Abstract: Carbonic anhydrase activity was studied in the quail metanephros by means of histochemical, histophotometrical and biochemical methods. Male and female samples were examined separately in order to show sex-related differences in enzyme activity and localization. The staining patterns revealed differential distribution of reaction product in the different tubular segments. The initial portion of proximal tubules showed positivity on the brush border in female kidneys only. Extra situ investigations provided further evidence of sexual dimorphism resulting in higher values of enzyme activity for female than for male kidneys. In both sexes, marked staining was detected at the distal tubule level where histophotometric analysis confirmed the highest amount of reaction product. Moreover, the intracellular staining distribution at this site proved to be similar to that observed for mammalian proximal convoluted tubules. In the collecting ducts, a mosaic-like pattern was found with respect to both carbonic anhydrase staining and metachromatic properties. The functional significance of the presence of enzyme in the different renal tubules is discussed by comparison with the mammalian kidney. A model is proposed whereby the distal tubules represent the main sites of urinary acidification and bicarbonate reabsorption.

Abstract: The effects of embryonic exposure to androgen disrupting chemicals (ADCs) on growth and fluctuating asymmetry (FA) were determined in Japanese quail chicks. Embryos were exposed to an anti-androgenic chemical, 1,1,1-Trichloro-2,2-bis(p-chlorophenyl)ethane (p,p′-DDE) at 20 or 40 μg , or to an androgenic chemical, trenbolone acetate, at 5 or 50 μg on day one of incubation. Growth was measured by body weight and tarsus and culmen lengths from day of hatch until day 29. FA was measured as differences in right versus left lengths of the tarsus, radius, zygomatic process, and premaxilla in day old carcasses. No differences in FA were observed for either treatment. Embryonic exposure to DDE resulted in no significant differences in all measures of growth, although the same quail exhibited significant differences in immunological, reproductive, and behavioral measurements (reported elsewhere). Chicks exposed to trenbolone exhibited no differences in body weight or measures of FA at day of hatch, however, subsequent growth was inhibited. This study shows that although growth and FA are often used as measures of chemical stress experienced during embryonic development, they are not sensitive measures for exposure to these ADCs at these levels in Japanese quail.

Abstract: We established an endodermal epithelial cell culture model (EEC) for studying the function of certain enzymes and proteins in mediating nutrient utilization by avian embryos during development. Fertilized Japanese quail eggs were incubated at 37 °C for 5 days and then yolk sac membranes (YSM) were collected to establish the EEC culture system. We isolated the embryonic endoderm layer from YSM, and sliced the membrane into 2 - 3 mm pieces and partially digested with collagenase before seeding in 24-well culture plates. The EECs proliferate out of the tissue and are ready for cell culture studies. We found that the EECs had typical characteristics of YSM in vivo, for example, accumulation of lipid droplets, expression of sterol O-acyltransferase and lipoprotein lipase. The partial digestion treatment significantly increased the successful rate of EEC culture. Utilizing the EECs, we demonstrated that the expression of SOAT1 was regulated by the cAMP dependent protein kinase A related pathway. This primary Japanese quail EEC culture system is a useful tool to study embryonic lipid transportation and to clarify the role of genes involved in mediating nutrient utilization in YSM during avian embryonic development.
