Abstract: A simple, rapid, highly sensitive, and selective method for detecting protein in biological samples has been developed, which is based on the interaction between protein and m‐nitrophenylfluorone‐Mo(VI) complex as a spectral probe. The optimum condition for the reaction is investigated. Bovine serum albumin (BSA) obeys Beer's law up to 10 µg · mL−1, having a molar absorption coefficient of 8.51×106 L · mol−1 · cm−1 at 535 nm. Many amino acids and metal ions do not interfere. The results of determination for biological samples are comparable to those obtained by the Bradford method. Meanwhile, the binding number is also determined.

Abstract: Plasma (100.00 ml) obtained from bovine blood by centrifugation was fractionated into seven precipitates by cold ethanol precipitation. The yield (amount) of proteins in the precipitates calculated from the standard curve of BSA was 1160.83 mg, 806.57 mg, 1149.94 mg, 8.79 mg, 19.88 mg, 21.98 mg, and 13.97 mg respectively, while the total amount of protein obtained was 3180.00 mg. The precipitates were fractionated into 25 fractions each by Anion Exchange Chromatography (AEC) and the amount of protein in each fraction was obtained by Bradford protein assay. SDS-PAGE analysis was performed on the fractions with proteins and their estimated molecular weights were obtained with the aid of the molecular weight marker. The result indicated that cold ethanol precipitation and anion exchange chromatographic techniques are valuable tools for the purification of bovine blood to obtain high grade α, β and γ-fibrinogen, IgM (μ-globulin), IgG (γ -globulin), alpha (α –globulin), β-globulin (E-globulin) and albumin. Abstract: A qualitative and/or quantitative proportioning of proteins of the solution (has the ratio 4/1-2/1) is obtained by contacting the solution with a compound capable of reacting with proteins and carrying out interaction to colorimetric mode is by chemical, physical, physicochemical or immunological method. The reducer (0.04-0.1 M) is 2-mercaptoethanol, dithiothreitol (DTT) and/or thioglycolic acid. The fibers and the solution are contacted at a rate of 10 mg and heated at higher/equal to 100[deg]C. Independent claims are also included for: (1) a kit; and (2) a composition using the above method.
