Abstract: Objective To study the variation of apolipoprotein(Apo) A5 influence un-related Shandong han patients with coronary artery disease(CAD) and lipid disorder.Methods The study was investigated by case-control method. Polymorphism of ApoA5 SNP3 was identified by polymerase chain reaction-restriction fragment length polymorphism(PCRRFLP) and DNA sequence.Results The frequency of the ApoA5 SNP3 TC in CAD patients was significantly higher than that of the control group,and this difference existed after adjustment for age,gender,levels of lipid.In CAD patients,the levels of triglyceride(TG) were significantly elevated in ApoA5 SNP3 TC compared to two other genotype.Conclusions ApoA5 SNP3 maybe a genetic marker for CAD;the variation of ApoA5 SNP3 influence plasma TG.

Abstract: Objective To investigate the effect of ecdysterone on protein expression of insulin receptor(InsR) in insulin-resistant HepG2 cells. Methods After establishment of the insulin resistant HepG2 cell model, ecdysterone was added in culture fluid. The influence of ecdysterone and pioglitazone on glucose uptake rate was observed, as well as the influence of ecdysterone on the InsR protein express in IR HepG2 cell. Results Compared with IR model cells,1×10-5 mol/L ecdysterone incresed InsR protein express of IR HepG2 cell obviously. Conclusion IR sensitization of ecdysterone may have relation with IR signal transduction molecule such as IR protein strong express.

Abstract: Objective To observe the antitumor effect of sesamin on H22 hepatoma-bearing mice and explore its mechanism.Methods 64 mice embedded with H22 hepatoma were equally divided into 4 groups: model group(saline),cyclophosphamide group(0.2 g/L),high concentration sesamin group(15 g/L),low concentration sesamin group(1.5 g/L).Every group received corresponding treatments for 10 days.The inhibition rate of tumor,thymus,spleen index and expression intensity of proliferating cell nuclear antigen in the tumor tissue of the 4 groups were compared.Results Low concentration sesamin could significantly inhibit growth of H22 hepatocarcinoma cells in vivo,promot the immunity function and reduced the expression of PCNA in the tumors.Conclusion The inhibitory mechanism of sesamin on H22 hepatocarcinoma cells may be realized by promoting the immunity function and inhibiting the expression of PCNA.

Abstract: Objective To inves tigate the effect poly cyclic aromatic hydro carbons (PAH)s in stomach carcinoma genesis.Method PAHs in human gastric tumor and normal stomach were detected with solid phase extraction-high performance liquid chromatography (SPE-HPLC). Result Pyrene and B(a)P in human gastric tumor were significantly higher than those in normal stomach(P0.05), while there were non-significant difference with dimethylnaphthalene and phenanthrene in human gastric tumor and normal stomachs(P0.05). Conclusion Human gastric tissue has an extremely strong ability of bioconcentrating PAHs, which play an important role in the occurrence of human gastric tumor.

Abstract: Objective To investigate the effects of tongxinluo capsule on platelet aggregation rate and C-reactive protein(CRP) and thromboxan B2(TXB2) in patients with acute coronary syndrom(ACS)who had aspirin resistance(AR).Methods A total of 70 ACS patients who had AR were recruited.They were randomly assigned into conventional therapy group,tongxinluo group,clopidogrel group.platelet aggregation rate and CRP and TXB2 levels were detected before and after 1-month treatment.Results Tongxinluo and clopidogrel could both make platelet aggregation rate,CRP and TXB2 decrease in ACS with AR patients.The levels of platelet aggregation rate,CRP and TXB2 had statistical significance before and after treatment(P0.05,0.01).Conclusions Tongxinluo capsule has antiplatelet aggregation and anti-inflammatory effect in ACS patients with AR,and can improve AR in ACS patients in a certain degree.

Abstract: Objective To observe the expressional variability and correlation of Ezrin and CD44V6 between osteosarcoma and osteochondroma and explore their clinical significance.Methods Ezrin and CD44V6 were detected by SP immunohistochemical method in 36 cases of osteosarcoma and 20 of osteochondroma.Results The positive rate of Ezrin and CD44V6 expression were 55.56% and 72.22% in osteosarcoma,which were higher than those in osteochondroma,P0.01.There was a positive correlation between Ezrin and CD44V6(rs=0.444,P0.05).The positive expression of Ezrin in osteosarcoma was relative with the rate of 3-year metastasis-free.Conclusions Ezrin and CD44V6 may participate the progressions of infiltration and metastasis.The detection of them can early predicte potential metastatic risk and prognosis of osteosarcoma.

Abstract: Objective To investigate the single nucleotide polymorphism of visfatin promoter-3186 CT and its relationship with type 2 diabetes.Methods The single nucleotide polymorphism of visfatin promoter-3186 CT of 151 type 2 diabetes patients and 69 healthy control subjects were determined by ligase detection reaction(LDR) and microarray assay.Results The total frequencies of allele of CC\TT\CT and A\G of promoter-3186 CT in visfatin were 20.45%,50.0%,29.55% and 45.45%,54.55% respectively.No significant differences were found between type 2 diabetes group and the control group(P0.05).There were no significant differences between obese diabetes and nonobese diabetes(P0.05).The BMI average of allele of CC were higher than TT(F=4.939 P=0.028).Conclusions The single nucleotide polymorphism of visfatin promoter-3186 CT is found in the han population of this area and there is certain relationship between its genovariaton and BMI of T2DM patients.

Abstract: Objective To investigate expression and clinical significance of CXCR4 and hTERT in normal brain tissue and all grades of glioma.Methods Immunohistochemical staining and ISH for CXCR4 and hTERT were perfomed in 60 cases of glioma and 6 cases of normal brain tissue.Results In 60 cases of glioma,35 cases expressed CXCR4,36 cases showed hTERT,the positive rates were 58.3%,60.0% respectively.The expression level of CXCR4 and hTERT strengthened with the rise of tumor grade(P0.05).CXCR4 and hTERT protein was not found in normal brain tissue.CXCR4 is correlated with expression of hTERT(r=0.6358,P0.05).Conclusions Expression of CXCR4 and hTERT is related to grading of glioma.The united ditecting of CXCR4 and hTERT can be used to accurately assess the malignancy of glioma.

Abstract: Objective To explore the relations among the polymorphism of CYP1A1 gene and GSTM1 gene,environmental exposure and the susceptibility to lung cancer.Methods The polymorphisms of the CYP1A1 gene and GSTM1 gene were detected by the method of polymerase chain reaction and restriction fragment length polyorphism.Results Compared with control group,the frequencies of smoking and dust exposure were higher,While the frequencies of eating vegetables,fruits and drinking sanitized water were lower in lung cancer(P all 0.01).There was no statistical difference in the distribution of each genotype of CYP1A1 and GSTM1 in the two groups.There was a synergistic interaction to lung cancer among CYP1A1 mutant genotype,GSTM1 null genotype and tobacco exposure.Conclusions Smoking and dust exposure significantly increase the risk of lung cancer,intaking vegetables,fruits and sanitized water frequently decrease the risk of lung cancer.there is an elevated risk of lung cancer in CYP1A1 mutant genotype with exposure to tobacco,as well as GSTM1 null genotype.

Abstract: Objective To study the relationship between CpG island hypermethylation of MT3 gene and esophageal squamous cell carcinoma.Methods Four cell lines of squamous cell carcinoma of the esophagus TE-1,TE-13,TTN,ECA-109 were employed in this study.The comparison of CpG island hypermethylation status and the mRNA expression of MT3 gene of these four cell lines before and after treated by 5-Aza-2'-deoxycytidine(5-aza-CdR) was performed by using the techniques of methylation-specific PCR(MSP) and RT-PCR.Results Different degree of CpG island hypermethylation of MT3 gene existed in all the four cell lines of esophageal squamous cell carcinoma.The hypermethylation was demethylated after the treatment of 5-aza-CdR.CpG island hypermethylation mode was removed,the mRNA expression was also enhanced obviously(P0.05).Conclusions CpG island hypermethylation of MT3 gene in cell lines of esophageal squamous cell carcinoma can inhibit their mRNA expressions.When the hypermethylation is demethylated,the mRNA expression will be enhanced correspondly.

Abstract: Objective To study the diagnosis and classification indexes of newly diagnosed type 1 diabetes mellitus(T1DM) and type 2 diabetes mellitus(T2DM) in children.Methods Diabetes family history,the age of onset,the time of appearing symptom to established diagnosis,DKA incidence,acanthosis nigricans,waist circumstance /high ratio(WC/H),body mass index(BMI),HbA1C,FBG,FCP and FINS,IRI,islets cell autoimmune antibodies(ICA),glutamic acid decarboxylase(GAD) antibodies(GADA) and insulin dependence were compared between 110 newly diagnosed T1DM and 20 newly diagnosed T2DM in children.And related indexes were compared with that in healthy children.Results The TIDM characterized urgent incidence,small age of onset,high DKA incidence,less DM history,low BMI and WC/H,no acanthosis nigricans,high positive rate of ICA and(or) GADA,low FCP and FINS,no insulin resisitance and insulin dependence.The T2DM characterized high DM history,the age of onset 10 a,slight symptom of polyuria,polydipsia,polyphagia and weight loss,obvious acanthosis nigricans,obesity,lower positive rate of diabetes immunoantibodes,insulin resistance and no insulin dependence.Conclusions Acanthosis nigricans,obesity,insulin independence are characteristic indexes of newly onset T2DM.Decreased FCP and FINS,no insulin resistance and insulin dependence are characteristic indexes of newly diagnosed T1DM.Family history,the age of onset,DKA and GADA are important reference indexes.

Abstract: Objective To study the influence of TGF-β1 on proliferation and differentiation of HPDLFs in vitro.Methods HPDLFs were primarily cultured.The effect of TGF-β1 on the proliferative ability of HPDLFs was evaluated with MTT assay.The effects of TGF-β1 on the total amount of protein and the cell cycle were detected by coomassie brilliant blue(CBB) method and flow cytometry.The effects of TGF-β1 on the alkaline phosphatase(ALP) activity and expression of ALP mRNA were measured with the enzyme kinetic method and reverse-transcription polymerase chain reaction(RT-PCR).Results TGF-β1(2~ 10 μg/L)significantly promoted the proliferative ability of HPDLFs and the changes depended on dosage and time(P0.05).The total amount of protein was dramatically increased and the ALP activity was enhanced for 48 h with TGF-β1(2~ 10 μg/L),P0.05.After 48 h of TGF-β1(10 μg/L),the G1 phase cells reduced(P0.05),the S phase cells and proliferation index(PI) significanctly increased(P0.05).The expression of ALP mRNA was evidently up-regulated by different concentrations of TGF-β1(2~10 μg/L)after 48 h(P0.05).Conclusions These findings suggest that TGF-β1 was capable of producing an accelerative effect on the proliferation and differentiation functions on HPDLFs.It may be related to TGF-β1 enhancing cell DNA,protein synthesis,ALP mRNA expression and ALP activity.

Abstract: Objective To investigate the relation between the polymorphisms of β2 adrenergic receptor(β2-AR) in 16,27 position and chronic obstructive pulmonary disease(COPD).Methods The genetic mutation types were determined by sequencing in 49 COPD patients(COPD group)and 48 healthy controls(control group),and FEV1% value in COPD group was detected.Results There was no difference in gene frequency and allele freauency of β2-AR coding region 16,27 site between COPD group and control group(P0.05);β2-AR coding region 16 site polymorphism had significant relation with FEV1% value(P0.01),FEV1% values of patients with Gly/Gly and Arg/Gly genotypes were lower than patients with Arg/Arg genotype. The polymorphism of β2-AR coding region 27 site had obvious relations with FEV1% value(P0.05),FEV1% values of the patients with Gln/Gln and Gln/Glu genotype were lower than those of patients with Glu/Glu genotype.Conclusions Gene polymorphism of β2-AR coding region 16,27 site is not possible a primary cause of COPD,but it can influence pulmonary function of patients with COPD.

Abstract: Objective To investigate chemokine receptor CX3CR1 gene V249I polymorphisms and its frequency in patients with ischemic cerebrovascular disease(ICVD).Methods The CX3CR1 gene V249I polymorphisms and frequency of patients with ICVD(cerebral infarction,lacunar infarction,transient ischemic attack) and healthy controls were analyzed by multiplex polymerase chain reaction and restriction fragment length polymorphism.Results There were VV and VI genetypes of CX3CR1 gene V249I in control group,and VV,VI and II genetypes in ICVD group.The frequency of I249 allele was significantly higher in ICVD group than that in control group(P0.01).No significant differences were observed in ICVD subgroups in genetypes and I allele distribution.Conclusions CX3CR1 gene V249I polymorphism may be associated with ICVD.

Abstract: Objective To observe the effect of pure puerarin and pueraria crude extract on H446 cell proliferation, and explore its mechanism. Methods H446 cells grew in different concentrations of SP and CP, cell growth inhibition was detected by MTT assay.Cell cycle and apoptosis were detected by flow cytometry.Bcl-2 and Bax protein were detected by immunohistochemistry. Results The CP for 72 h 50% inhibitory concentration (IC50) was 435 μg/ml, for SP was 1 403 μg/ml、 435 μg/ml on H446 for 72 h,the apoptosis rate induced by CP and SP were respectively 14.71% and 2.61%, the percentages of G0/G1 phase cell numbers were respectively 79.20% and 72.20%, those of G2/M phase were respectively 8.94% and 10.50%, there was significant difference compared with control group(P0.05). Compared with the control group the Bcl-2 protein expression was lower, Bax expression is increased, P0.05. The expression proportion of Bax/Bcl-2 was higher than control group. Conclusions SP and CP inhibit H446 tumor cell proliferation. SP and CP can induce apoptosis, which may be related to block the cell cycle in the G0/G1 phase, up Bax Expression, lower Bcl-2 expression.

Abstract: Objective To observe the effect of p38 mitogen activated protein kinase inhibitor SB203580 on the progress of chondrocyte apoptosis.Methods 40 SD rats were randomly divided into A,B,C and D groups.Groups A,B and C underwent unilater ACLT.Rats of group A received 0.1 ml intra-articular injection of p38MAPK inhibitor at concentration of 100 μm/L after surgery,while those in group B received 0.1 ml 0.9% sodium chloride.Rats of group C received no treatments and those of group D were mormal.All of the rats were sacrificed at 8th week after surgery.Histological test and apoptosis index of each group were observed.Results Histological changes observed in group A were slighter than those in group B and C.Apoptotic chondrocytes were observed in all groups.Apoptosis index was significantly different between group D and other three groups,and the apoptosis index of group A was lower than that of group B and C.Conclusions Chondrocytes apoptosis was increased in rats with OA induced by ACLT,and intra-articular injection of p38MAPK inhibitor can suppress the chonsrocyte apoptosis effectively.

Abstract: Objective To investigate the expression of COX-2,EGFR and Ki67 in colorectal cancer and its significance.Methods To select 70 samples of colorectal cancer,21 samples of colorectal adenomas and 15 samples of normal colorectal mucosal tissues,the expressions of COX-2,EGFR and Ki67 were evaluated by immunohistochemical method of Envision.Results In normal colorectal mucosal tissues,colorectal adenomas and colorectal cancer,the positive rates of COX-2 expression were 33.3%,71.4% and 80.0%,respectively.In normal colorectal mucosal tissues,colorectal adenomas and colorectal cancer,the positive expression rates of EGFR were 0,23.8% and 65.7% respectively,there were statistical differences,when they compared with each other(P0.05).The positive expressions of Ki67 were 40.0%,76.2% and 82.8%,Ki67 expressions in colorectal cancer group and adenomas group were significantly higher than that of the normal group(P0.05).In colorectal cancer,the expression of COX-2 in the group with lymph nodes metastasis was significantly higher than the group with no lymph nodes metastasis(P0.05).The expression of EGFR in the group of poorly differentiation,high stage and lymph nodes metastasis was significantly higher than the group of the well differentiation,low stage and no lymph nodes metastasis in colorectal cancer(P0.05).Furthermore,the studies showed that there were significant correlation between the expression of COX-2 and EGFR,COX-2 and Ki67 as well as EGFR and Ki67(r=0.316,0.435,0.314,P all 0.05).Conclusions COX-2,EGFR and Ki67play an important role in the carcinogenesis and development of colorectal cancer.

Abstract: Objective To observe the association of plasmahomocysteine(Hcy) and polymorphism in 5,10 methylene tetrahydrofolate reductase(MTHFR) genes with cerebral infarction in the elderly.Methods 102 elderly patients with cerebral infarction and 100 controls were studied,The Hcy levels were measured by highperformance liquid chromatography fluorescence detection.The polymorphism in MTHFR was determined by a polymerase chain reaction(PCR) assay and subsequent restriction enzyme digestion.Results The genotype distribution of MTHFR R677 site and the frequency of alleles were significant difference in cerebral infarction group and control group(P all0.05).The frequency of T alleles and the fast Hcy levels were higher in the cerebral infarction group compared with those in the control group(P all 0.05).Conclusions The MTHFR gene mutations can lead to hyperhomocysteinemia increase in the elderly patients with cerebral infarction.The mutations in MTHFR and hyperhomocysteinemia are risk factors for cerebral infarction in elderly.

Abstract: Objective To investigate the effects of Aucubin on blood glucose levels and mitochondria antioxidant capacity in streptozotocin-induced diabetic rats. Methods Diabetic rats were induced by intraperitoneal injection of STZ. Rats were randomly allocated into diabetic group, low dose group,high dose group and normal control group. During and after the treatment, the general state, body weight, blood gloucose, lipid per-oxidative level and antioxidative enzymes activities in mitochondria were observed.Results The level of blood glucose and MDA in the diabetic group significantly increased, and the activities of CAT, GSH-Px and SOD vigor reduced significantly. After the administration of Aucubin, the level of blood glucose and MDA in the diabetic group reduced significantly, as well as the activities of CAT, GSH-Px and SOD increased significantly. Conclusion This study showed that Aucubin administration could reduce the level of blood glucose, improve mitochondria antioxidant capacity in diabetic rats. Thus, Aucubin may be implicated as a preventive medicine against diabetes mellitus.

Abstract: Objective To investigate the inhibitory action of astemizole on wild type(WT) and Y652 mutational HERG channels and observe the molecular mechanism.Methods HERG channels were expressed in Xenopus oocytes,and currents were measured using two-microelectrode voltage-clamp technique.The effects of astemizole on HERG channels blockade were observed.Results Astemizole blocked HERG current in voltage-concentration-and time-dependent manner.Compared with the WT,Y652A and Y652R significantly attenuated the blockade by astemizole.Conclusion Astemizole preferentially blocks open state HERG channel.Compared with the WT-652 is a critical residue in the astemizole-binding site.

Abstract: Objective To study the expression of Livin,survivin and IGF-2 in human hepatocellular carcinoma(HCC) and its clinical significance.Methods Expression of Livin,survivin and IGF-2 in 60 HCC patients were assessed by immunohistochemistry method.Twelve candidate factors associated with prognosis were analyzed by the COX proportional hazards model and Log-rank test.Kaplan-Meier was used to estimate the survival rate.Results The positive rate of Livin,survivin and IGF-2 expression were 32/60(53.3%),39/60(65.0%),31/60(51.7%) respectively.The expression rate of Livin and survivin in HCC tissue was higher than that in the adjacent non-tumorous liver tissue and the normal liver tissue.There was no significant correlation between the expression of Livin and survivin in HCC and the tumor size,capsule status,cirrhosis,the classification of Child-Paugh,TNM stage,except the portal vein tumor thrombus.There was no significant correlation between the expression of IGF-2 and capsule status,the classification of Child-Paugh and cirrhosis.However,the positive rate of IGF-2 was correlated significantly with portal vein tumor thromb,liver cirrhosis and TNM stage.There was a close correlation between the expression of Livin,survivin and IGF-2.The multivariae analysis showed that Livin,survivin and IGF-2 were independent significant prognostic factors of HCC.Conclusion The upexpression of Livin,survivin and IGF-2 has a close correlation and holding a prognostic significance in HCC.

Abstract: Objective To observe the morphological changes of the apoptosis and degeneration of A549 human lung cancer cells in vitro after treatment with the raw extract of Grub.Methods The growth inhibition rates and cytotoxic effects of A549 cells were measured with MTT assay.Morphology changes were observed by microscope,HE and scan electron microscope.Results The Raw Grub extract could significantly inhibit the growth and proliferation of A549 cells in dose-dependent and time-dependent manner.Inverted microscope revealed that the A549 cells showed the apoptosis changes after treatedment with 80 μg/ml of the Grub petroleum ether extract before 24 h and necrosised;after 72 h.Conclusion The Grub extract has an influence on apoptosis induction and prdiferation inhibition of A549 human lung cancer cells in vitro.

Abstract: Objective To study the relationship between helicobacter pylori(Hp) infection and cerebral infarction as well as transient ischemic attack.Methods By means of enzyme linked immunosorbent assay(ELISA),serum density of helicobacter pylori antibody Ig-G(Hp-IgG) and cytotoxin associated gene A IgG(CagA-Hp-IgG) in 80 cases of cerebral infarction patients and 80 cases of transient ischemic attack(TIA) patients as well as 60 control group were measured.At the same time,serum concentrations of total cholesterol,triglyceride,apolipo protein A,apolipo protein B,high density lipoprotein-cholesterol,low density lipo protein-cholesterol were measured.Results In cerebral infarction group and TIA group,the positive rate of serum Hp-IgG was 65.40% and 67.90%,CagA-Hp-IgG was 52.56% and 50.23%,and they were significantly higher than those of controls(47.15% and 35.20%).Conclusions Hp infection is closely associated with transient ischemic attack and cerebral infarction.It can also influence the level of blood fat.

Abstract: Objective To investigate the role of KISS-1 and MMP7 in the genesis and development of NSCLC.Methods The expression of KISS-1 and MMP7 was detected in 58 NSCLC tissues and 23 normal lung tissues as control by immunohistochemical method SABC.Results The expression positive rate of KISS-1 protein in the tissue of NSCLC was significantly lower than that of the control(P0.05),the positive rate of MMP7 protein was significantly higher than that of the control(P0.05).The expression level of KISS-1 and MMP7 protein were closely related to lymph node metastasis,clinical stages(P0.05),but not to differentiations,histological classification,smoke,age,and sex(P0.05).Conclusion Abnormal expression of KISS-1 and MMP7 may play an important role in the growth and metastasis in human NSCLC.Detection of KISS-1 and MMP7 expression may be helpful to predict the prognosis of patients with NSCLC.

Abstract: Objective To study the expression of Livin and p53 in non-small cell lung cancer(NSCLC),explore the relationship between Livin and p53 expression.Method Livin mRNA and protein were detected in lung tissues of 48 patients with NSCLC and 12 patients with benign lung diseases by RT-PCR and immunohistochemical staining,respectively. p53 protein were detected by Immunohistochemical staining. Result The positive rates of Livin and p53 in NSCLC group were significantly higher than those in normal control and benign lung diseases. The positive rate of Livin in lung adenocarcinoma reached 46.2 % ,much more than that of lung squamocellular carcinoma and large cell carcinoma(P0.01) . Positive correlation was found between p53 and livin protein(r=0.457,P0.01).Conclusions Increased expression of Livin protein can be served as characteristic parameters for NSCLC. As molecular marker of lung adenocarcinoma ,Livin may be served as a new target for apoptosis inducing therapy of NSCLC.
