TLDR
Insight gained from studies of the signaling pathways downstream of TCR and BCR stimulation is likely to contribute significantly to future understanding of mechanisms responsible for lymphocyte differentiation and for the discrimination of self from nonself in developing and mature cells.

TRANSCRIPTION FACTORS OF THE NFAT FAMILY:Regulation and Function

TL;DR: Recent data on the diversity of the NFAT family of transcription factors, the regulation of NFAT proteins within cells, and the cooperation ofNFAT proteins with other transcription factors to regulate the expression of inducible genes are discussed.
Journal ArticleDOI

Small GTP-Binding Proteins

TL;DR: In this review, functions of small G proteins and their modes of activation and action are described.

Cellular functions regulated by src family kinases

TL;DR: This chapter reviews the evidence implicating Src family kinases in specific receptor pathways and describes the mechanisms leading to their activation, the targets that interact with these kinases, and the biological events that they regulate.

Three-dimensional segregation of supramolecular activation clusters in T cells

TL;DR: The three-dimensional distribution of receptors and intracellular proteins that cluster at the contacts between T cells and APCs during antigen-specific interactions, Surprisingly, instead of showing uniform oligomerization, these proteins clustered into segregated three- dimensional domains within the cell contacts.

Antioxidant and Redox Regulation of Gene Transcription

TL;DR: The efficacy of different antioxidants to favorably influence the molecular mechanisms implicated in human disease should be a critical determinant of its selection for clinical studies.

Calcineurin is a common target of cyclophilin-cyclosporin A and FKBP-FK506 complexes

TL;DR: The results suggest that calcineurin is involved in a common step associated with T cell receptor and IgE receptor signaling pathways and that cyclophilin and FKBP mediate the actions of CsA and Fk506 by forming drug-dependent complexes with and altering the activity of calcineURin-calmodulin.
Journal ArticleDOI

The mechanism of action of cyclosporin A and FK506

TL;DR: Recent findings that indicate CsA and FK506 operate as prodrugs are reviewed: they bind endogenous intracellular receptors, the immunophilins, and the resulting complex targets the protein phosphatase, calcineurin, to exert the immunosuppressive effect.
Journal ArticleDOI

Identification of calcineurin as a key signalling enzyme in T-lymphocyte activation

TL;DR: It is reported here that overexpression of calcineurin in Jurkat cells renders them more resistant to the effects of CsA and FK506 and augments both NFAT- and NFIL2A-dependent transcription.

Genetic evidence for the involvement of the lck tyrosine kinase in signal transduction through the T cell antigen receptor.

TL;DR: Results indicate that lck is required for normal signal transduction through the TCR, and expression of the lck cDNA in JCaM1 restores the ability of the cell to respond to TCR stimulation.
