Abstract: The invention specifically relates to a preparation method and novel application of a C-glycosylflavone compound, which belongs to the fields of medicine and chemistry of natural products. According to the invention, normal-phase (silica gel) column chromatography is employed for separation at first and inversed-phase (open ODS) column chromatography for separation next, and an isovitexin crystal is naturally precipitated. A dry-process sample introduction way is employed during separation via silica gel column chromatography, while a wet-process sample introduction way is employed during separation via open ODS column chromatography. The invention also provides application of isovitexin to myocardial preservation.

Abstract: Background: Clinacanthus nutans, also known as ‘Sabah snake grass’ or ‘Belalai gajah’, is a herb well known locally for its medicinal values. The primary chemical constituents of the leaves are schaftoside, vitexin, isovitexin, orientin and isoorientin, and antiviral activity is shown by two glycoglycerolipids. Despite the importance of C. nutans, complete information with respect to commercial production and postharvest handling of the herb in the local herbal industry is still lacking. Thus, the objective of this study was to determine the optimum postharvest handling processes that could retain the phytochemicals quality of C. nutans. Materials and Methods: The flavonoid compounds of C. nutans were analysed by using ultra fast liquid chromatography (UFLC). Total phenolic content and antioxidant activity were determined using a spectrophotometer. Results: The total phenolic compounds and antioxidant activity in C. nutans were found to be higher in the young vegetative stage than in the mature vegetative stage. Flavonoid compounds (schaftoside, isovitexin, vitexin and orientin) were also found to be highest in the young vegetative plant compared to the mature vegetative plant. All of the assayed phytochemicals and flavonoid compounds levels were found to be highest in oven dried samples compared to the sun, air and solar dried samples. Conslusion: This study suggests that oven-drying young vegetative C. nutans plant material is the optimum method to retain postharvest quality.

Abstract: Quantitative and qualitative analysis of flavonoids in showy tick trefoil (Desmodium canadense L., Fabaceae family) herbs was performed using HPLC techniques during perennial vegetation at the Department of Medicinal Herbs of the Kaunas Botanical Garden (Vytautas Magnus University). A set of 15 flavonoids of the aglicone and glycoside nature were determined, including apigenin, apigenin-7-O-glycoside, luteolin, rutin, 2-vicenin, vitexin, isovitexin, vitexin rhamnoside, orientin, homoorientin, quercitrin, quercetin, hyperoside, astragalin, and kaempferol. The maximum amount of identified flavonoids was observed at the flowering phase: 2.45 % on the second vegetative year and 1.28 % on the sixth vegetative year, i.e., their amount constantly decreased during the plant perennial vegetation process. Among these, orientin and homoorientin varied from 45.9 to 38.9 % of the total amount of flavonoids; 2-vicenin varied from 16.3 to 12.8 %, vitexin, from 12.7 to 8.6%; isovitexin, from 13.0 to 7.7 %; and rutin, from 1.6 to 6.9%. The optimum time of collecting showy tick trefoil herbs as raw plant material is the flowering and preflowering phases of the herbs regrowing upon mowing. The recommended optimal exploitation of plantation is up to six years.
