Abstract: An alcoholic extract of freshly collected Eclipta alba exhibited dose-dependent (62.5–500.0 mg/kg p.o.) significant hepatoprotective activity against carbon tetrachloride-induced liver injury in rats and mice. It indicated its protective role on parameters such as hexobarbitone-induced sleep, zoxazolamine-induced paralysis, bromosulphalen (BSP) clearance, serum levels of transaminases, bilirubin and protein. The extract did not show any signs of toxicity and the minimum lethal dose was greater than 2.0 g/kg when given orally and intraperitoneally in mice.

Abstract: This study was designed to evaluate the protective effect of kolaviron (KV), a biflavonoid from Garcinia kola seeds, against gamma-radiation (5 Gy)-induced oxidative stress in brain of Wistar rats. Vitamin C (VC) served as standard antioxidant. Forty-four rats were divided into 4 groups of 11 animals each. One group was un-irradiated (normal), two groups were treated with KV and VC (250 mg/kg) for 6 weeks prior to and 8 weeks after irradiation, and fourth group was only irradiated. Rats were sacrificed 1 and 8 weeks after irradiation. Cellular alterations were monitored using changes in the levels of malondialdehyde (MDA)-an index of lipid peroxidation, superoxide dismutase (SOD), glutathione-S-transferase (GST), reduced glutathione (GSH), catalase (CAT), alanine and aspartate aminotransferases (ALT and AST), urea and creatinine. MDA levels increased significantly (p<0.05) by 90% and 151% after 1 and 8 weeks of irradiation. Furthermore, levels of GSH and antioxidant enzymes were significantly (p<0.05) decreased in gamma-irradiated animals. GSH and GST decreased by 61% and 43% after 1 week, and by 75% and 74%, after 8 weeks of exposure, respectively. gamma-Irradiation decreased SOD and CAT levels by 53% and 68%, respectively, and caused significant (p<0.05) increases in serum ALT, AST and urea after 8 weeks of exposure. Treatment with KV and VC significantly decreased the levels of MDA, ALT, AST and urea. The antioxidant indices were significantly ameliorated in KV-treated animals. These data suggest that kolaviron may protect against gamma-radiation-induced oxidative stress in brain of exposed rats.

Abstract: Freshwater snails are used as sensitive biomarkers of aquatic ecosystem pollution. The potential impacts of zinc oxide nanoparticles (ZnONPs) on aquatic ecosystems have attracted special attention due to their unique properties. The present investigation was designed to evaluate the possible mechanisms of ecotoxicological effects of ZnONPs on freshwater snail Biomphalaria alexandrina. ZnONPs showed molluscicidal activity against B. alexandrina snails, and the LC50 was 145 μg/ml. Two tested concentrations of ZnONPs were selected: The first concentration was equivalent to LC10 (7 μg/ml), and the second was equivalent to LC25 (35 μg/ml). Exposure to ZnONPs (7 and 35 μg/ml) for three consecutive weeks significantly induced malondialdehyde and nitric oxide with concomitant decreases in glutathione and glutathione-S-transferase levels in hemolymph and soft tissues of treated snails. Moreover, ZnONPs elicited a significant decrease in total protein and albumin contents coinciding with enhancement of total lipids and cholesterol levels as well as activities of aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase in hemolymph and soft tissues of treated snails. This study highlights the potential ecological implications of ZnONP release in aquatic environments and may serve to encourage regulatory agencies in Egypt to more carefully monitor and regulate the industrial use and disposal of ZnONPs.

Abstract: The combined hepatoprotective effect of Bi- herbal ethanolic extract (BHEE) was evaluated against carbon tetra chloride (CCl4) induced hepatic damage in rats. Ethanolic extract from the leaves of Eclipta alba and seeds of Piper longum at a dose level of 50 mg/kg body weight was administered orally daily once for 14 days. The substantially elevated serum marker enzymes such as SGOT, SGPT, ALP, LDH, ACP, gGT and 5’ Nucleotidase, due to CCl4 treatment were restored towards normalization. The biochemical parameters like total protein, total bilirubin, total cholesterol, triglycerides, and urea were also restored towards normal levels. In addition, BHEE significantly decreased the liver weight of CCl4 intoxicated rats. Silymarin at a dose level of 50 mg/kg was used as a standard reference also exhibited significant hepatoprotective activity against CCl4 induced hepatotoxicity. The results of this study strongly indicate that BHEE has got a potent hepatoprotective action against CCl4 induced hepatic damage in rats. Abstract: The present study has been conducted to evaluate the curative effect of propolis extract, a honey bee-hive product, against acetaminophen (APAP) induced oxidative stress and dysfunction in liver and kidney. Animals were challenged with APAP (2 g/kg, p.o.) followed by treatment of propolis extract (100 and 200 mg/kg, p.o.) once only after 24 h. Release of transaminases, alkaline phosphatase, lactate dehydrogenase, and serum bilirubin were increased, whereas hemoglobin and blood sugar were decreased after APAP administration. Antioxidant status in both the liver and kidney tissues were estimated by determining the glutathione, malondialdehyde content and activities of the CYP enzymes, which showed significant alterations after APAP intoxication. In addition, activities of adenosine triphosphatase, acid phosphatase, alkaline phosphatase, and major cell contents (total protein, glycogen and cholesterol) were also altered due to APAP poisoning. Propolis extract successfully reversed the alterations of these biochemical variables at higher dose. Improvements in hepatorenal histoarchitecture were also consistent with biochemical observations. The results indicated that ethanolic extract of propolis has ability to reverse APAP-induced hepatorenal biochemical and histopathological alterations probably by increasing the antioxidative defense activities due to various phenolic compounds present in it.
