Abstract: Objective To study the relationship between apolipoprotein B( ApoB) gene polymorphism and the tendonvessel stagnation of steroid induced osteonecrosis of the femoral head( SONFH). Methods A total of 108 patients with SONFH were included in this study,including 64 patients with tendon-vessel stagnation syndrome of TCM( tendon-vessel stagnation syndrome group) and 44 patients with the deficiency of liver and kidney syndrome of TCM( deficiency of liver and kidney syndrome group). There were 98 patients who adopt adrenal cortical hormone for treatment in 6 months without SONFH collected as control group. DNA of each group were extracted from the blood sample. The direct sequencing polymerase chain reaction and ligase detection reaction( PCR-LDR) technique were used to analyze XbaⅠ( rs693) and EcoRⅠ( rs1042031) singlenucleotide polymorphism of ApoB gene. Results Two gene type,X-X-and X + X-,were detected in XbaⅠ site. The amounts of X + X- genotype and X + allele frequencies of XbaⅠ polymorphism in tendon-vessel stagnation syndrome group were higher than those in deficiency of liver and kidney syndrome group( all P 0. 05). Two gene type,which were E + E + and E +E-,were detected in EcoRⅠ site. The amounts of E + E-genotype and E-allele frequencies of EcoRⅠ polymorphism in tendonvessel stagnation syndrome group were higher than those in deficiency of liver and kidney syndrome group( all P 0. 05).However,the difference between the deficiency of liver and kidney syndrome and the control group showed no significance( P 0. 05). Conclusion The XbaⅠ X + X- genotype,XbaⅠ X + allele frequencies,EcoRⅠ E + E- genotype and EcoRⅠE- allele frequencies may be related to the tendon-vessel stagnation syndrome of SONFH.

Abstract: Objective To investigate the chemical compositions of Cortex Juglans Mandshurica and to evaluate anti-tumor effects of these compounds in human hepatoma cells Methods Cortex Juglans Mandshurica were extracted with 75% ethanol,concentrated with chloroform and ethyl acetate,eluted radiantly on silica gel,and purified by preparative HPLC Physicochemical constants and spectroscopic analysis were used to identify their structures MTT assay was used to evaluate their anti-hepatocarcinoma bioactivities Results Two compounds were isolated and elucidated as luteolin and formononetin Luteolin had strong cytotoxic effect on HepG2 and LO2 with IC50 values of( 103 32 ±21 34) and( 131 13 ±13 48) μmol /L; Formononetin showed lighter cytotoxic effect on HepG2 with IC50 200 μmol /L Conclusion Luteolin and formononetin are isolated from Cortex Juglans Mandshurica for the first time,and luteolin shows significant anti-tumor activity in human hepatoma cells

Abstract: Objective To evaluate the application effect of minimally invasive transverse aortic constriction procedure in the construction of heart failure animal model in mice. Methods Thirty mice were randomly divided into the sham operation group( n = 6) and the constriction group( n = 24). Mice in the constriction group underwent minimally invasive operation ligation of the aortic arch through neck incision,and mice in the sham operation group were not ligated,but other operations were the same as the constriction group. B ultrasonography was performed on the left and right common carotid artery and heart in the constriction group in order to evaluate the effect of established models after the operation. Results Twelve rats died in the constriction group,and no one in the sham operation after the surgery. As the prolonged operation time,the right common carotid artery diameter increased gradually in the constriction group and the left common carotid artery reduced; at the same time point,the blood flow velocity and diameter in the right common carotid artery were greater than the left( all P 0. 05); left ventricular ejection fraction,left ventricular shortening fraction gradually lowered,the diastolic interventricular septum thickness,left ventricular posterior wall thickness,left ventricular correction capacity and quality gradually increased( all P 0. 05). Conclusion Minimally invasive transverse aortic constriction procedure can successfully prepare the mouse models of heart failure.

Abstract: Objective To analysis the characteristics of proton magnetic resonance spectroscopy(1H-MRS) on hippocampus in patients with post-stroke depression. Methods A total of 32 patients with post-stroke depression( research group),30 patients with stroke( case-control group) and 30 normal controls( control group) were assessed in this study. A multi-voxel1H-MRS was given to all the subjects on hippocampus within 24 hours they got in hospital. The NAA,Cho and Cr were measured and the ratios of NAA / Cr and Cho / Cr were determined. Results In left and right hippocampus, the NAA / Cr ratio in research group demonstrated lower than that in other two groups. In left hippocampus,the Cho / Cr ratio in research group showed higher than that in other two groups( P 0. 05); in right hippocampus,the Cho / Cr ratio was higher than that in other two groups( P 0. 05). In left and right hippocampus,the NAA / Cr ratio in the severe depression group was lower than that in the mild depression group( P 0. 05),there was no significant difference in the Cho / Cr ratio among the different degree of depression( P 0. 05). In left and right hippocampus,the NAA / Cr ratio in research group showed a significant negative correlation with the HAMD scores( P 0. 05); the Cho / Cr ratio not showed a significant correlation with The HAMD scores( P 0. 05). In the three groups,there were no significant differences in NAA / Cr and Cho / Cr between bilateral hippocampus( P 0. 05). Conlusions Abnormal metabolism of hippocampal neurons maybe the neurobiological basis of post-stroke depression. There is no significantly lateralized asymmetry for metabolite levels such as NAA,Cho among post-stroke depression,stroke patients and normal subjects.

Abstract: Objective To investigate the changes of the function of mouse dendritic cells under the condition of hypothermia and the regulation of Yu Ping Feng pulvis. Methods Fifty BALB / c mice were randomly divided into 5 groups: the control group at room temperature of 25 ℃,model group at low temperature of 10 ℃,low dose,medium dose and high dose groups of Yu Ping Feng pulvis. And on the fourth day of medication,the other four groups except the control group were placed into the equipment of hypothermia for another three days,and each day for six hours. The expression of MHC class II molecules,co-stimulatory molecules CD86and chemokine receptor CCR7 on dendritic cells was detected by flow cytometry; the levels of CD86,CCR7 mRNA were measured by reverse transcription-polymerase chain reaction( RT-PCR). Results Compared with the control group,the rectal temperature of model group at hypothermia was lower,and so was the expression of MHC class II,CCR7,CD86and the mRNA expression of CCR7,CD86( all P 0. 05); compared with the model group,Yu Ping Feng pulvis enhanced the expression of MHC-II,CCR7 and CD86( all P 0. 05). Conclusions Under the condition of hypothermia,the expression of MHC class II,CCR7 and CD86on the surface of dendritic cells is down-regulated,while the phenomena could be reversed by Yu Ping Feng pulvis and then it enhances the function of information transfer of dendritic cells.

Abstract: Objective To observe the expression changes of Raf kinase inhibitor protein( RKIP) and epithelial cadherin( E-cadherin) in gastric cardia adenocarcinoma( GCA) and to explore their correlation. Methods The expression of RKIP and E-cadherin were detected in 160 cases of GCA tissues and 160 cases of adjacent cancerous tissues by immunohistochemical SP method. We analyzed the relationship between their expression and clinical pathological parameters as well as their correlations. Results The expressions of RKIP and E-cadherin in GCA tissues were lower than that in the adjacent cancerous tissues( all P 0. 05). The expression of RKIP and E-cadherin in highly,moderately,poorly differentiated GCA was significantly different( P 0. 01) and the expression in GCA tissues with lymphatic metastasis was lower than that without lymphatic metastasis( all P 0. 05). The expression of RKIP and E-cadherin in GCA tissues were positively correlated( r =0. 388,P 0. 05). Conclusion The expression of RKIP and E-cadherin in GCA tissues is decreased and positively correlated with each other.

Abstract: Objective To explore the correlation between miR-7 and expression of EGFR /PI3 K signal pathway related protein( EGFR,PI3 K and AKT2) in brain glioma tissues and U251 cells. Methods The tumor tissues were obtained from 42 patients with brain glioma by excision,including 8 cases of tumor adjacent normal brain tissue. U251 glioma cells were cultured,and then were divided into the blank control group( only with liposomes),nonsense sequence group( nonsense sequence and liposomes) and miR-7 transfection group( transfection of miR-7 gene fragments). The expression of miR-7 in the glioma tissues and U251 cells was determined by RT-PCR. The expression of EGFR,PI3 K and AKT2 protein in glioma tissues was detected by immunohistochemitry,and RT-PCR and Western blotting were used to detect the mRNA and protein expression of EGFR,PI3 K and AKT2 in the U251 cells. Results The mRNA expression of miR-7 in the glioma and normal brain tissues were 1. 47 ± 0. 27 and 3. 47 ± 0. 15,respectively( P 0. 05). With the increase of pathological grades of glioma,the expression of miR-7 decreased,the EGFR,PI3 K and AKT expression increased( all P 0. 05). The expression of miR-7 was negatively correlated with EGFR,PI3 K and pAKT expression in the glioma tissues( r =-0. 754,-0. 528 and-0. 467,respectively; all P 0. 05). The EGFR,PI3 K and AKT expression was significantly reduced in the U251 cells after being transfected with miR-7( all P 0. 05). Conclusions The expression of miR-7 in the glioma tissues is down-regulated,which is negatively correlated with the EGFR /PI3 K pathway related protein. The expression of EGFR,PI3 K and AKT is decreased in the U251 cells after being transfected with miR-7. The EGFR /PI3 K pathway may be one of the important mechanisms of miR-7 in regulating the occurrence and development of gliomas.

Abstract: Objective To explore the spontaneous chloride currents in poorly differentiated nasopharyngeal carcinoma( CNE-2Z) cells.Methods The whole-cell patch-clamp technique was used to detect the Cl-currents.The characteristics of the currents were clarified by ATP depletion in the pipette solution,extracellular applications of chloride channel blockers and hypertonic challenges.Results A chloride current was activated spontaneously and occasionally in the isotonic solution in CNE-2Z cells,showing significant outward rectification but no time-dependent inactivation.The reversal potential of the current was(- 7.6 ± 0.9) mV,which was close to the calculated equilibrium potential for Cl-.The induction of the chloride channel was dependent on the intracellular ATP.The spontaneous chloride currents were inhibited by extracelluar applications of the chloride channel blocker ATP and 47% hypertonic solution.Conclusion A chloride channel can be activated spontaneously and occasionally under the isotonic condition in CNE-2Z cells,showing the similar characteristics to the volume-sensitive chloride channel.

Abstract: Objective To investigate the effect of ecdysterone on the expression of insulin receptor substrate 2( IRS-2) in liver cell of type 2 diabetic rats Methods 14 of the 42 rats were randomly selected to the normal control group,and was given ordinary rat food The other 28 rats were made into type 2 diabetes mellitus rat models by using the high fatty diet and small dose of SIZ,and then diveided into model group and treatment group The rats in model group or treatment group was given high fatty diet or high fatty diet fecdysterone IRS-2 protein and mRNA in liver tissues of rats were detected by immunohistochemistry assay and RT-PCR assay after five weeks Results The expression of IRS-2 protein and mRNA in normal group and treatment group were higher than those of the model group( P 0 01 or 0 01) Conclusions Ecdysterone led to up-regulation expression of IRS-2 protein and IRS-2 mRNA of liver tissue in diabetic rats This might play a role in ecdysterone-improved hepatic insulin resistance

Abstract: Objective To analyze the risk factors of acute ischemic stroke( IS),and to investigate the combined detections of plasma homocysteine( Hcy) and ESSEN score in predicting progressive stroke( PS). Methods Totally 126 patients with acute IS of onset within 24 h were included. According to PS definition,patients were divided into the PS group( 30 cases) and non-PS group( 96 cases). The age,gender and history of hypertension in the two groups were compared. and multivariate analysis were used to assess the risk factors of acute IS developing into PS. According to Hcy level,all patients were divided into two groups: the normal group( ≤15 μmol /L) and the increased group( 15 μmol /L),and then according to ESSEN score ≤2 and 2,they were respectively divided into two groups. Hcy level ≤ 15 μmol /L and ESSEN score 2 was designed as baseline,other groups were respectively compared with the baseline group to evolve the short-term risk of PS. Results Fibrinogen( FIB) level,unstable carotid plaque,intracranial artery stenosis,dysphagia,Hcy and ESSEN score were higher in the PS group than those in the non-PS group( all P 0. 05); the number of cases with diabetes history was less in the PS group than that of the non-PS group( P 0. 05). Multivariate analysis showed that FIB level,intracranial artery stenosis,Hcy level and ESSEN score in the PS group were higher than those in the non-PS group( all P 0. 05). Patients with Hcy level 15 μmol /L and ESSEN score 2 had higher short-term risk of PS. Conclusion FIB level,intracranial artery stenosis,Hcy level and ESSEN score are the risk factors of PS. Hcy combined with ESSEN score in predicting risk of PS has good predictive value.
