Abstract: Objective:To optimize processing technology parameters of Strychnos nux-vomica heated with sand,in order to control quality of S.nux-vomica effectively.Method:With the content of brucine and strychnine as indexes,orthogonal test was used to evaluate effect of oil-sand degree,sand-material ratio,frying temperature and time on processing technology parameters of S.nux-vomica.Result:Optimum processing technology parameters were as followings:medium sand(300-600 μm),sand-material ratio 7∶ 1,frying temperature(190±5) ℃,frying time 4 min.Conclusion:Optimized processing technology was suitable and feasible,it was suitable for mass production.

Abstract: 3-Bromo-2,2′,4,4′,6,6′-hexamethyl-3′-nitro-, 2,2′4,5′,6-pentamethyl-3-nitro-, 3-bromo-2,2′,4,6-tetramethyl-5′-nitro-, and 2,2′,4,4′,6,6′-hexamethyl-3-methoxycarbonylmethyl-3′-nitro-benzophenone were prepared by Friedel–Crafts acylation. The last compound was hydrolysed to 3-carboxymethyl-2,2′,4,4′,6,6′-hexamethyl-3′-nitrobenzophenone, which was resolved by way of the brucine salt; this demonstrates for the first time the existence of optical isomerism in suitably substituted benzophenones because of steric interactions between the o- and o′-substituents.

Abstract: Objective:To establish the RP-HPLC method for the content determination of brucine and strychnine in Yaotongning Patch.Method: The Intersil C8-3(4.6 mm×250 mm,5 μm) column was used;the mobile phase consisted of methanol-water-acetic acid(20∶96∶7),adjust pH to 3.10 with triethylamine.The flow rate was 1.0 mL·min-1 and the detection wavelength was at 254 nm;the column temperature was at 30 ℃.Result: The linear range of strychnine was between 0.020 56-0.123 36 μg(r=0.999 8),the average recovery was 100.28% with a RSD of 1.33%(n=9);the linear range of brucine was between 0.013 84-0.083 04 μg(r=0.999 9);the average recovery was 99.82% with a RSD of 0.84%(n=9).Conclusion: The method is fast,reliable and accurate,it can be used in the quality control of strychnine and brucine in Yaotongning Patch.

Abstract: OBJECTIVE To study the transdermal absorption characteristics of brucine in vitro. METHODS The oil-water partition coefficient of brucine was measured. Drug permeation tests were performed through excised rat skin in improved Franz diffusion cell. Brucine concentration in samples was determined by HPLC. RESULTS The oil-water partition coefficient of brucine was between 4.05 and 5.02 at different temperatures. The permeation rate of 0.5 and 1 mg/ml brucine solution were (1.83 +/- 0.85) and (3.74 +/- 1.54) microg/cm2 x h, respectively. The cumulative permeation ratio in 24 hours were (51.30 +/- 18.51)% and (50.01 +/- 12. 80)%, respectively. CONCLUSION The research provides experimental datas for the design of transdermal delivery system of brucine.
