Abstract: Overexpression of acyl-CoA binding protein (ACBP) was induced in a rat hepatoma cell line (McA-RH 7777) by stable integration of rat ACBP cDNA. The transfected cells (ACBP-27) had 3.5-fold higher concentrations of ACBP than control cells (14 vs. 4 ng/μg DNA). Both ACBP-27 and control cells were cultured in the presence of various concentrations of radiolabeled palmitic acid; and the effects of ACBP on lipogenesis and β-oxidation were studied. Incubation of the cells with 100 μM palmitic acid resulted in 42% greater incorporation of the fatty acid in ACBP-27 cells as compared to that in the control cells. This increased incorporation of the fatty acid was observed predominanly in the triglyceride fraction. Higher concentrations of palmitic acid (200 to 400 μM) were associated with a significant decrease in the production of 14CO2 in the ACBP-27 cell line than in the control cells, while lower concentrations had not effect. Our data suggest a role for ACBP in the partitioning of fatty acids between esterification reactions leading to the formation of neutral lipids and β-oxidation. ACBP may play a regulatory role by influencing this important branch point in intermediary lipid metabolism.

Abstract: A computer model is presented in which the role of cytoplasmic fatty acid-binding protein (FABP) in the intracellular translocation of fatty acids (FA) from one membrane to an opposite membrane is studied. The model consists of a cubical space, in which FABP and FA are allowed to diffuse at random. The amount of FA released from the donor membrane and reaching an opposite acceptor membrane is calculated in a variety of conditions. The data provided by the various simulations suggest that FABP can play a significant role in intracellular FA transport only if FABP is able to take up FA directly from FA containing membranes and to directly deliver FA to an acceptor membrane, thus preventing the unfavourable thermodynamical situation in which FA must solubilize in an aqueous environment prior to binding to FABP.

Abstract: Fatty acid binding protein 7 (FABP7), abundant in the embryonic brain, binds with the highest affinity to docosahexaenoic acid (DHA) and is expressed in the early stages of embryogenesis. Here, we have examined the consequences of the exposure to different DHA levels and of the in utero depletion of FABP7 on early rat brain development. Neurodevelopment was evaluated through the contents of two proteins, connexin 43 (Cx43) and cyclin-dependent kinase 5 (CDK5), both involved in neuroblast proliferation, differentiation, and migration. The dams were fed with diets presenting different DHA contents, from deficiency to supplementation. DHA brain embryos contents already differed at embryonic day 11.5 and the differences kept increasing with time. Cx43 and CDK5 contents were positively associated with the brain DHA levels. When FABP7 was depleted in vivo by injections of siRNA in the telencephalon, the enhancement of the contents of both proteins was lost in supplemented animals, but FABP7 depletion did not modify phospholipid compositions regardless of the diets. Thus, FABP7 is a necessary mediator of the effect of DHA on these proteins synthesis, but its role in DHA uptake is not critical, although FABP7 is localized in phospholipid-rich areas. Our study shows that high contents of DHA associated with FABP7 are necessary to promote early brain development, which prompted us to recommend DHA supplementation early in pregnancy. © 2015 Wiley Periodicals, Inc.
