TLDR
It is shown that, under conditions of dysregulated IL15 expression in vivo in patients with celiac disease and in vitro in healthy individuals, multiple steps of the NKG2D/DAP10 signaling pathway leading to ERK and JNK activation are coordinately primed to activate direct cytolytic function independent of TCR specificity in effector CD8 T cells.

Up on the tightrope: natural killer cell activation and inhibition.

TL;DR: Recent advances in the understanding of the structural properties and signaling pathways of the inhibitory and activating NK cell receptors are highlighted, with a particular focus on the ITAM-dependent activating receptors, the NKG2D-DAP10 receptor complexes and the CD244 receptor system.

The effects of a smoking cessation intervention on 14.5-year mortality: a randomized clinical trial.

TL;DR: The smoking cessation program was associated with cumulative reduced decline in lung function (FEV1) that was largest in participants who stopped smoking early in the study; inhaled ipratropium produced a small noncumulative increase in FEV1 that disappeared when the drug was withdrawn.

The biology of interleukin-2 and interleukin-15: implications for cancer therapy and vaccine design.

TL;DR: The biology of these cytokines will affect the development of novel therapies for malignancy and autoimmune diseases, as well as the design of vaccines against infectious diseases.

Immunity, Inflammation, and Allergy in the Gut

TL;DR: In both mouse and man, mutations in genes that control innate immune recognition, adaptive immunity, and epithelial permeability are all associated with gut inflammation, which suggests that perturbing homeostasis between gut antigens and host immunity represents a critical determinant in the development of gut inflammation and allergy.

American Gastroenterological Association (AGA) Institute Technical Review on the Diagnosis and Management of Celiac Disease

TL;DR: This technical review addresses the state of evidence for celiac disease epidemiology, detection by serologic testing, diagnosis by biopsy, treatment, and outcome.

HLA-E binds to natural killer cell receptors CD94/NKG2A, B and C

TL;DR: The identification of ligands for HLA-E is reported, which shows that a subset of HLA class I alleles has been shown to inhibit killing by CD94/NKG2A+ NK-cell clones, and only the HLA alleles that possess a leader peptide capable of upregulating Hla-E surface expression confer resistance toNK-cell-mediated lysis.
Journal ArticleDOI

Tumour-derived soluble MIC ligands impair expression of NKG2D and T-cell activation

TL;DR: It is shown that binding of MIC induces endocytosis and degradation of NKG2D, a mode of T-cell silencing that may promote tumour immune evasion and, by inference, compromise host resistance to infections.

Roles of the NKG2D immunoreceptor and its ligands

TL;DR: This review summarizes recent findings that concern NKG2D, its ligands, its signalling properties and its role in disease, and provides a framework for considering how the induction of immune responses can be regulated by cellular responses to injury.
