Abstract: Objective To establish the method to determine the content and encapsulation efficiency of liposomes containing total alkaloids from seed of Strychnos nux-vomica L.Methods The total alkaloids were extracted from seeds of Strychnou nux-vomica L.and the content was analyzed.Liposomes containing the total alkaloids were prepared by ammonium sulfate transmembrane gradients.The total alkaloid content of dissolved liposomes was determined by UV spectrophotometry at 254 nm,which was performed with strychnine as the reference substances.The encapsulated and free drugs were separated with Sephadex G-50 column,and then their contents were determined to calculate the encapsulation efficiency.Results The average extracted efficiency of total alkaloids from seed of Strychnos nux-vomica L.was 3.49 %.Strychnine and brucine accounted for 46.76 % and 26.51 % in the extract,respectively.Strychnine at the concentration of 2.4-20 μ g/mL had a good linear relationship with the absorbance.The recovery of strychnine was in the range from 96.66 % to 102.62 %.The recovery of Sephadex G-50 column was(100.85± 0.79) %(n=3).Conclusion The established method is simple,accurate and stable,and can be used for the quality control of liposomes containing total alkaloids from seed of Strychnos nux-vomica L.

Abstract: Objective To study the anagesic effect of compound brucine and its pharmacodynamics. Methods The mice were treated ip only once with compound brucine (32.6, 22.8, 16 mg/kg) and morphine (10 mg/kg) as positive control. Then the change of the latencies of licking paws on hot-plate were observed. The dose-effect and time-effect curves were studied. The apparent parameters of pharmacodynamics were estimated based on time-effect curves of compound brucine 32.6 mg/kg ip. Results Compound brucine can prolong the incubation period of pain caused by hot-plate in mice obviously. A significant dose-effect relationship was noted in the anagesic experenment. The potency of this drug (22.8, 16 mg/kg) ip was equivalent to that of the morphine 10 mg/kg ip. The time-effect curve after compound brucine 32.6 mg/kg ip was found to fit a 1-compartment open model with first order elimination. The apparent pharmacodynamic parameters were: Ka=0.09477/min, T1/2a=7.3 min, Ke=0.00203/min, T1/2e=341.4 min, Tb=1.3 min, Tmax=41.4 min, Emax=238.5, Te=598.7 min, Tc=597.4 min, respectively. Conclusion Compound brucine has remarkable analgesic effect with power potency and long duration.

Abstract: The inhibiting mechanisms of brucine to cancer cells included inducing the apoptosis of cancer cells, inhibiting the proliferation of cancer cells, preventing cell adhesion and transfer. This paper reviews the function of brucine and its anti-tumor preparations, and compares the efficacy and adverse reactions between brucine related preparations and traditional drugs to evaluate its anti-tumor effects. Furthermore, more exploration on the inhibiting mechanisms is necessary for the patients to obtain safe and effective drugs. Abstract: Objective To develop an RPHPLC method for determination of strychnine and brucine in Semen Strychni and its processed products and study the principle of Semen Strychni processed products with Jiangxi innovated method.Methods A Hypersil BDS C_(18)(4.6 mm×200 mm,5 μm) column was used with acetonitrile-water-diethylamine(34∶100∶1) as the mobile phase,the content was detected at a wavelength of 260 nm.Results The contents of strychnine and brucine in the processed products with Jinngxi innovated method were lower.Conclusion This method is accurate,simple and reliable.
