Abstract: Acid catalysed condensations of various 2 substituted 1,3-oxazinanes 3 and 1,3-oxazolidines 4 with cyclic carbon nucleophiles viz. 5,5-dimethyl-1,3-cyclohexanedione and 1,3-cyclohexanedione furnish xanthene derivatives, whereas a Knoevenagel reaction proceeds with acyclic nucleophiles. In case of 4b and 4c, a unique synthesis of functionalised α-tetralones has emerged. Reactions of mixtures of cyclic and acyclic carbon nucleophiles with 3 provide some functionalised and partially reduced benzopyran derivatives.

Abstract: In this study, the practical construction of a pilot library with benzopyranylpyrazole, a novel core skeleton synthesized through the recombination of privileged structures, benzopyran and pyrazole, was successfully conducted through the efficient utilization of solution-phase parallel synthesis using solid-phase reagents and solid-phase parallel synthesis. We have also developed a novel procedure for the synthesis of benzopyranylpyrazoles via regioselective condensation of substituted hydrazines with β-keto aldehydes. The diversity of this core skeleton was expanded by the regioselective introduction of alkyl- and aryl-substituents at the R1 diversity point on the pyrazole moiety and by the introduction of piperazine on the benzopyran substructure, which provide the R2 diversity point. Lastly, the introduction of a nitro group on the benzopyran moiety was found to accelerate the nucleophilic aromatic substitution of piperazine and provide the R3 diversity point at the aniline moiety through the reduction...

Abstract: The mass spectrometric behaviour of some 2-hydroxymethyl-3,4,5,6,7,8-hexahydro-2H-1-benzopyran-5-ones is described and discussed with the aid of exact mass measurements, linked scans, labelling experiments and low electron energy measurements. The unusual presence of abundant [MH3O]+ species in the mass spectra of 4-phenyl substituted compounds is discussed in detail and a mechanism is proposed.
