Effect of corticosterone on adrenal 5-ENE-3β-hydroxysteroid dehydrogenase in estrogen-treated rats

Abstract: Treatment of adult male rats with estradiol-17 beta for 7 days results in adrenal hyperplasia, increased level of serum ACTH along with reduction in serum level of alpha 2u-globulin and inhibition of adrenal 5-ene-3 beta-hydroxysteroid dehydrogenase (5-ene-3 beta-HSD) activity. Administration of corticosterone in estrogen-treated rats reverses the effects of estrogen while in normal rats corticosterone treatment reduces adrenal weight, serum ACTH and adrenal 5-ene-3 beta-HSD activity. In vitro experiments show that alpha 2u-globulin fails to change adrenal 5-ene-3 beta-HSD activity in corticosterone pretreated rats while in normal and estrogen pretreated rats alpha 2u-globulin increases 5-ene-3 beta-HSD activity.

Abstract: The aim of the present study was to determine the role of adrenals in gonadal activity in the male toad during the breeding season. Exogenous administration of corticosterone or metapyrone for 6 days inhibited adrenal Δ 5 -3β(delta 5−3 beta) hydroxysteroid (Δ 5 -3β-HSD) and testicular 17β (17 beta) hydroxysteroid dehydrogenase (17β-HSD) activities, decreased the serum levels of testosterone and inhibited spermatogenesis. When toads were treated with corticosterone a significant rise of serum corticosterone was noted while metapyrone treatment appeared to decrease serum corticosterone levels. It is concluded that adrenocortical hormone plays an indirect role in testicular activity in toads during the breeding season.

Abstract: Administration of estradiol-17 beta for 7 days to the adult male rat results in adrenal hyperplasia, decreased serum corticosterone along with elevation in serum ACTH and inhibition of adrenal 5-ene-3 beta-hydroxysteroid dehydrogenase activity (5-ene-3 beta-HSD). Treatment with alpha 2u-globulin for following 14 days of estrogen-treated rats reversed the effects of estrogen while in normal rats alpha 2u-globulin treatment increased adrenal 5-ene-3 beta-HSD activity and serum corticosterone level while causing a fall in serum ACTH. It is concluded that alpha 2u-globulin may play a role in ACTH secretion by inducing corticosterone synthesis.

Abstract: The mean immunoglobulin levels in 20 normal human sera were found to be 12.4 mg/ml for IgG (7 S γ2-globulins); 2.8 mg/ml for IgA (γ1A or β2A-globulins) and 1.23 mg/ml for the IgM (18 S γ1M-globulins). Type K (I) and Type L (II) immunoglobulins were similarly determined. The results by the antibody-agar plate method are similar to those obtained by the isotopic immune inhibition technique, except for the serum IgA and Type K (I) and Type L (II) immunoglobulin levels. With each of these proteins the values obtained by the isotopic immune inhibition test are higher than those found by the antibody-agar plate test. The basis for this difference is discussed.

Abstract: A radioimmunoassay of plasma ACTH, useful for several mammalian species, has been adapted for the rat. The antibody permitting this versatility was produced in the rabbit by immunization with the species-common, steroidogenic 1–24 amino acid sequence of ACTH. Binding studies, using polypeptide fragments of ACTH, showed that the antibody bound most effectively with the 1–24 fragment and nearly as effectively with the 1–39 native human and porcine molecules. The antibody failed to react significantly with αMSH, βP-MSH or with synthetic 1–16 amide or αp 17–39 fragments of ACTH. A close similarity was found between bioactivity and immunoreactivity in 3 tested specimens of rat plasma. Physiological validation of the method was obtained from the following studies. The immunoreactive plasma ACTH concentration at 9 AM in “resting” male rats was 23±4 pg/ml (mean±SE), when the plasma corticosterone concentration was 4 ± 0.4 μg/100 ml. Abstract: A major urinary protein of the male rat was purified by a combination of (NH 4 ) 2 SO 4 fractionation and chromatography on DEAE-cellulose. The molecular weight, sedimentation coefficient ( s 0 20, w ) and isoelectric point were determined to be 26 400, 2.2.S and pH 3.4, respectively. Hexose, glucosamine and sialic acid were shown tobe 2%, 1.6% and 1.4%. The amino acid composition was also established. The electrophoretic migration was studied in starch-gel and immunoelectrophoretic systems. In the immunoelectrophoretic system, the purified protein migrated similarly to one of the major components of the total urinary proteins previously designated as a α 2 -globulin.
