Abstract: The characteristic color-forming responses of lignified tissue and some lignin preparations on treatment with certain organic and inorganic reagents were recognized as early as the 19th century when Runge (1834) noted that spruce wood turned greenish blue after treatment with phenol and hydrochloric acid. Color reactions have since found widespread use in the detection and identification of lignin and several have also been adapted for use in the quantitative determination of lignin. Recently, color reactions have been proposed for the detection of specific groups within the lignin molecule.
