Abstract: The monoterpenoid indole alkaloids are a large family of structurally diverse compounds encompassing some of the most important plant medicinais discovered by man. Although these alkaloids were one of the last major groups of well-known natural products to have their biogenesis delineated, they have become one of the first classes of plant secondary metabolites to be studied at the enzymatic level. A contributory factor has been the excellent progress made by in vivo tracer work, which within a little more than a decade has led to the elucidation of the major features of a highly complicated pathway1,2.

Abstract: Non-phenolic oxidative coupling: The first total synthesis of the C-2 symmetric indole alkaloid P-(+)-dispegatrine (1) is reported. A late-stage thallium(III)acetate mediated intermolecular oxidative coupling was employed to construct the C(9)-C(9′) bond with complete regio- and stereocontrol. The exclusive formation of a single atropodiastereomer 12 in this critical step arises due to internal asymmetric induction, as planned. In addition, the first total synthesis of four other monomeric sarpagine indole alkaloids is described.
