Abstract: A protocol is described for rapid and large-scale propagation of the woody aromatic and medicinal shrub Vitex negundo by in vitro culture of nodal segments from mature plants. Of the three different cytokinins – N6-benzyladenine (BA), kinetin, and thidiazuron – evaluated as supplements to Murashige and Skoog (MS) medium, BA at an optimal concentration of 2.0 mg/l was most effective in inducing bud break. Although callus-free multiple-shoot formation was a function of cytokinin activity alone, faster bud break coupled with an enhanced frequency of shoot development (92%) and internode elongation were dependent on the synergistic influence of gibberellic acid (GA3) when used at an optimal concentration (0.4 mg/l) along with BA (2.0 mg/l). The frequency of shoot proliferation was markedly influenced by the explanting season. By repeated subculturing of nodal segments harvested from the in vitro-formed axenic shoots on MS containing 1.0 mg/l BA and 0.4 mg/l GA3, prolific shoot cultures free from proximal callusing and showing a high-frequency multiplication rate were established. The percentage shoot multiplication (98–100%) as well as the number of shoots per node (six to eight) were highest during the first three culture passages, after which there was a gradual decline in shoot development. Rooting was best induced (94%) in shoots excised from proliferated shoot cultures on half-strength MS medium augmented with an optimal combination of indole-3-acetic acid and indole-3-butyric acid each at 1.0 mg/l. Vermi-compost was the most suitable planting substrate for hardening inside a plant growth chamber and its use ensured high-frequency survival (93%) of regenerated plants prior to outdoor transfer. Micropropagated plants established in garden soil were uniform and identical to the donor plant with respect to growth characteristics as well as vegetative and floral morphology.

Abstract: Raffinose family oligosaccharides (RFOs) have been implicated in mitigating the effects of environmental stresses on plants. In seeds, proposed roles for RFOs include protecting cellular integrity during desiccation and/or imbibition, extending longevity in the dehydrated state, and providing substrates for energy generation during germination. A gene encoding galactinol synthase (GOLS), the first committed enzyme in the biosynthesis of RFOs, was cloned from tomato (Lycopersicon esculentum Mill. cv Moneymaker) seeds, and its expression was characterized in tomato seeds and seedlings. GOLS (LeGOLS-1) mRNA accumulated in developing tomato seeds concomitant with maximum dry weight deposition and the acquisition of desiccation tolerance. LeGOLS-1 mRNA was present in mature, desiccated seeds but declined within 8 h of imbibition in wild-type seeds. However, LeGOLS-1 mRNA accumulated again in imbibed seeds prevented from completing germination by dormancy or water deficit. Gibberellin-deficient (gib-1) seeds maintained LeGOLS-1 mRNA amounts after imbibition unless supplied with gibberellin, whereas abscisic acid (ABA) did not prevent the loss of LeGOLS-1 mRNA from wild-type seeds. The presence of LeGOLS-1 mRNA in ABA-deficient (sitiens) tomato seeds indicated that wild-type amounts of ABA are not necessary for its accumulation during seed development. In all cases, LeGOLS-1 mRNA was most prevalent in the radicle tip. LeGOLS-1 mRNA accumulation was induced by dehydration but not by cold in germinating seeds, whereas both stresses induced LeGOLS-1 mRNA accumulation in seedling leaves. The physiological implications of LeGOLS-1 expression patterns in seeds and leaves are discussed in light of the hypothesized role of RFOs in plant stress tolerance.

Abstract: The sites of gibberellin (GA) synthesis in light grown sunflower plants were studied. The results of organ excision and the exogenous application of indole acetic acid and gibberellic acid indicated that gibberellin synthesis occurred in the young leaves of the apical bud. This was substantiated using a combination of diffusion and extraction techniques. Diffusion of sunflower apical buds on agar for 20 hours revealed a level of gibberellin greater than that obtained by solvent extraction of a similar number of apices, indicating that synthesis of gibberellin was occurring in those apices during the diffusion period. The gibberellin level of apices extracted following a 20 hour diffusion period was the same as that obtained from buds extracted immediately following excision from the plant, again suggesting that apical buds are sites of gibberellin synthesis. A similar experiment was conducted with young internodal sections, the results indicating that they were not sites of gibberellin synthesis.The ability of various parts of sunflower plants to produce gibberellin as measured by the agar diffusion technique was correlated to internode elongation, the region of maximum gibberellin production being associated with the region of maximum extension growth.Root tips were also shown to be sites of gibberellin synthesis, this ability being confined to the apical 3 to 4 mm region of the root.

Abstract: Cotyledons of cucumber plants grown in the dark for 7 days were treated with various concentrations of cytokinins for 14 h and then moved into light. After 3 h the treated cotyledons had up to 450% more chlorophyll than the water controls. This suggests that cytokinins have an important role in the formation of chlorophyll. The increase in chlorophyll level was proportional to cytokinin concentration and was apparent at concentrations as low as 0.001 mg/l. Sensitivity to cytokinins depended on the age of the cotyledons and the time of exposure to light. Gibberellic acid, indoleacetic acid, adenine and sucrose did not cause a similar increase in chlorophyll levels. This effect of cytokinins on chlorophyll formation is valuable as a simple, rapid bioassay for cytokinins.
