TL;DR: Tricyclic compounds containing a cyclopropyl carboxylic acid (CARA) or carboxyl acid derivative fused to a bicyclic ring, including pharmaceutically acceptable salts and prodrugs thereof, are useful as therapeutic compounds, particularly in the treatment of Type 2 diabetes mellitus, and of conditions that are often associated with this disease, including obesity and lipid disorders, such as mixed or diabetic dyslipidemia, hyperlipidemic, hypercholesterolemia, and hypertriglyceridemia.
Abstract: Tricyclic compounds containing a cyclopropyl carboxylic acid or carboxylic acid derivative (e.g. amide) fused to a bicyclic ring, including pharmaceutically acceptable salts and prodrugs thereof, are agonists of G-protein coupled receptor 40 (GPR40) and are useful as therapeutic compounds, particularly in the treatment of Type 2 diabetes mellitus, and of conditions that are often associated with this disease, including obesity and lipid disorders, such as mixed or diabetic dyslipidemia, hyperlipidemia, hypercholesterolemia, and hypertriglyceridemia.

Abstract: 132 CBI analogues of CC-1 065 and the duocarmycins having dimeric monocyclic, bicyclic, and tricyclic heteroaromatics substituents were synthesized by a parallel route. The resultant analogues were evaluated with respect to their catalytic and cytotoxic activities. The relative contribution of the various dimeric monocyclic, bicyclic, and tricyclic heteroaromatics substituents within the DNA binding domain were characterized. Several of the resultant CBI analogues of CC-1065 and the duocarmycins were characterized as having enhanced catalytic and cytotoxic activities and were identified as having utility as anti-cancer agents.

Abstract: Herein we describe a rhodium-catalyzed (4+1) cyclization between cyclobutanones and allenes, which provides a distinct [4.2.1]-bicyclic skeleton containing two quaternary carbon centers. The reaction involves C–C activation of cyclobutanones and employs allenes as a one-carbon unit. A variety of functional groups can be tolerated, and a diverse range of polycyclic scaffolds can be accessed. Excellent enantioselectivity can be obtained, which is enabled by a TADDOL-derived phosphoramidite ligand. The bridged bicyclic products can be further functionalized or derivatized though simple transformations.
