Abstract: A culture enrichment technique was used to isolate phytase-producing microorganisms. Also, microorganisms from various culture collections were tested for their phytase-producing ability. A number of the Aspergillus niger group produced extracellular phytase which dephosphorylated calcium phytate in acidic solution. A soil isolate, A. ficuum NRRL 3135, produced the most active phytase in a cornstarch-based medium. Production of phytase was strongly repressed by inorganic phosphates and required a high carbon to phosphorus ratio in the medium.

Abstract: The highest yield of protoplasts in the strainAspergillus niger K 10 was obtained from young, freshly harvested hyphae, grown on simple medium of sucrose-asparagine type on a rotary shaker. The residual cultivation medium has to be washed from the mycelial suspension with a solution of high osmotic pressure. Lyophilized snail digestive juice in concentration of 1 %, temperature 31° C, and incubation in Erlenmayer flasks on reciprocal shaker were optimal for the release of protoplasts. Good stabilizers of released protoplasts (in combination with CaCl2) were for example galactose, mannitol, inositol and mixture of NaCl with glucose, sucrose or lactose.

Abstract: Optically active sulphoxides are formed when unsymmetrical thioethers are treated, under aerobic conditions, either with growing Aspergillus niger or with the insoluble material obtained by extracting A. niger with acetone. The stereoselectivity of the oxidation ranges from ca. 4 to 100% depending on the thioether used. t-Butyl p-tolyl sulphide is oxidised most selectively, the R(+)-sulphoxide obtained having a rotation higher than that recorded before. In thioethers of structure, p-X·C6H4·S·CH2Ph, the compound where X = Me is oxidised more selectively than related sulphides where X = H or But.

Abstract: Mycelial mats of Aspergillus niger 1617 grown on cellophane membrane placed on agar media were examined for activities of enzymes related to phosphorus metabolism during the course of cultural development.Most enzymes tested, phosphomonoesterase, pyrophosphatase, ribonuclease, and deoxyribonuclease, showed two pH optima at acid and neutral to slightly alkaline regions except phosphodiesterase, which had an optimum at pH 5.8. In general, enzymes showing higher pH optima were active in younger mycelia and those of lower pH optima were active in aged ones.Ribonucleases of T1-type (optimum pH 6.0) together with T2-type (optimum pH 4.5) were found in younger mycelia, while that of T2-type was most active in aged mycelia. The latter enzyme was considered to be responsible for the ribosome degradation.

Abstract: The respiratory components of tightly coupled mitochondria from the filamentous fungus Aspergillus niger were studied. Cytochromes a + a(3), b, and c + c(1) were identified by difference spectra. The cytochrome spectra were qualitatively similar to yeast and rat liver mitochondria. The mitochondria contained, per gram of protein, an average of 2.9 and 7.0 mumoles of ubiquinone and nicotinamide adenine dinucleotide, respectively.

Abstract: A relatively simple procedure for isolation of mitochondria from Aspergillus niger was described. The organelles satisfied standard criteria for structural and functional integrity and were characterized by a somewhat higher degree of respiratory control than that associated with mitochondria from fungi reported by others. Mitochondria from a nitroaryl-resistant strain respired more vigorously and had higher respiratory control ratios than mitochondria from the parent wild strain. These observations may explain previous demonstrations that the former strain had high citrate cycle activity and no measurable direct oxidative pathway, whereas the wild strain metabolized glucose predominantly via gluconate and had very little citrate cycle capability. Abstract: SUMMARY: The rates of growth of five strains of Aspergillus niger (M, MUBLI, NRRL 334, NRRL 323, NRRL 346) were followed in an optimum nutrient solution. The time course of growth of all the strains showed a steep decrease after 3 days, except that of strain NRRL 346 which tended to flatten out. A correlation between sporulation and mycelial dry weight was observed. All the five strains showed a marked decrease in mycelial yield in the absence of manganese from the basal medium. Their quantitative responses to graded concentrations of Mn in the medium were compared with spore and mycelial inocula. Statistical analyses of the result suggest that strain MUBLI is the most suitable test organism for the bioassay of Mn. The effective range of concentrations of Mn which coincided with the maximum differences in mycelial dry weights was 0.001–0.006 µg./ml.

Abstract: Effect of digestion with various cellulase preparations including Vercellase from Aspergillus niger was examined on the extraction of green tea components from tea leaves. By the treatment, especially by that with Vercellase, significant improvement in yield of solubles such as caffeine and tannin was obtained without any unfavorable effect on the flavor. Such treatment will be available to manufacture instant green tea.

Weiterhin konnte gefunden werden, das die Saccharoseaufnahme gleichmasig uber die Hyphen junger Pellets erfolgt. Eine verstarkte radioaktive Markierung der Hyphenspitze nach langerer Inkubation ist die Folge des Spitzenwachstums. Altes Mycel verhalt sich anders. Hier sind Orte intensiven Stoffwechsels proliferierende Conidientrager.

The pH value of the culture media differs from strain to strain for the good yield of citric acid. When there is good yield of citric acid the sugar consumption is less and mycelial growth is also less.
