Abstract: High-performance liquid chromatography with ultraviolet spectrometer (HPLC-UV) was used to simultaneously detect the four flavone C-glycosides, i. e. orientin, isoorientin, vitexin and isovitexin. Analytes were separated with Waters XTerra MS C18 column (250 mm x 4.6 mm, 5 μm) using acetonitrile and 0.5% (φ) formic acid as mobile phase. The flow rate was set at 1.0 mL · min(-1) with the column temperature at 30 °C, and the detection wavelength was 360 nm. The calibration curve was linear over the concentration range of 0.1-10.0 mg · L(-1) for the mixed standard solution. Analytes were separated in 22 minutes, and the relative standard deviation values were all above 0.999. LOD values of standards were found to be between 0.03 and 0.07 mg · L(-1), and LOQ values were in the range of 0.04-0.08 mg · L(-1). After comparing the spectra (240-400 nm) of four flavone C-glycosides in mixed standards and the final product purified by macroporous resin, respectively, the curve shape and characteristic ultraviolet absorption wavelength of each flavone C-glycoside including orientin, isoorientin, vitexin and isovitexin were fitted well. The bamboo leaves sample was extracted by ethanol under reflux, and then partitioned with water and petroleum ether. The aqueous phase was added onto macroporous resin (AB-8), and the fraction of ethanol-water (40%, φ) was concentrated. It was found that the contents of orientin, isoorientin, vitexin and isovitexin relative to the fraction of ethanol- water were 13.73, 49.68, 7.85 and 30.70 mg · g(-1), respectively. In addition, the average recovery of the four flavone C-glycosides ranged from 34.90% to 87.64% with RSD values from 0.41% to 10.83%. The results showed that bamboo leaves sample had good stability and repeatability. The new method was used to analyze the four flavone C-glycosides quickly and provide quality control for commercial products.

Abstract: Ficus deltoidea (Moraceae), Mas cotek, is a local medicinal plant which has been used for its health benefits including antioxidant, improving blood circulation and anti-hypertension effects. This study aims to determine the vitexin and isovitexin content in various varieties of F. deltoidea, and to study the extract’s inhibitory effect on angiotensin converting enzyme (ACE) activity as anti-hypertension marker. Five varieties were studied including F. deltoidea var deltoidea, F. deltoidea var. angustifolia, F. deltoidea var tranguensis, F. deltoidea var. Telinga Beruk, and F. deltoidea var. Tapak Itik. Five extracts were prepared; water, methanolic, ethanolic, 50% methanolic and 50% ethanolic, and ACE inhibitory effect was studied in vitro. Vitexin and isovitexin concentration was in the range 0.001–0.35% and 0.001–7.025% (w/w), respectively. The highest ACE inhibition was obtained from F. deltoidea var. Tapak Itik (FD-TI) extracts where the 50% ethanolic and 50% methanolic extracts showed the most promising results. A bivariate correlation analysis may indicate existence of relation between vitexin in FD extracts and the ACE inhibition (r = 0.58), whereas the isovitexin results do not show any relationship (r= -0.06). The median inhibitory concentration (IC50) of the 50% ethanolic extract of FD-TI was 22±3.8μg/ml, and that of vitexin and isovitexin was 3.1±0.5μg/ml, 18.6±1.3μg/ml, respectively. Our results showed the ACE inhibitory effect is partly due to existence of vitexin but not isovitexin, and the 50% ethanolic extract of the F. deltoidea var. Tapak Itik as a possible antihypertension candidate.

Abstract: Isovitexin, a bioactive flavonoid constituent isolated from Desmodii Styracifolii, is considered an adjuvant for antiurolithiasis diseases. In this study, an ultra-high-performance liquid chromatography coupled with hybrid triple quadruple time-of-flight mass spectrometry method was developed to characterize and compare the metabolic profiling of isovitexin experimented on normal and kidney stone model rats. The comparative research indicated that 28 metabolites (18 phase I and 10 phase II) in normal rats and 33 metabolites (20 phase I and 13 phase II) in kidney stone model rats were initially identified. The results of relative quantitative determination reflected that the contents of metabolites produced by deglycosylation, reduction, and isomerization in kidney stone model rats were greater than those in healthy rats. Instead, the levels of oxidative and dehydrogenated metabolites in normal groups were higher than those in kidney stone model groups. The results of this study are valuable and important for understanding the metabolic process of isovitexin in clinical application, and especially the metabolism study in kidney stone model rats could provide a beneficial reference for the further search of effective substances associated with the treatment of kidney stones.

Abstract: The composition of flavonoids of 12 species of plants of the genus Silene introduced into Western Siberia was studied for the first time. The C-diglycoside of apigenin, shaftoside, was isolated from the aboveground part of S. chalcedonica, the structure of which was identified by HPLC, mass spectrometry, and NMR spectroscopy. In all other studied species, the presence of shaftoside and other C-glycosides of flavones was established on the basis of HPLC data. C-diglycoside of apigenin - vicenin-2 was first identified in S. sendtneri, S. roemeri, S. viridiflora, S. paradoxa, S. nemoralis, S. frivaldszkyana, S. colpophylla, S. linicola, S. caramanica. The most common in the studied Silene species are the apigenin monoglycosides vitexin and isovitexin and a number of their derivatives, luteolin derivatives orientin and others are found a little less frequently. The highest content of flavonoids in the aboveground part of the studied species is characterized by S. nemoralis (6.75%), high – S. chalcedonica, S. paradoxa, S. frivaldszkyana, S. caramanica, S. sendtneri (3.35–5.15%). The major component in the sum of flavonoids is shaftoside, the proportion of which in most species ranges from 50 to 90%. It is shown that the composition of flavonoids changes during vegetative development. The phases of maximum accumulation of flavonoids in the aboveground part of 4 Silene species have been determined. In general, the studied Silene species are promising sources of C-glycosylflavones.

Their structures were elucidated by spectroscopic data as saikogenin F (1), 3-O-beta-D-fucopyranosylsaikogenin F (2), clinoposaponin XV (3), saikosaponin A (4), 6"-acetylsaikosaponin A (5), clinoposaponin I (6), bupleuroside I (7) , clinoposaponin XII (8) , saikoponin b3 (9), isovitexin (10) , swertisin (11) , isoorientin (12), 3',4',5-trihydroxy-7-methoxy-6-C-beta-D-glucopyranosyl-flavone (13). Compounds 1-10, 12-13 were all isolated from Comastoma genus for the first time.
