Abstract: Le dosage colorimetrique des acides amines separes par chromatographie sur papier est effectue par un reactif a la ninhydrinehydrindantine apparente a celui de Moore & Stein. Les ions ammonium contenus dans le papier sont elimines avant le dosage par vaporisation d'une solution de potasse methanolique et sechage, ce qui permet de rendre nulle la valeur a blanc du papier lors du dosage. La methode permet de doser de 0,02 a 0,5 micromole d'acides amines.

Abstract: En ajoutant a un acide amine ou peptide N-acyle dans un solvant organique une amine tertiaire (tri-n-butylamine), puis du chloroformiate ( = chlorocarbonate = chloromethanoate) d'ethyle (ou de methyle) en quantites equimoleculaires, on obtient immediatement une solution contenant des anhydrides mixtes de la forme: Si, a une telle solution, on ajoute une solution aqueuse ou organique d'un acide amine, d'un peptide, d'un ester de ceux-ci, ou d'une amine, on obtient sur le champ la formation d'une liaison peptidique, avec degagement de CO2. Grâce a cette methode simple et rapide, nous avons prepare divers dipeptides ainsi qu'un tetrapeptide (a partir de deux dipeptides). Avec l'ammoniac, ces anhydrides mixtes alcoylcarboniques donnent des amides.

Abstract: Publisher Summary This chapter focuses on the accuracy of the peptide theory and on the possible existence of non- peptide bonds in proteins. Partition chromatography in the form of paper chromatography is a method that is identified as breakdown products of proteins where more peptides are used. This technique had been identified by the classical methods of organic chemistry. Three types of polypeptide chains are possible, open, cyclic and branched. There are two types of terminal residues, those with a free amino group and those with a free carboxyl group. The Dinitrophenyl (DNP) method is used for the separation and identification of peptides containing lysine. Elution can be effected with acid ethanol and the peptides subsequently fractionated on suitable partition chromatograms. The theoretical possibility exists that synthetic reactions may occur during the partial hydrolysis of proteins. Acid hydrolysis is the most used method of degrading proteins, and it is almost universally employed when amino acids are to be isolated or estimated, since it leads to complete hydrolysis with a minimum of destruction. Paper chromatography is preferred for the final fractionation of a simplified peptide mixture because of the good resolutions obtained, the ease and rapidity of technique, and the possibility of using the two-dimensional method. The chief disadvantage is that only a very small amount of each peptide can be obtained from a chromatogram, though it is usually possible to obtain sufficient material to determine the structure of di- and tri-peptides.

Abstract: Consden, Gordon and Martin reported a very ingenious method for the separation of amino acids by filter paper chromatography in which phenol or some other appropriate organic solvent sweeps the applied amino acids along the filter paper, capillarity causing the organic solvent to move. The percentage of light intermission along the chromatogram is measured and this transmission has been plotted on semi-log graph paper against the distance along the filter paper strips, and the areas of the segments above the plotted curve measured with a planimeter. It has been found that the areas so determined are over a limited concentration range a simple function of the concentration of the amino acids. The various factors which influence the reliability of this method are reported.
