Lysosomal enzyme activity in rats with adjuvant-induced arthritis.

01 May 1970-Annals of the Rheumatic Diseases (Ann Rheum Dis)-Vol. 29, Iss: 3, pp 307-313
TL;DR: It is concluded that the extensive tissue breakdown in adjuvant arthritis is due to the release and degradative action of lysosomal enzymes on connective tissue components.
About: This article is published in Annals of the Rheumatic Diseases. The article was published on 1970-05-01 and is currently open access. It has received 80 citations till now. The article focuses on the topics: Arthritis & Enzyme assay.

TL;DR: It is suggested thatospholipases are freed from the lysosomes of leucocytes overactive in the endocytosis of carrageenin, and that these attack cell membrane phospholipid to give arachidonic acid which is converted to prostaglandin by freely available tissue enzymes.

TL;DR: It is concluded that the extract contains as yet unidentified anti-inflammatory principle(s) and contains alkaloids, saponins, steroids and flavonoids.
Abstract: 1. The anti-inflammatory effect of an alcoholic extract from the flower of Vernonia cinerea (Asteraceae; Less) was tested in adjuvant arthritic rats. 2. Changes in paw volume, body and tissue weights and serum and tissue enzyme activities of ALT, AST, ACP and cathepsin-D in adjuvant rats were reversed by oral administration of 100 mg/kg body weight (BW) of the flower extract. 3. The extract also reversed the major histopathological changes in the hindpaws of the arthritic rats. 4. Phytochemical studies revealed the presence of alkaloids, saponins, steroids and flavonoids. 5. It is concluded that the extract contains as yet unidentified anti-inflammatory principle(s).

Abstract: The pathological and biochemical features of the polyarthritis in rats induced by Freund's adjuvant are briefly reviewed. The object of this is to highlight recent studies that provide a basis for specifically improving procedures employed in screening the activity and mode of action of anti-inflammatory/antiarthritic drugs-especially those with possible disease modifying activity.

TL;DR: This review is mainly concerned with the mechanism of action of those non-steroid anti-inflammatory drugs used to ameliorate the signs and symptoms of human acute and chronic inflammation which have the common property of inhibiting fatty acid oxidative cyclization, in particular the formation of prostaglandins and throm-boxanes.
Abstract: This review is mainly concerned with the mechanism of action of those non-steroid anti-inflammatory drugs used to ameliorate the signs and symptoms of human acute and chronic inflammation which have the common property of inhibiting fatty acid oxidative cyclization, in particular the formation of prostaglandins (PGs) and throm-boxanes. We have retained for this group of substances the term PG synthetase inhibitors because thromboxane synthetase can be selectively inhibited by substances which do not affect PG synthesis and do not show anti-inflammatory activity (Moncada et al., 1976). Other types of non-steroid anti-inflammatory agents are reviewed in subsequent chapters.
