Abstract: This paper overviews the recent work on compressive sensing, a new approach to data acquisition in which analog signals are digitized for processing not via uniform sampling but via measurements using more general, even random, test functions. In stark contrast with conventional wisdom, the new theory asserts that one can combine "low-rate sampling" with digital computational power for efficient and accurate signal acquisition. Compressive sensing systems directly translate analog data into a compressed digital form; all we need to do is "decompress" the measured data through an optimization on a digital computer. The implications of compressive sensing are promising for many applications and enable the design of new kinds of analog-to-digital converters, cameras, and imaging systems.

Abstract: We report here on the current state of our efforts in automated molecular microscopy. Our primary automated data acquisition software system, Leginon, has been completely redesigned over the past two years. The new distributed system has been developed using the Python programming language and is compatible with both Linux and Windows operating systems. The new flexible architecture was designed to allow for the development of customized data collection protocols, several of which are described here. The system has been used to acquire data for ∼150 experiments and we have demonstrated the capacity for high throughput data acquisition by acquiring images of more than 100 000 particles in a single session at the microscope.

Abstract: Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) produces complex, time-dependent signals. These require significantly different treatment both during data acquisition and reduction from the more steady-state signals produced by solution sample introduction. This paper discusses, in detail, data acquisition and reduction considerations in LA-ICP-MS analysis. Optimum data acquisition parameters are suggested. Equations are derived for the calculation of sample concentrations and LOD when time-resolved data acquisition is employed, sensitivity calibration is obtained from reference materials with known analyte concentrations and naturally occurring internal standards are used to correct for the multiplicative correction factors of drift, matrix effects and the amount of material ablated and transported to the ICP.
