Abstract: The plastic monomere bisphenol A (BPA) and the flame retardant tetrabromobisphenol A (TBBPA) were examined for estrogen-like developmental effects on the reproductive organs in avian embryos. The synthetic estrogen diethylstilbestrol (DES) was used as a positive control. The test compounds were injected into the yolk of quail and chicken eggs early during incubation and the embryos were examined 2 d before anticipated hatching. At 200 microgram/g egg, BPA induced Mullerian duct (embryonic oviduct) malformation in female quail embryos and feminization of the left testis (ovotestis) in male chicken embryos. The estrogenic potency of BPA compared with DES was species and endpoint specific. Mullerian duct malformation was the most sensitive endpoint in quail embryos, whereas ovotestis formation was the most sensitive response in chicken embryos. Tetrabromobisphenol A caused high embryo mortality at 45 microgram/g egg in both species, but no estrogen-like effects were observed. Bisphenol A caused mortality only in chicken embryos at 67 and 200 microgram/g egg. To our knowledge, this is the first report on estrogen-like or embryolethal effects of BPA and TBBPA in birds.

Abstract: The aim of this study was to perform a morphometric analysis of the various parts of the intestine of the domestic quail. Twenty-four female quails (Coturnix coturnix japonica) aged 37 weeks were used and accommodated in laying cages for 12 weeks. Each group was fed a standardized diet containing different quantities of calcium: 2.0%, 2.5%, 3.0% and 3.5%. The birds were weighed, killed, and samples of 1 cm were collected from the duodenum, jejunum and ileum and submitted to the histological routine. The sections obtained were stained in haematoxylin & cosin (H&E). For morphometric analysis, 30 villi and 30 crypts of each segment of the small intestine were measured in order to determine the height and area of the villi, as well as the depth of the crypts. The results showed that although the integrity of the gastrointestinal tract was maintained in all the birds treated with the different calcium levels, a calcium level of 3.0% showed the most promise. as the levels of 2.0% and 2.5% did not cause any alteration in the intestinal tract. Furthermore, a calcium level of 3.5% led to a significant reduction in the height of the villosities, and in consequence reduced the digestive and absorptive capabilities.

Abstract: Experiments were conducted to determine the effect of injection of lipopolysaccharide (LPS, from S. typhimurium) or muramyl dipeptide (MDP, N-acetylmuramyl-L-ala-isoglutamine) in Japanese quail. Doses of MDP between 0.3 and 10 mg/kg body wt. had no effect on body temperature. In contrast, doses of 1.0-22.5 mg LPS/kg body wt. caused significant increases in body temperature. None of the doses of LPS or MDP resulted in mortality. The febrile response to LPS was diminished following a second injection 48 h after the first, and was absent following a third injection. Plasma zinc, an indicator of the acute phase response, was significantly reduced by either LPS or MDP after the first injection (P<0.001), but not after the second or third injection. Splenic interleukin 1-beta (IL-1beta) mRNA expression was increased after the first and last injection of LPS (P<0.001), but only after the first injection of MDP (P<0.005). Hepatic IL-1beta mRNA expression was increased after the first, but not the third injection of LPS (P<0.001), while MDP had no effect. These data indicate that Japanese quail are less sensitive to MDP than LPS, and that quail demonstrate tolerance to LPS following repeated injections.

Abstract: 1. HG and LG quail lines selected for high and low relative weight gain between 11 and 28 d of age (RG11-28), respectively, and an unselected C line were compared. Mature body weight of both selected lines was held at that of the C line. Progeny of generation 6 were used for analysis. 2. Divergent selection for RG11-28 brought about opposite changes in the growth rates shortly after hatching. 3. Parameters of the Richards function were used to describe the growth curve. The largest differences between HG and LG lines occurred in age (t+) and body weight (y+) at the inflection point of the growth curve (on average for both sexes 28% and 20%, respectively). For HG quail, the parameter t+ was 5 d later than that for LG quail (18.6 vs 14.1 d for males and 20.6 vs 15.6 d for females, respectively), and consequently the parameter y+ was greater (90.3 vs 84.0 g for males and 104.5 vs 96.1 g for females, respectively). The shape of the growth curve expressed by the y+/A ratio was substantialy different for HG and LG quail (44.8% vs 39.6% for males and 43.5% vs 36.8% for females, respectively). 4. The food/gain ratios for the fattening period (3 to 35 d of age) were 3.21, 3.47 and 3.34 for the HG, LG and C lines, respectively. The HG quail started to utilise food more efficiently than the LG quail as early as 10 to 14 d, that is, at the age when their relative growth rate first became greater. 5. The relative deviations of the HG and LG lines from the C line are discussed.

Abstract: Sexual interactions can cause changes in plasma hormone levels and activate immediate early genes within the mammalian brain. There are marked anatomical differences between the regions activated that relate directly to the sexual specific behaviour and neuroendocrinology of each sex. The aim of this study was to determine if such a sexual dimorphism exists in birds by examining the brain regions stimulated in adult virgin female Japanese quail (Coturnix japonica) during sexual behaviour and comparing this to previously reported data concerning males. Female quail were allowed to freely interact with adult males and both female and male sexual behaviour was recorded. Contrary to previous findings in male quail, no significant induction of Fos-like immunoreactive (FLI) cells was observed following sexual interactions in the preoptic area of females; this area is fundamentally involved in the control of male-type copulatory behaviour. Sexual interactions significantly induced FLI cells in the hyperstriatum ventrale, the part of the archistriatum just lateral to the anterior commissure, and the nucleus intercollicularis. Moreover, prominent activation was detected throughout most of the ventromedial nucleus of the hypothalamus, a region reported to be rich in oestrogen receptors. FLI induction was not a consequence of sexual behaviour induced changes in luteinizing hormone (LH) as plasma LH levels were unaltered. Instead, brain activation must be a consequence of copulation-associated somatosensory inputs or direct stimuli originating from the male. Male quail, like the majority of other birds, lack an intromittant organ (penis) so that the somatosensory inputs to the female are rather different from those in mammals; the precise nature of these inputs is yet to be determined.

Abstract: The insulin-like growth factor (IGF) system is regulated by various stimuli, including hormones, growth factors and nutritional status. We examined the effects of vitamin A on components of the IGF system in Japanese quail. Male quail (1 d old) fed a vitamin A-deficient diet for 14 or 21 d developed vitamin A deficiency, as confirmed by a depletion of serum retinol and hepatic retinyl palmitate. Consuming the vitamin A-deficient diet for 14 d did not affect growth rate, but decreased the serum IGF-I concentrations by 22% compared with the control group. The decreased serum IGF-I levels were accompanied by 21-52% lower levels of IGF-I mRNA in the testis, lung, liver and heart, whereas IGF-I receptor (IGF-IR) and insulin receptor (IR) gene expressions were unaffected in these tissues. Continuous feeding of the vitamin A-deficient diet for 21 d retarded growth and further decreased the levels of serum IGF-I and tissue IGF-I mRNA. Serum IGF-I levels were reduced by approximately 50%; IGF-I mRNA levels were > 90% lower in the liver and lung and approximately 60% lower in the heart and testis. In contrast, levels of the IGF-IR and IR mRNAs were approximately 100% greater in some tissues examined. When vitamin A-deficient quail received a single injection of retinol or retinoic acid (0.1 mg/bird), tissue IGF-I, IGF-IR and IR gene expressions did not change after 4 h. These results suggest a possible physiologic role of the IGF system in mediating vitamin A-supported growth of Japanese quail.

Abstract: A sexually dimorphic nucleus is located in the preoptic area of Japanese quail and plays a key role in the activation of male copulatory behavior. The medial preoptic nucleus (POM) is significantly larger in adult male than in adult female quail. Its volume is steroid-sensitive in adulthood and consequently decreases after castration but is restored to normal levels by a treatment with exogenous testosterone. This volumetric difference appears to result only from a sex difference in the adult hormonal milieu and is not affected by embryonic treatments that permanently modify sexual behavior (no organizational effects). In contrast, some cytoarchitectonic features of the POM such as the size of neurons in the dorso-lateral part of nucleus appear to be irreversibly affected by embryonic steroids. The POM is characterized by the presence of a wide variety of neurotransmitters, neuropeptides, and receptors and can be specifically identified by the presence of a dense cluster of aromatase-immunoreactive cells, by a high density of neurotensin-immunoreactive cells and fibers and by a dense vasotocinergic innervation. Some of these neurochemical markers of the dimorphic nucleus are themselves modulated by steroids. Many of these neurochemical changes appear to play a causal role in the control of male sexual behavior. The quail POM thus represents an excellent model for the analysis of steroid-induced brain plasticity in a behaviorally relevant context.

Abstract: Summary Insulin-like growth factor-I (IGF-I) is involved in the regulation of growth and differentiation of a variety of vertebrate tissues. The biological actions of IGF-I are mediated mainly by the IGF-I receptor (IGF-IR) and partly by the insulin receptor (IR) and modulated by IGF binding proteins (IGFBP). We conducted studies designed to clarify the possible roles of IGF system in the development of the avian reproductive organs. We cloned cDNAs of IGF-I, IGF-IR, IR and IGFBP-2 of Japanese quail and simultaneously measured the expression of these genes in the quail liver, testis and oviduct at different ages using a lysate RNase protection assay. Hepatic IGF-I mRNA levels increased rapidly and remained elevated during the rapid-growing period, which coincided with the period of rapid increase in testicular weight. IGF-I mRNA was detected at each stage of developing testis examined. Its level was high at the early stage and decreased with age. IGFBP-2 mRNA in testis exhibited a similar expression pattern to that of IGF-I, whereas a divergence in IGF-I and IGF-IR gene expression was observed. Both IGF-IR and IR mRNAs increased when the testis grew rapidly and decreased when sexual maturation was almost completed. These results suggest that IGF-I may serve as an autocrine/paracrine regulator as well as an endocrine regulator in the testicular development and function of Japanese quail. In the oviduct, IGF-I, IGF-IR, IR and IGFBP-2 mRNAs were also developmentally regulated. A rapid growth of the oviduct was accompanied by a significant increase in the level of IGF-I mRNA. The expression of genes encoding IGF-IR, IR and IGFBP-2 in the oviduct exhibited a similar developmental change to that of IGF-I. These results suggest that IGF-I mainly works in an autocrine and/or paracrine manner in the oviduct during the development of this organ. The findings of the present study provide further evidence of an important role for IGF system in the development and function of the avian reproductive system.

Abstract: Hydroxyindole-O-methyltrasferase (HIOMT) plays an important role as the final enzyme in the synthesis of melatonin. In this study, the expression of the HIOMT gene in Japanese quail was investigated with respect to tissue distribution and the effects of light and vitamin A deficiency. HIOMT mRNA in the pineal gland and eye had a clear daily rhythm with peak values in daytime. The testis also contained a detectable amount of HIOMT mRNA, which did not display a rhythmic change over a 24-h period. When birds were rendered vitamin A deficient through feeding with a vitamin A-free diet, the daily rhythm of the HIOMT gene almost disappeared in both the pineal gland and eye due to increases in the nighttime values. Our previous observations and these results suggest that vitamin A and a photo-signal are required to maintain the rhythmic expression of the HIOMT gene as well as the arylalkylamine N-acetyltransferase gene.

Abstract: We investigated the effects of vitamin A (VA) nutritional status on the levels of expression of retinoic acid (RA) receptor-beta (RARbeta) gene in the various tissues of Japanese quail. VA deficiency caused a significant decrease in the mRNA levels of brain, liver, heart, lung and kidney RARbeta2/beta4, whereas no change was observed in the level of testis RARbeta2 transcript. In contrast, reduction in the RARbeta1 transcript caused by VA depletion was observed only in the lung, remaining unchanged in the other tissues. The administration of RA to the VA-deficient quail rapidly induced the expression of RARbeta2/beta4 mRNAs in all the tissues examined, but RA increased the expression of RARbeta1 transcript in the liver, heart, lung and kidney at a lower magnitude. RA could not change the expression of the brain RARbeta1 transcript, while it induced the expression of the testis RARbeta1 mRNA in a temporal way. These results clearly indicate that VA nutritional status differently regulates the expression of RARbeta1 and RARbeta2/beta4 transcripts in a tissue-specific manner.

Abstract: We investigated the effect of day length on mixed protein fractional synthesis rates (K-S) in 14- and 21-d-old Japanese quail (Coturnix c. japonica) habituated to either a long day length, 18 h light/6 h dark (LDL), or short day length, 6 h light/18 h dark (SDL), with free access to food during the light period. Rates of protein synthesis were measured by a flooding dose of L-[1-C-13]leucine. In both groups, we measured K-S of pectoral muscle, liver and heart after an overnight period of food deprivation and after 2-h food access at dawn. Rates of protein synthesis were also measured in LDL quail starved for 18 h and refed for 2 h. SDL chicks were smaller and had lower tissue weights at 2 wk of age than did LDL chicks (P <0.05). Starvation led to a lower rate of protein synthesis in those animals starved for 18 h. Food availability after starvation for 18 h induced a significant rise in tissue protein synthesis in both SDL and LDL quail (P <0.05). This increase was absent in LDL quail after a 6-h starvation period. There was an increase in K-S to ad hoc changes in food supply. By determining the daily period in which feeding can occur, day length has a major effect on protein synthesis rates. This effect will determine the overall growth chicks are able to achieve that have been subjected to different day lengths.
