This work has constructed a series of strains to facilitate the generation and analysis of clones of genetically distinct cells in developing and adult tissues of Drosophila, providing an unprecedented opportunity to perform systematic genetic screens for mutations affecting many biological processes.Abstract:
We have constructed a series of strains to facilitate the generation and analysis of clones of genetically distinct cells in developing and adult tissues of Drosophila. Each of these strains carries an FRT element, the target for the yeast FLP recombinase, near the base of a major chromosome arm, as well as a gratuitous cellautonomous marker. Novel markers that carry epitope tags and that are localized to either the cell nucleus or cell membrane have been generated. As a demonstration of how these strains can be used to study a partic ular gene, we have analyzed the developmental role of the Drosophila EGF receptor homolog. Moreover, we have shown that these strains can be utilized to identify new mutations in mosaic animals in an efficient and unbiased way, thereby providing an unprecedented opportunity to perform systematic genetic screens for mutations affecting many biological processes. TL;DR: A genetic mosaic system in Drosophila is described, in which a dominant repressor of a cell marker is placed in trans to a mutant gene of interest, which allows for the study of gene functions in neuroblast proliferation, axon guidance, and dendritic elaboration in the complex central nervous system.

An evolutionarily conserved function of the Drosophila insulin receptor and insulin-like peptides in growth control
TL;DR: This study shows that the Drosophila insulin receptor autonomously controls cell and organ size, and that overexpression of a gene encoding an insulin-like peptide is sufficient to increase body size.

Evidence that stem cells reside in the adult Drosophila midgut epithelium
TL;DR: The ability to identify, manipulate and genetically trace cell lineages in the midgut should lead to the discovery of additional genes that regulate stem and progenitor cell biology in the gastrointestinal tract.

A novel class of evolutionarily conserved genes defined by piwi are essential for stem cell self-renewal.
TL;DR: The Drosophila piwi gene is cloned and characterized and it is shown that it is required for the asymmetric division of GSCs to produce and maintain a daughter GSC but is not essential for the further differentiation of the committed daughter cell.

Mutations affecting segment number and polarity in Drosophila
TL;DR: The phenotypes of the mutant embryos indicate that the process of segmentation involves at least three levels of spatial organization: the entire egg as developmental unit, a repeat unit with the length of two segments, and the individual segment.

Genetic Transformation of Drosophila with Transposable Element Vectors
TL;DR: A rosy transposon (ry1), constructed by inserting a chromosomal DNA fragment containing the wild-type rosy gene into a P transposable element, transformed germ line cells in 20 to 50 percent of the injected rosy mutant embryos indicating that the visible genetic defect in the host strain could be fully and permanently corrected by the transferred gene.

TL;DR: In this article, six monoclonal antibodies have been isolated from mice immunized with synthetic peptide immunogens whose sequences are derived from that of the human c-myc gene product.
