Abstract: Plants in the genus Hernandia of the family Hernandiaceae have been used ethnobotanically to treat malaria, postpartum hemorrhage, and morning sickness. Additionally, crude extracts of H. moerenhoutiana and H. voyronii have been reported to exhibit certain antioxidant, antimalarial, cytotoxic, and spasmolytic activities, as well as to inhibit platelet aggregation and prostaglandin biosynthesis. Of the approximately 24 species in this genus, about a quarter have been subjected to detailed phytochemical investigation, namely, H. cordigera, H. guianensis, H. ovigera, H. ovigera var. mascarenensis, H. peltata ( H. nymphaeifolia ), H. sonora, and H. voyronii. From over 130 compounds obtained from these species, the major secondary metabolites are represented by various classes of isoquinoline alkaloids and lignans, along with minor constituents of other alkaloidal (isoindole and pyridine) types, as well as benzenoids, flavonoids, monoterpenoids, and sterols. Some of these isolates have been found to exhibit various biological activities, such as antiplasmodial action, antiplatelet aggregation, cytotoxicity, inhibition of Epstein-Barr virus early antigen activation induced by 12- O -tetradecanoylphorbol 13-acetate, and inhibition of the transformation of JB6 mouse epidermal cells. The biologically active constituents of the genus Hernandia are discussed in detail.
