as follows: Compared with the control group(25 ℃),the MDA contents in serum decreased slightly(P0.05) after heat stress with 30 ℃ and it was significantly lower at 35 ℃ and 40 ℃(P0.01).The SOD activity in serum of Rex Rabbit was in rising trend and T-AOC was in declining trend.The SOD activity and T-AOC-size in every heat stress group had very significant differences comparing with the control group.During heat stress with different time,the MDA contents in serum raised and reached the peak with 4 hour(P0.01)and then decreased.There were no differences between heat stress with 6 hour and 0.5 hour.The SOD activity and T-AOC-size in serum of Rex rabbit were in fluctuation.Compared with heat stress 0.5 hour,the SOD activity and T-AOC-size in every heat stress group were very significantly increased(P0.01).The T-AOC-size in serum of Rex rabbit was significantly increased after heat stress with 1 hour(P0.05)and very significantly decreased after heat stress with 1.5 hour,2 hour(P0.01).There were not differences between other groups for T-AOC-contents in serum of Rex Rabbit.

Abstract: The chytridiomycosis is a significant emerging disease of amphibians in recent years which seriuosly does harm to the wild and breeding population.To ascertain the condition of the Batrachochytrium dendrobatidis infection history in the population of toad,the medicinal economic animal,and enhance the prevention for this disease,32 toad specimen collected from Sichwan keeping in a museum were assayed by Taqman-MGB fluorescence probe quantitative polymerase chain reaction to detect Batrachochytrium dendrobatidis.Besides,the products of QPCR were cloned and sequenced to identify the origin of the pathogen by sequence alignment and phylogenetic analysis.Finally we got the standard curve:Y=-3.0X+32.39 and the related coefficient:R2=0.999 6. Total 12 positive samples reported the detection rate as 37.5%.Meanwhile the phylogenetic analysis indicated that a certain extent differentiation of the chytrid fungi in our country exists.One kind of the tested strain showed highly genetic relationship with the strains from the North America,South America and Europe.Another one is obviously different from the strains found in the other areas of the world with special characteristics.It suggest that the prevention and treatment for chytridiomycosis of toad should be more targeted and should not copy foreign experience aimlessly.

Abstract: The Blue Sheep are good at climbing and jumping,the variations of Blue Sheep′s gait motion are the bases of the bionic quadruped robots′ mechanism studying.The research set out from bionics,regarding Blue Sheep as the research object,establishing three-dimensional gait experimental system of Blue Sheep,and had gained the acquisition of sportive video through VFW,processed the image by professional image processing software Sigma Scan,eventually obtained the location of key points of the Blue Sheep limbs and the acceleration curve.Through the experiment to obtain the complete information of blue sheep gait,its analysis could be used for design and control of bionic blue sheep robot.
