Abstract: Synthetic antioxidants such as butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT) are found to be toxic, hence non-carcinogenic naturally occurring radical scavengers especially flavonoids have gained considerable importance in the past two decades. In the present investigation, the radical scavenging activity of C-glycosyl flavonoids is evaluated using theoretical approach which could broaden its scope in therapeutic applications. Gas and solvent phase studies of structural and molecular characteristics of C-glycosyl flavonoid, isovitexin is investigated through hydrogen atom transfer mechanism (HAT), Electron transfer-proton transfer (ET–PT) and Sequential proton loss electron transfer (SPLET) by Density functional theory (DFT) using hybrid parameters. The computed values of the adiabatic ionization potential, electron affinity, hardness, softness, electronegativity and electrophilic index indicate that isovitexin possess good radical scavenging activity. The behavior of different –OH groups in polyphenolic compounds is assessed by considering electronic effects of the neighbouring groups and the overall geometry of molecule which in turn helps in analyzing the antioxidant capacity of the polyphenolic molecule. The studies indicate that the H–atom abstraction from 4’–OH site is preferred during the radical scavenging process. From Mulliken spin density analysis and FMOs, B–ring is found to be more delocalized center and capable of electron donation. Comparison of antioxidant activity of vitexin and isovitexin leads to the conclusion that isovitexin acts as a better radical scavenger. This is an evidence for the importance of position of glucose unit in the flavonoid.

Abstract: This study aimed to investigate the proximate and phytochemicals present in seeds of 24 mung bean (Vigna radiate L.) genotypes from four provinces of China for estimating their nutritional and antioxidant properties. Proximate analysis of mung bean genotypes revealed that starch, protein, fat, ash and water-soluble polysaccharide ranged from 39.54–60.66, 17.36–24.89, 4.24–12.18, 2.78–3.53 and 1.99–2.96 g/100 g respectively. The five principal fatty acids detected in mung beans were stearic acid, palmitic acid, linoleic acid, oleic acid, and linolenic acid. The contents of insoluble-bound phenolic compounds, soluble phenolic compounds, and flavonoids ranged from 0.78 to 1.5 mg GAE g− 1, 1.78 to 4.10 mg GAE g− 1, and 1.25 to 3.52 mg RE g− 1, respectively. The black seed coat mung bean genotype M13 (Suheilv 1) exhibited highest flavonoid and phenolic contents which showed strong antioxidant activity. Two flavonoids (vitexin and isovitexin) and four phenolic acids (caffeic, syringic acid, p-coumaric, and ferulic acids) were identified by HPLC. Vitexin and isovitexin were the major phenolic compounds in all mung bean genotypes. The content of soluble phenolic compounds had positive correlation with DPPH (r2 = 0.713) and ABTS (r2 = 0.665) radical scavenging activities. Principal component analysis indicated that the first two principal components could reflect most details on mung bean with a cumulative contribution rate of 66.1%. Twenty-four mung bean genotypes were classified into four groups based on their phenolic compounds contents and antioxidant activities. The present study highlights the importance of these mung bean genotypes as a source of nature antioxidant ingredient for the development of functional foods or a source of health promoting food.

Abstract: Objective To isolate and identify the flavones in the leaves of Isatis indigotica Fort.(Folium Isatidis).Methods The ethanol extract of Folium Isatidis crude drug was extracted with chloroform,and the aqueous layer was separated by macroporous resin,silica gel and polyamide column chromatography,respectively.The compounds obtained were identified by mass spectra and NMR data.Results The compounds separated were identified as isoscoparin(1),isoscoparin-3″-O-glucopyranoside(2),isovitexin(3),isovitexin-3″-O-glucopyranoside(4).Conclusions Among them,compound 1,2 and 4 are isolated from Isatis indigotica Fort.for the first time.

Abstract: Objectives: This study aimed to isolate phenolic compounds from the 70% aqueous methanol extract of the aerial parts (leaves, flowers and stems) of Centaurea aegyptiaca , evaluate in-vivo hepatoprotective activity and determine total phenolic and total flavonoid contents. Methods: 70% aqueous methanol extract of the aerial parts subjected to different chromatographic separation techniques. The identities of the isolated compounds were established on the basis of their spectral data and comparing with previously reported data. The crude extract was evaluated for in-vivo hepatoprotective activity on CCl4-induced hepatotoxicity in rats. Total phenolic and total flavonoid contents of the aqueous methanol extract were determined using Folin-Ciocalteu and aluminum chloride colorimetric methods, respectively. Results: The aqueous methanol extract of the aerial parts of Centaurea aegyptiacaafforded seven compounds. A phenolic acid ester; protochatechuic acid methyl ester (1) along with six known flavonoids; apigenin-6-C-β-D-glucopyranoside (isovitexin) (2), apigenin-8-C-β-D-glucopyranoside (vitexin) (3), quercetin-3-O-b-D-glucopyranoside (isoquercetin) (4), apigenin (5), 3-O-methylquercetin (6) and quercetin (7). Administration of the extract at doses of 100 and 200 mg/kg b.wt showed a hepatoprotective activity similar to that of the standard drug; silymarin at a dose of 25 mg/kg b.wt. Comparative histopathological study of liver exhibited moderate changes in liver histoarchitecture when compared to the CCl4 group. The methanolic extract showed high concentration of phenolic and flavonoid contents. Conclusion: The aqueous methanol extract of the aerial parts of C. aegyptiaca afforded seven phenolic compounds for the first time from this species, with promising in-vivo hepatoprotective activity.
