Publisher Summary The securinega group comprises sixteen alkaloids isolated mainly from two or three species of the Securinega and the Phyllanthus genera. Apart from many investigations establishing their high alkaloid content, studies have shown that some Securinega species contain coumarins, flavonoids, triterpenoids, and other products. This chapter discusses securinine-type alkaloids—their skeletal structure, relative and absolute configuration and conformation, and the piperidyl–cyclohexene acetic acid γ-lactone to isoquinuclidine rearrangement. It describes the NMR spectra of securinine, allosecurinine, and some of their hydrogenated derivatives. The absolute configuration of securinine was confirmed by X-ray analysis on its hydrobromide dihydrate. In the crystal state, it was found that ring A exists in a boat conformation with the nitrogen atom approximately tetrahedrally arranged and its lone pair in a cis relationship with the C-2 hydrogen. The X-ray study also indicated the nonplanar nature of the C-12, C-13 and C-14, C-15 double bonds. The proposed major fragmentation pathways of securinine are obtained by high-resolution mass spectrometry.
