Abstract: Que peut nous apprendre un ver sur le fonctionnement du cerveau humain? Les donnees acquises sur le systeme nerveux du nematode Caenorhabditis elegans demontrent l’existence d’une fascinante conservation de la biologie moleculaire et cellulaire du neurone au travers de plus de 550 millions d’annees d’evolution separant les nematodes des mammiferes. C. elegans possede un systeme nerveux simple forme de 302 neurones et d’environ 7000 synapses. Des outils genetiques puissants permettent d’isoler de nouveaux genes et d’attribuer a des genes connus de nouvelles fonctions dans la mise en place et le fonctionnement du reseau neuronal du nematode. Nous montrerons par quelques exemples comment les decouvertes faites chez C. elegans ont pu etre rapidement transposees a la biologie du systeme nerveux des mammiferes.

Abstract: Myeloproliferative disorders are clonal haematopoietic stem cell malignancies characterized by independency or hypersensitivity of haematopoietic progenitors to numerous cytokines. The molecular basis of most myeloproliferative disorders is unknown. On the basis of the model of chronic myeloid leukaemia, it is expected that a constitutive tyrosine kinase activity could be at the origin of these diseases. Polycythaemia vera is an acquired myeloproliferative disorder, characterized by the presence of polycythaemia diversely associated with thrombocytosis, leukocytosis and splenomegaly. Polycythaemia vera progenitors are hypersensitive to erythropoietin and other cytokines. Here, we describe a clonal and recurrent mutation in the JH2 pseudo-kinase domain of the Janus kinase 2 (JAK2) gene in most (> 80%) polycythaemia vera patients. The mutation, a valine-to-phenylalanine substitution at amino acid position 617, leads to constitutive tyrosine phosphorylation activity that promotes cytokine hypersensitivity and induces erythrocytosis in a mouse model. As this mutation is also found in other myeloproliferative disorders, this unique mutation will permit a new molecular classification of these disorders and novel therapeutical approaches.

Abstract: Pour se construire une identite coherente, l’homme s’est toujours attache a distinguer ce qui appartenait au monde perceptif, ce qui lui appartenait en propre et ce qui appartenait au monde interieur d’autrui. Les psychanalystes ont particulierement etudie les structures psychiques fantasmatiques delimitant le monde interieur individuel : leur constitution, leurs caracteristiques, leur devenir, leurs perturbations et leurs defaillances. Dans le meme mouvement, a la suite des ethologues, ils ont pointe l’importance du role de la mere et des echanges precoces mere-enfant (tactiles, en particulier) pour que l’enfant devienne un individu autonome se sentant en securite dans ses limites corporelles et psychiques. S’est ainsi developpe le lien metaphorique entre la peau reelle, celle etudiee par les anatomistes et les dermatologues, qui limite le corps et se situe a la frontiere du monde exterieur et du monde interieur de l’individu, et une structure psychique fantasmatique qui joue le role de limite, de frontiere entre le monde interieur, psychique, de l’individu et le monde exterieur. C’est dans une telle dynamique d’elaboration theorique psychanalytique que Didier Anzieu, psychanalyste francais, a elabore le concept du Moi-peau. Il a ainsi favorise des collaborations theoriques et pratiques fructueuses entre les psychanalystes et les dermatologues.

Abstract: Inflammatory bowel diseases (IBD) are idiopathic, chronic and relapsing inflammatory conditions of the gastrointestinal tract. New insights into the pathogenesis of IBD have been provided by three lines of research: (1) studying susceptibility genes involved in the detection of bacterial components and in the regulation of the host immune response, (2) highlighting the disruption of tolerance towards the commensal microbiota and (3) unravelling the critical role of environmental factors such as sanitation and hygiene. This review presents current etiological hypothesis of IBD which argue that pathogenic intestinal bacteria and/or infectious agents initiate and perpetuate the inflammation of the gut in an individual with genetic vulnerability leading to impaired epithelial barrier function and abnormal mucosal immune responses.
