Abstract: 目的:分析自噬基因Beclin1和核因子NF-κB(nuclear factor-kappaB,NF-κB)在原发性肝细胞性肝癌(primary human hepatocellular carcinoma,HCC)中的表达及其临床意义.方法:收集顺德第一人民医院2003-01/2007-12手术切除经病理学证实的HCC50例,肝炎后肝硬化手术切除或活检标本30例,肝炎患者肝脏穿刺组织30例和正常肝组织10例.应用免疫组化S-P法检测肝脏组织中Beclin1和NF-κBp65蛋白的表达.结果:原发性肝细胞性肝癌、肝硬化、肝炎和正常肝组织的Beclin1蛋白阳性表达率分别是78.00%(39/50)、26.67%(8/30)、53.33%(16/30)、10.00%(1/10),四者之间的差异有统计学意义(χ2=28.31,P〈0.05),且Beclin1在原发性肝细胞性肝癌组织表达明显高于在肝硬化组织,肝炎组织,正常肝组织中的表达(χ2=20.39,5.31,14.41,P〈0.05);在肝炎组织表达明显高于在肝硬化组织,正常肝组织中的表达(χ2=4.44,χ2=4.12,P〈0.05).原发性肝细胞性肝癌、肝硬化、肝炎和正常肝组织的NF-κBp65蛋白阳性表达率分别是74.00%(37/50)、36.67%(11/30)、30.00%(9/30)、20.00%(2/10),四者之间的差异有统计学意义(χ2=22.00,P〈0.05),且NF-κBp65在原发性肝细胞性肝癌组织表达明显高于在肝硬化组织,肝炎组织,正常肝组织中的表达(χ2=10.89,χ2=14.85,χ2=8.44,P〈0.05).结论:Beclin1和NF-κBp65在HCC中的表达关系密切,呈正相关;Beclin1和NF-κBp65异常表达与HCC的发展密切相关,在HCC的发展过程中起重要的作用.

Abstract: 目的:研究拟胆碱药卡巴胆碱对烫伤大鼠口服补液时小肠TNF-α及水通道蛋白-1(AQP-1)表达的影响. 方法:♂Wistar大鼠50只,随机分为假烫(N)、单纯烫伤(S)、肠内葡萄糖-电解质液(GES)、肠内卡巴胆碱组(CAR)和肠内葡萄糖-电解质液+卡巴胆碱组(GES/CAR)5组(n=10).大鼠背部用沸水造成35%TBSA烫伤.N,GES和GES/CAR组于伤后30min开始补液.免疫组化法测定肠组织AQP-1的表达,ELISA法检测肠组织TNF-α含量,酚红法测定大鼠小肠对水的吸收率. 结果:S组大鼠小肠AQP-1与N组比明显降低(90.3±18.4 vs 4851.6±654.5,P〈0.01):CAR,GES和GES/CAR组AQP-1与S组相比均显著增加(1806.1±110.1,2272.3±113.8,3322.0±595.9 vs 90.3±18.4,均P〈0.01).给予卡巴胆碱组(CAR,GES/CAR)与未给予卡巴胆碱组(S,GES)相比肠组织TNF-α含量明显下降(0.9±0.3,1.0±0.47 vs 1.8±0.3,1.9±0.2,P〈0.05).GES/CAR.CARTLS组AQP-1表达量与TNF-α含量成负相关(r=0.9030,-0.9602,-0.9866,均P〈0.05).GES/CAR组水吸收率较GES组明显升高(21.0%±0.1% vs 12.7%±0.1%,P〈0.05). 结论:卡巴胆碱可抑制促炎因子TNF-α的释放,上调小肠AQP-1表达,改善大鼠烫伤早期肠道对水的吸收.

Abstract: N A D P H o x i d a s e ( n i c o t i n a m i d e a d e n i n e dinucleotide phosphate-oxidase, NOX) is a multiprotein complex producing reactive oxygen species (ROS), present both in phagocytes, being essential in host defense and in non-phagocytic cells, regulating intracellular signaling. In liver, NADPH oxidase plays a central role in fibrogenesis. A functionally active form of NADPH oxidase is expressed not only in Kupffer cells (phagocytic cell type) but also in hepatic stellate cells (HSCs) (non-phagocytic cell type), suggestive of its role the non-phagocytic NADPH oxidase in HSCs activation. This paper reviewed effects of NOX in liver fibrogenesis.

Abstract: AIM: To invest igate the gene expression profiles in tissues of normal human liver, chronic hepatitis B, cirrhosis and hepatocellular carcinoma (HCC) by oligonucletide chip, and to screen HCC-related genes. METHODS: The total RNA was extracted and reverse transcribed into double-strand cDNA, and then transcribed into biotin-labled cRNA target probes. The probes were hybridized with oligonucleotide chip containing 19378 genes respectively. Subsequently, the signal images were scanned by gene Scanner 3000 and analyzed with GenePix Pro 3.0 software. RESULTS: Eighty-one genes differentially expressed among tissues of chronic hepatitis B, cirrhosis and HCC were screened out in the gene expression profiles. Of the 81 genes, 53 genes were consistently up-regulated, while 28 genes were consistently down-regulated. CONCLUSION: The oligonucleotide chip of gene expression profile is powerful for screening HCC-related genes. Our results indicate that multiple genes take part in the carcinogenesis of HCC.

Abstract: 目的:检测Ezrin及细胞黏附分子E-cadherin、CD44在胃癌细胞的表达,了解Ezrin对瘤细胞黏附分子调节及对胃癌浸润生长的影响.方法:手术切除胃癌标本80例(其中62例附癌旁胃组织)行Ezrin、E-cadherin和CD44v6免疫组化染色.结果:Ezrin的表达与胃癌的转移呈正相关,与分化程度呈负相关(均P〈0.05)、CD44的表达与胃癌的浸润深度,淋巴结转移呈正相关,与分化程度呈负相关(均P〈0.05).E-cadherin的表达与胃癌的浸润深度、转移及分化程度呈负相关(均P〈0.05).Spearman等级相关分析显示,Ezrin与CD44的表达呈显著正相关(rs=0.305,P〈0.05),Ezrin与E-cadherin的表达呈负相关(rs=-0.314.P〈0.05).结论:胃癌组织Ezrin增加与肿瘤细胞的分化有关,并可调节E-cadherin、CD44而影响胃癌细胞的黏附性.
