Abstract: Objective To establish human endometrial adenocarcinoma cell line.Methods Tumor tissues from 1 case with well differentiated endometrial adenocarcinoma were made into single-cell suspension and cultured in DMED/F-12 medium supplemented with 10% fetal bovine serum.The morphology of the cells was observed by inverted microscope.The immunofluorescent staining technique was used to detect cytokeratin in endometrial carcinoma cells.Results The cells were subcultured more than 50 generations,monolayer cultured cells were polygonal in shape and had a tendency to pile up without contact inhibition.The expression of cytokeratin was positive in the cells.Conclusion The novel endometrial adenocarcinoma cell line is successfully established.

Abstract: In this study, a simple and high-resolution four-degrees-of-freedom (4-DOF) motion error measuring system based on DVD pickup has been proposed. it can simultaneously measure the horizontal straightness error and three angular errors of a single-axis linear moving platform. The horizontal straightness error is measured based on the focus technique of DVD pickup and the inverse algorithm is used to solve pitch, yaw, and roll angular errors through the kinematic analysis. The grating is mounted on the moving axis of a single-axis linear moving platform and reflects incident laser beam into several diffractive rays. Photodiode IC (PDIC) of DVD pickup is set up for detecting the focus error signal (FES) of zero-order diffraction ray from grating and quadrant detectors are set up for detecting the position of ±1st-order diffraction rays from grating. The 4-DOF motion error measuring system detected the three angular errors with a related accuracy of about ±1 arc sec for a measuring range of ±100 arc sec, and detected horizontal straightness with a related accuracy of about ±2 μm for a measuring range of ±150 μm.

Abstract: Objective To investigate the different preparations of radix isatidis on the nucleoprotein(NP) of influenza virus,so as to look for the active ingredient of anti-influenza A virus herbs.Methods The radix isatidis decoction and isatis agglutinin were separately decocted and concentrated by boiled leaching and organic solvent immersion method,and radix isatidis injection was purchased,the test concentrations of which were 1.95,2.60,0.78 mg/ml detected by means of cytopathic effects(CPE).The cultured Hela cells were divided into 8 groups,the plasmid group was transfected with pcDNA3.1(+)/NP,the empty plasmid group was transfected with pcDNA3.1(+);after transfected with pcDNA3.1(+)/NP,the experimental groups of injection,decoctionand,agglutinin were treated with the three preparations of radix isatidis at above test concentrations,the control groups corresponding the three preparations were not transfected and treated with the preparations only.Indirect immunofluorescence and colloidal gold immunochromatographic strip assay were used for determining the expression of NP gene.Results In the plasmid group and the decoction group and injection group of the radix isatidis,colloidal gold immunochromatography showed positive results,and strong green fluorescent was appeared in transfected Hela cells,and the NP were expressed in cytoplasm;while in the agglutinin group and the empty plasmid group,no strong green fluorescent were found in the cells expressed NP and,colloidal gold immunochromatography showed negative results.Conclusion Radix isatis agglutinin can inhibit the expression of NP gene,which provide new thoughts for the research of drugs anti-influenza virus.

Abstract: Objective To investigate the relationship between adiponectin gene exon 2 polymorphism and type 2 diabetes mellitus(T2DM) in Han population of Binzhou.Methods A total of 100 T2DM patients and 100 controls were enrolled in our study.PCR-RFLP method was used to determine the polymorphism of the SNP+45(T/G) in exon 2 of adiponectin gene.Results G/G genotype frequency and G allele frequency were significantly higher in T2DM group than in the control group(P0.01).T allele frequency was significantly lower in T2DM group than in the control group(P0.01).Conclusion Adiponectin gene may play a major role in susceptibility to T2DM in Han population of Binzhou.

Abstract: Objective To observe the effect of tissue factor(TF)on invasion inhibition and apoptosis inducing effect of oxaliplatin in human gastric cancer cell line SGC7901.Methods SGC7901 cells wich has been successfully transfected with TF-pcDNA3 recombinant(Experimental group)and empty vector pcDNA3(Control group) were incubated with oxaliplatin.Transwell chamber were used to show the change of invasive ability of cells.Caspase-3 activity were detected by colorimetric method;Annexin-V/PI double-staining method was used to detect the apoptosis rate.Results Compared with the control group,invasion ability increased but Caspase-3 activity and apoptosis rate decreased in the experimental group,P0.05.Conclusion TF can reduce the invasion inhibition and apoptosis inducing effect of oxaliplatin in human gastric cancer cell in vitro.

Abstract: Objective To investigate the association of single nucleotide polymorphisms(SNP)45 T→G and SNP276 G→T in the adiponectin gene with metabolic syndrome(MS).Methods 183 patients with MS and 144 participants were selected.Adiponectin gene SNP45 of them were analyzed by TaqMan technology.Adiponectin gene SNP276 of them were analysised by direct sequencing.Results The distributions of SNP45 genotypes were different between MS and the control group(P0.05).The analysis of allele frequency showed the major allele was G in MS patients(P0.05).The subjects with SNP45 TG or GG genotype had lower serum adiponectin levels than those with TT genotype in MS groups(P0.05).The distribution of genotype and allele frequencies of SNP276 were not different in MS and control groups(P0.05).Conclusion SNP45 G/G genotype seems to be the susceptive gene in MS patients of Chinese Han.

Abstract: Objective To study the effect of flavones extract from Juglans mandshurica Maxim stem-barks on the p73-gene expression in hepatocarcinoma cell Bel-7402.Methods Eight groups of person liver cancer Bel-7402 cell were raised in vitro,total RNA were extracted after the cells were processing by different concentrations of Hickory bark extract flavonoids,and the mRNA of p73-gene in Bel-7402 cells were detected by RT-PCR.Results Sustained action for 48 hours,The target band grayscale amount of lower dose group,low dose group,middle dose group and high dose group were respectively(49.2±2.6)%,(47.5±2.2)%,(29.6±1.6)% and(13.3±1.8)% of total grayscale amount.Comparison among one of lower dose group and low dose group,middle dose group,high dose group were significantly different(all P0.05).The p73-gene mRNA expression cut down gradually with the dose stepping up,and the inhibitory effect would be reinforced with the dose to raise.Conclusions The effects of flavones extact from Juglans mandshurica Maxim stem-barks inhibiting tumor cell proliferation,promoting apoptosis may be implemented by inhibiting the overexpression of the p73 gene.

Abstract: Objective To study the relationship between integrin alpha-2(ITGA2) gene C807T polymorphisms and cerebral infarction(CI) and the effect of integrin gene polymorphisms on plasma lipid on lipoprotein levels,and to analyze the nosogenesis of CI.Methods The polymorphism of ITGA2 gene was analyzed by the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method in patients with CI group and healthy controls.The plasma lipid and lipoprotein levels were measured by routine method.Results The distributions of ITGA2 gene C807T polymorphism was significantly different between CI group and control group(P0.05),The relative risk suffered from CI of T allele was 1.635 times of the C allele(OR=1.635,95%CI:1.232~2.171),The level of plasma lipid T allele carriers was significantly higher than that of no carriers(P0.05).Conclusions ITGA2 gene C807T polymorphism associates with CI,T allele carrier may be an increasing risk by affecting the levels of plasma lipid and lipoprotein of CI.

Abstract: Objective To explore the effect of Jiaweibugan decoction on peripheral neuropathy in experimental diabetic rats.Methods Forty diabetic rats were induced by 1% streptozotocin in which 20 rats were cured by Jiaweibugan decoction.Rats in each group were killed on 4th or 8th week after the beginning of treatment.The c-jun and PKC-βⅡ mRNA of sciatic nerve were detected by using RT-PCR and the brain-derived neurotrophic factors(BDNF) protein of sciatic nerve was detected by method of immunohistochemistry assay.Results The expression of c-jun and PKC-βⅡ in rats treated by Jiaweibugan decoction were lower than those in non-treated model rats(P0.05).The expression of BDNF protein in rats treated by Jiaweibugan decoction were higher than those in non-treated model rats(P0.05).Conclusion Jiaweibugan decoction can reduce the oxidative stress reaction and has a preventive and therapeutic effect on peripheral neuropathy in experimental diabetic rats.

Abstract: Objective To study the inhibitory effects of silver nanoparticles on influenza virus H3N2.Methods MTT test,cytopathic effect(CPE) test,transmission electron microscopy(TEM) and flow cytometry were used to determine the inhibitory effects of silver nanoparticles on influenza virus H3N2.Results The maximal atoxic concentration(TD0) of silver nanoparticles and the tissue culture infective dose(TCID50) of H3N2 on MD-25 cell was 25 μg/ml and 10-3.5/0.1 ml,respectively.The apoptosis rate of MD-25 cell was(5.51±1.06)% after the 50 μg/ml nanosilver solution mixed with 40 TCID50 influenza virus H3N2 for 2 hours and the CPE was negative,the apoptosis rate of MD-25 cell in 20 TCID50 influenza virus H3N2 group was(22.54±2.57)% and the CPE was positive(P0.01).Conclusion Nanosilver has obvious inhibitory effects on influenza virus H3N2.

Abstract: Objective To study the effect of bisphenol A(BPA) on the proliferation of 3AO,HHUA and DU145 cells.Methods 3AO cells,HHUA cells and DU145 cells were exposed to BPA and 17β-Estradiol(E2) in different concentrations.The proliferation activities of different cells were detected by MTT assay at 48 h.Then the absorbance values of different cells treated with 10-9 mol/L BPA or 10-7 mol/L E2 were obtained at 0 h,24 h,48 h and 72 h.Results Compared with solvent control,BPA could significantly stimulate 3AO,HHUA and DU145 cells to proliferate in the concentration of 10-5、10-9 mol/L at 48 h(P0.05).In addition,10-9 mol/L BPA could significantely promote HHUA and DU145 cells to proliferate in a time-dependent manner(P0.05).Conclusions The contamination of BPA might be associated with the occurrence and development of hormone-related tumors.

Abstract: Objective To investigate mechanism of cryodamage on cell viability.Methods Human hepatoma carcinoma cell line BEL-7402 were cryopreserved by the same cooling method with different concentrations of electrolytes and cryoprotective agents.The apoptosis,cell necrosis,cryosurvival and debris rates of the cells,and cell cycle of BEL-7402 before and after freezing were analyzed by flow cytometry.Results Cell necrosis and debris were obvious(P0.05) without cryoprotective agents after freezing.When the concentrations of electrolytes and DMSO increased,cell necrosis and debris diminished obviously,while apoptosis turned to be significant gradually(P0.05);There were no significant variation of cell cyle after freezing(P0.05).Conclusions There are two independent kinds of cryodamage factors: one is intracellular ice damage,which can bring about cellular necrosis,and the other is solute-related damage(including electrolyte factors and cryoprotectants),which may induce apoptosis.Both of these two kinds of cryodamage have no cyclic specificity.

Abstract: Objective To study the effects of Tan ⅡA on the expression of HIF-1α,c-Myc in gastric cancer SGC-7901 cell under hypoxia and the correlation between the expression of HIF-1α and c-Myc.Methods The model of hypoxia was established by CoCl2.There were three groups: normal control group hypoxia control group and hypoxia combined with different concentrations of Tan ⅡA group.After Tan ⅡA being added to the media with 0.5,2.0 and 10.0 mg/L respectively for 48 h during hypoxia,the expression of HIF-1α and c-Myc protein were detected by immunocytochemical staining.Results Immunocytochemical staining revealed that the expressions of HIF-1α and c-Myc protein were inhibited by Tan ⅡA in a dose-dependent manner under hypoxia(P0.05).There existed high positive correlation between the expressions of HIF-1α and c-Myc(r=0.901,P0.05).Conclusion The expressions of HIF-1α and c-Myc protein are inhibited by Tan ⅡA under hypoxia,suggesting that Tan ⅡA might play an important role in inhibiting anaerobic glycolysis of tumor and VEGF expression.

Abstract: Objective To explore the association between GSTM1 gene and oligozoospermia.Methods The PCR technique was used to analyze the GSTM1 gene polymorphisms in 75 patients with oligozoospermia(observed group) and 36 healthy men controls(control group).Results The miss rate of GSTM1 gene in the observed group was significantly higher than that in the control group(P0.05).Conclusion The polymorphisms in GSTM1 may have modulating effects on human oligozoospermia,the particula mechanism needs further studies.

Abstract: Objective To explore the correlation between vitamin D receptor(VDR) Fok Ⅰ polymorphism and the breast cancer in Han nationality of North China.Methods After the preparation of gene DNA by common method,VDR genotypes were determined by Fok Ⅰ restriction endonuclease digestion of PCR-amplified DNA in 81 breast cancer patients and 78 healthy persons.Results The genotype frequencies of FF,Ff,ff and F,f allele frequencies of VDR in healthy persons were 33.33%,56.41%,10.26%,61.5%,38.5%,respectively,which were both significantly different as compared to breast cancer patients with respective frequencies of 19.75%,59.26%,20.99%,49.4%,50.6%(P0.05).The odds ratio of ff and Ff genotypes suffering breast cancer were 1.84 and 1.30 respectively(P0.01).Conclusion VDR f allelic may be a genetic mark gene of breast cancer.

Abstract: objective To study the effect and mechanism of evodiamine on invasion and proliferation in human gastric cancer cell.Methods Human gastric cancer SGC7901 cell were treated with different doses of evodiamine,and the growth of anchorage independence of cancer cell was studied by colony formation in soft agar,and invasion ability were determined by Boyden chamber,and the level of midkine gene mRNA and protein were detected by real time RT-PCR and Western blot assay,respectively.Results Evodiamine could significantly inhibit both invasion ability and anchorage independence growth in dose-dependent manners.The level of mRNA and protein of midkine of cancer cells treated with evodiamine reduced in time-and dose-dependent manners.Conclusion Evodiamine can inhibit invasion of gastric carcinoma cell through down-regulating of midkine expression.

Abstract: Objective To study the effect of umbilical cord blood stem cell transplantation therapy on decompensated cirrhosis.Methods Before operation,the conventional liver protective therapy were performed on 86 patients of decompensated cirrhosis.The stem cells for transplantation were got from umbilical cord blood using cell isolating technique,and then injected into the patients' hepatic artery,leading to the liver.Patients were followed up liver functions on the first week,fourth week,tenth week and sixteenth week after stem cell transplantation respectively,and were followed up liver CT examination on tenth week and sixteenth week.Results Compared with preoperation,the symptoms of all 86 patients improved significantly during the first to tenth weeks.The serum albumin levels and PTA improved greatly on sixteenth week after operation(all P0.01);ALT and AST significantly decreased(all P0.05).The liver CT of sixteenth week after operation showed that the RAmax of liver increased significantly campaired with preoperation(P0.05).Conclusions Umbilical cord blood stem cell transplant therapy could rescue liver injury,improve liver function and enhance the patients' quality of life.

Abstract: Objective To investigate the value of combined examination of three kinds of tumor markers.Methods The neuronspecific enolase(NSE),cell keratin 19 fragments(CYFRA21-1)and carcinoembryonic antigen(CEA)levels in pleural effusion and serum of 86 patients with lung cancer(lung cancer group),52 patients with benign lung diseases(TB group) and 67 normal adults(control group)were measured by electro-chemi-luminescence immunoassay assay.Results The levels of NSE,CYFRA21-1 and CEA in pleural effusion and serum of the lung cancer group were all significantly higher than those of the TB group and control group(all P0.01);the levels of NSE in patients with small-cell lung cancer were significantly higher than those in patients with the other subtypes of lung cancer,the levels of CYFRA21-1 in patients with squamous-cell carcinoma were significantly higher than those in patients with the other subtypes of lung cancer(all P0.01).The sensitivity of the three kinds of tumor markers for diagnosing lung cancer was significantly enhanced when they were combined.Conclusion Combined examination of the aboved three kinds of tumor markers in pleural effusion can increase the diagnostic sensitivity for lung cancer.The examination of NSE and CYFRA21-1 is helpful for the differentiation of different subtype lung cancer.

Abstract: Objective To investigate the influence of HBV persistent infection on autoantibody profile.Methods ELISA method was employed for ANA,anti-ss-DNA and RF from serum of 259 HBV persistent infections(observed group)and 260 healthy donors(control group).Results The positive rate of ANA,RF and anti-ss-DNA in observed group were significantly higher than those in control group(all P0.05),but no distinct difference were found between HBV-DNA positive group and HBV-DN A negative group.Conclusions The changement of autoantibody profile is associat ed with HBV persistent infection.It provide the foundation for further study of the effect of autoantibody to viral hepatitis type B development and final outc ome and the trigger mechanism of autoimmunity for viral hepatitis type B patient s.

Abstract: Objective To evaluate the effects and safety of HIFU on the patients wi th unresectable pancreatic cancer.Methods Thirty-six cases with unresectable pa ncreatic carcinoma were divided into treatment group(n=21) and control group(n=1 5) randomly.Both groups were given routine supporting therapy,the treatment gro up received HIFU treatment furthermore.When the course of treatment finished,t he clinical benefit reponse(CBR),clinical curative effect,serum level of CA19-9, life span,survival rate and adverse reaction were observed.Results Compared to t he control group,the positive rate of CBR,the remission rate of single pain,the general effective rate and survival rate of 12,18 months increased in the treatm ent group,the rate of PD,the incidence of adverse reaction and serum level of CA 19-9 decreased,the life span prolonged,P0.05 or 0.01.Conclusions HIFU treatmen t can alleviate sufferings,improve life quality and extend life span of patient s with pancreatic carcinoma,and also posses high safety.

Abstract: Objective To observe the change of stathmin and β-tubulinⅢ expression in NSCLC and its influence to taxol sensitivity.Methods Expression of stathmin and β-tubulinⅢ in 32 patients with NSCLC who received radical surgery paclitaxel-based chemotherapy were determined by immunohistochemical method.It is that the correlation of their expression analized with prognosis and prediction value in chemotherapy.Results The positive rates of stathmin,β-tubulinⅢ were 52.8% and 62.6%,significant more than inflammatory pseudotumor(P0.05).The positive rates of stathmin have significant difference in poorly differentiated group and lymphatic metastasis group(P0.05);The positive rates of β-tubulin Ⅲ have significant difference in poorly differentiated group and Ⅲ~Ⅳ state group(P0.05).The expression of stathmin,β-tubulinⅢ in the patients who had postoperative chemotherapy were subdivided into two group(all positive,all negative),there showd significant survival difference between all positive and all negative.Conclusion The expression of stathmin and β-tubulin Ⅲ increase in NSCLC tissues.It suggests that the tumor is malignant taxol-unsensitive and poor prognosis.

Abstract: Objective To investigate the expression of forkhead transcription Foxp3 in hepatocarcinoma cell lines and to explore its significance in tumor immune micro-environment.Methods Conventional RT-PCR technique and cDNA expression profile were used to detect the expression of Foxp3 mRNA in 10 hepatocarcinoma cell lines.Protein expression was assessed by using Western blot,immunocytochemistry and flow cytometry methods.Down-regulation of Foxp3 was used by Foxp3 shRNA in hepatocellular carcinoma(HCC) cells.The immune-related inhibitory functions in Foxp3-expressing HCC cells were evaluated by co-culturing tumor cells with native CD+4T cells.Results Foxp3 mRNA and protein were detected in all HCC cell lines.The hepatoma cells could significantly inhibit of CD+4T cell proliferation in the co-culture system.There could be a significant reversal of CD+4T cell proliferation and the expression of surface marker after silencing the expression of Foxp3 by shRNA in hepatoma cells and decrease the expression of Foxp3 in CD+4T cells.The Foxp3-expressing HCC cells could inhibit the CD+4effector T cells proliferation.Conclusion Foxp3 expressed in all HCC cells and it might be involved in tumor cell mediated immune-suppression.

Abstract: Objective To investigate the relationship between the levels of serum transforming growth factor β1(TGF-β1) and liver fibrosis in chronic hepatitis B patients.Methods The levels of serum TGF-β1,hyaluronic acid(HA),procollagenⅢ(PCⅢ) and laminin(LN) were respectively detected by enzyme-liked immunosorbent assay and radioimmunoassay in 73 patients with chronic hepatitis B(CHB) and liver cirrhosis(LC),and liver tissues in 42 cases of patients were stained.The change of those factors in different stages of liver fibrosis were observed.Results The levels of serum TGF-β1,HA,PCⅢ and LN were gradually increased with the progression of the disease(P0.05).There was positively relationship between TGF-β1 and HA,PCⅢ(r=0.457,0.363,all P0.05).The levels of serum TGF-β1 and HA in S1~2,S3~4 phase were significantly higher than that in S0 phase(all P0.05),and the levels of serum TGF-β1 and HA in S3~4 phase was significantly higher than that in S1~2 phase(P0.05).Conclusions The levels of serum TGF-β1 are closely related with liver fibrosis.Detecting HA and PCⅢ could judge the degree of liver fibrosis.

Abstract: Objective To explore the anti-tumor mechanism of curcumin furtherly.Methods The proliferation inhibition rate of Hep1 cells treated with different concentrations of curcumin for different time was observed by CCK-8 method,the apoptosis and cycle phase of which was measured by FCM.Results The proliferation inhibition rate of Hep1 cell significantly increased after the treatment of curcumin in a dose-dependent manner.After the treatment of 15 μmol/L curcumin,cells in G0+G1 phase increased significantly,while cells in G2+M phase reduced significantly and apoptosis rate increased(P0.05).Conclusions Curcumin can effectively inhibit the cell growth and proliferation of Hep1 in a clear dose-response relationship,and the mechanism may be that curcumin can interfere cell cycle progression and promote cell apoptosis.

Abstract: Objective To evaluate the effects and mechanism of carvedilol on intimal proliferation in rats with balloon injury.Methods Ninety male Wistar rats were randomly divided into the sham-operated group,the injury group and the CAR group with 30 in each,and the model of carotid balloon injury were established in the later two groups.Carotid tissues of the three groups were harvested 1,3,7,14 and 28 days after the injury with 6 rats at each time point.The intimal proliferation was observed by optical microscope.The expression of IGF-1 mRNA at different time points after ballon injury was examined with the method of RT-PCR.Results Compared with the injury group,the neointima areas reduced in the CAR group at the 14th day and 28th day,the IA/MA ratios decreased(P0.01)and the lumen areas increased(P0.01).Compared to the sham-operated group,the expression of IGF-1mRNA significantly increased in the the injury group after 3,7,14,28 days of balloon injury,and reached the maximum at the 14th day(P0.01).The expression of IGF-1mRNA at every time points in the CAR group were significantly lower than that in the injury group,especially at the 14th day(P0.01).Conclusion Carvedilol can significantly attenuate intimal proliferation of the rat carotid arteries after balloon injury,the mechanism may be related to its inhibitory effect on the expression of IGF-1mRNA.

Abstract: Objective To investigate the antifibrotic action and mechanism of artesunate furtherly.Methods Human embryonic lung fibroblasts(HELF) cell lines HFL-I were cultured to log phase in vitro and divided into 3 observed groups and control group,the former 3 groups were treated with artesunate at the mass concentration of 1,10,100 mg/L,the control group were given equivalent DMEM/F-12.The collegan synthesis was determined by sirius red staining,the expression of type Ⅰ procollagen and type Ⅲ procollagen mRNA was detected by RT-PCR.Results Compared with the control group,the expression of collagen and type Ⅰ procollagen,type Ⅲ procollagen mRNA in observed groups decreased significantly,and remarkable differences were also seen among observed groups(P0.05).Conclusion Artesunate can resist pulmonary fibrosis and reduce the secretion of collagen in some extent,the mechanism may relate to the reduction of the expression of type Ⅰ procollagen and type Ⅲ procollagen mRNA.
