Abstract: Summary.Uredospores ofPuccinia gramintis (Pers.) tritici (Eriks. andHenn.) wereuniformly labeled with14Cbypermitt.ng thehost(Triticuml aestivum L.)tocarry outphotosynthesis in1"CO2during theprocess ofsporeproduction bytheobligate parasite. Theuseof14Clabeled spores iprovided advantages inastudy oftheutilization ofendogenous substrates atfrequent intervals withsma'l amounts ofspores undercondinions conducive togermination. Because ofprevious uncertainties aboutthenature ofthesubstrates ofimportance to germination, adetailed study ofcarbohydrate andlipid conponents, bothinthespores andinthegermination medium, wasmadeduring thefirst 7 hoursafter placing the -pores on aqueousmedia.D.ethyl etherand80% ethanol soluble metabolites each constituted approximately 20% ofthetotal sporecarbon. Duringthefirst hournearly 60% ofthe80% alcohol solubles disappeared fromthespores whll'e thctotal ethler soluble material didnotchange appreciably. A significant partofthe80% ethanol soluble materials appeared inthegermnation medium. During germination andgermtubeextension, there wasrapid utilization oftrehalose, arabitol andmann.tol eventhough appreciabe amounts ofthese materials werepresent asexogenous pools inthegermnation med'um.Although thetotal amountsofether soluble components didnotchange asdrastically asthecarbohydrate fraction, there was extensive utilization ofpalmitic, oleic, linolenic and9,10-epoxyoctadecanoic acids. Theresults indicate thatthegermination iprocess inspores ofobligate parasites isnot based so!ely ontheutilization oflipids andsomepossible roles ofthechanges ininterna' andexternal pools ofsoluble carbohvdrates arediscussed.
